High-resolution crystal structures of large ribosomal subunits from Deinococcus radiodurans complexed with tRNA-mimics indicate that precise substrate positioning, mandatory for efficient protein biosynthesis with no further conformational rearrangements, is governed by remote interactions of the tRNA helical features. Based on the peptidyl transferase center (PTC) architecture, on the placement of tRNA mimics, and on the existence of a two-fold related region consisting of about 180 nucleotides of the 23S RNA, we proposed a unified mechanism integrating peptide bond formation, A-to-P site translocation, and the entrance of the nascent protein into its exit tunnel. This mechanism implies sovereign, albeit correlated, motions of the tRNA termini and includes a spiral rotation of the A-site tRNA-3' end around a local two-fold rotation axis, identified within the PTC. PTC features, ensuring the precise orientation required for the A-site nucleophilic attack on the P-site carbonyl-carbon, guide these motions. Solvent mediated hydrogen transfer appears to facilitate peptide bond formation in conjunction with the spiral rotation. The detection of similar two-fold symmetry-related regions in all known structures of the large ribosomal subunit, indicate the universality of this mechanism, and emphasizes the significance of the ribosomal template for the precise alignment of the substrates as well as for accurate and efficient translocation. The symmetry-related region may also be involved in regulatory tasks, such as signal transmission between the ribosomal features facilitating the entrance and the release of the tRNA molecules. The protein exit tunnel is an additional feature that has a role in cellular regulation. We showed by crystallographic methods that this tunnel is capable of undergoing conformational oscillations and correlated the tunnel mobility with sequence discrimination, gating and intracellular regulation.  相似文献

Collagenous transmembrane proteins are an emerging group of biologically versatile molecules which function as both cell surface receptors and matrix molecules. The seven group members have interesting structural similarities: they are integral membrane proteins in type II orientation and have one or more collagenous domains in the extracellular C-terminus; interspersed by non-collagenous stretches which confer structural flexibility to the ectodomain. A conserved coiled-coil sequence (linker domain) immediately adjacent to the extracellular face of the cell membrane presumably serves as a nucleus for trimerization and triple-helix folding of each collagen. Intriguingly, the ectodomains of at least some of these molecules are proteolytically shed from the cell surface, releasing a shorter form of the collagen into the extracellular matrix. Collagenous transmembrane proteins are expressed in many different tissues and cells, and are involved in a broad spectrum of biological functions, reaching from epithelial and neural cell adhesion, and epithelial-mesenchymal interactions during morphogenesis to host defense against microbial agents. Several group members are involved in the molecular pathology of genetic and acquired human diseases including epidermolysis bullosa, ectodermal dysplasia, bullous pemphigoid or Alzheimer disease. An extensively investigated member is collagen XVII, a keratinocyte surface protein, which attaches the epidermis to the basement membrane in the skin. In this review, the structure and functions of the currently known collagenous transmembrane proteins are summarized and, as a 'prototype' of the group, collagen XVII and its biology and pathophysiology are delineated.  相似文献

