Summary In earlier ultrastructural studies of the supraoptic nucleus in adult rats we noted free and incompletely covered postsynaptic densities (collectively referred to here as vacant postsynaptic densities) on dendritic shafts. Free postsynaptic densities have been reported in other parts of the central nervous system of normal rodents. We investigated the possibility that physiological activation of the supraoptic cells, which produces changes in many aspects of their morphology, would alter the incidence of the free or incompletely covered postsynaptic densities on dendrites in the supraoptic basal dendritic zone. The cells of the supraoptic nucleus are activated to increase cell firing and secretion of oxytocin and/or vasopressin in response to dehydration, gestation, and lactation. We have examined: (i) untreated virgin females; (ii) untreated males; (iii) 24 h water-deprived males; (iv) prepartum (21st day of gestation) females; (v) postpartum females (on the day of parturition); (vi) lactating females (14 days of suckling); (vii) mothers 10 days after weaning their pups; (viii) females given 2% saline to drink (dehydrated) for 10 days; and females or males given 2% saline to drink for 10 days, then given tap water for (ix) 2 or (x) 5 weeks to allow rehydration. Only long-term activation of the supraoptic nucleus by lactation or by drinking saline for 10 days brought about significant decreases in the percentage of dendrites with vacant postsynaptic densities. These densities did not reappear in saline treated rats which had been rehydrated for 2 weeks, but did return in both the 5-week rehydration and the 10-day postweaning groups. Short-term activation of the supraoptic nucleus, such as occurs at parturition or in acute dehydration