Zusammenfassung  Während das Mortalitätsrisiko für Vögel durch Drahtanflug und Stromschlag an Hochspannungsleitungen vielfach untersucht wurde, könnte auch ein Einfluß von Hochspannungstrassen (110 kV) auf Vögel bestehen, die sich am Boden aufhalten. Dieser wurde 1994/95 am Beispiel der Flächennutzung überwinternder Bläß- und Saatgänse (Anser albifrons, A. fabalis) am Unteren Niederrhein untersucht. Da Gänse offene ungestörte Äsungsflächen beanspruchen, wurde der Arbeitshypothese nachgegangen, daß trassennahe Bereiche von Gänsen gemieden bzw. vermindert genutzt werden. Die Besuchsfrequenz von vier häufig genutzten trassenüberspannten Äsungsflächen wurde ermittelt. Im Anschluß an die einzelnen mehrtägigen Beäsungsphasen wurde die Losungsdichte (LD) als Maß der Flächennutzung senkrecht zum Trassenverlauf bestimmt. An 16–20 m hohen Trassen stieg die LD mit zunehmender Entfernung zur Trasse. Eine signifikante Abnahme der LD trat unterhalb von 40–80 m Abstand zur Trasse ein. Kleine Teilflächen, die durch die Trassen von größeren Bereichen abgetrennt sind, wurden weniger genutzt. An einer 60 m hohen, den Rhein Überquerenden Trasse, konnte kein trassenbedingter Unterschied der LD festgestellt werden. Mögliche Ursachen für die beschriebenen Effekte werden diskutiert.
The impact of power lines on field selection and grazing intensity of wintering White-fronted- and Bean GeeseAnser albifrons, A. fabalis
In addition to studies about the mortality risk by collision and electrocution power lines may have an impact on birds during foraging behaviour on the ground. We examined this question in geese (Anser albfrons, A. fabalis) wintering in the lower Rhine area of North Rhine Westfalia (FRG) 1994/95. Because geese prefer large areas with low levels of disturbance by human activities or urbanic structures like buildings or roads, we hypothesized that they might be disturbed by the presence of power lines in their foraging sites and thus avoid areas next to the lines. To quantify the grazing intensity in relation to the distance from the power lines (110 kV), dropping densities have been repeatedly counted after independant grazing intervals in four different feeding sites. Our data show that: 1. Grazing intensity increases with the distance to power lines of low height. Significantly reduced grazing levels have been found in distances less than 80 to 40 m to the power lines. 2. No effects have been observed for a power line in an area next to the river rhine where the wires crossed the ground in a height of about 60 metres. 3. In small pasture or field areas, cut off by power lines from large ones, a generally reduced grazing amount is evident. Several conceivable reasons for the described effects are discussed.

Plasma progesterone levels in heparinized blood collected at 10 min intervals for 8 continuous hours from four nulliparous Holstein cows on day 3 (early luteal), day 10 or 11 (mid-luteal) and day 18 or 19 of the estrous cycle were found to decline over time when blood was incubated at ambient temperature. The loss was more obvious during the mid-luteal collection period than either the day 3 or day 18 or 19 periods in all cows. This appeared to be associated more with high progesterone levels on day 10 or 11 rather than with differences in the period of the estrous cycle. There was an average decrease in progesterone levels of 3.4, 1.0 and 1.5 ng/ml between samples having the shortest and longest incubation periods on day 10 or 11, day 3 and day 18 or 19, respectively. This apparent decrease in levels of progesterone from bovine blood indicates need in the future for careful consideration concerning the handling of bovine blood collected for subsequent radioimmunoassay (RIA) of progesterone. Further work to elucidate the mechanism which is responsible for the apparent loss is also warranted.  相似文献

Summary A simple effective technique for the fractionation of protoplast populations is described. Protoplasts are separated by low-speed centrifugation in an iso-osmotic, discontinuous density gradient system on the basis of differences in their buoyant densities. At a constant osmolality of 660±20 mOs/kg H2O, the gradients provide a density range from 1.017 to 1.069 g/cm3 at 20 °C which corresponds to the buoyant densities of most protoplast types studied. Characteristics of the KMC/S-density gradient system and factors affecting the fractionation were investigated. Protoplasts were isolated from various tissues and cultivars of tobacco, barley, wheat, rye, oat and maize. Their density-dependent distribution profiles in KMC/S-gradients and their average buoyant densities were determined under standardized conditions. Great differences in the buoyant densities were found between protoplasts of different tissues. Mixed populations of two types of protoplasts, differing in buoyant density by about 15–20 mg/cm3, were separated to give highly purified fractions. Factors affecting the buoyant densities of protoplasts have been investigated. Ploidy level and species differences did not significantly affect the fractionation profiles. However, an age-dependent variation in the average buoyant density of tobacco mesophyll protoplasts was observed. Fractionation of tobacco mesophyll protoplasts and their subsequent regeneration to plants demonstrates the practicability and physiological compatibility of the KMC/S-density gradient system under sterile conditions. The morphogenetic potential of protoplasts was not affected by the separation procedure or the gradient components.  相似文献
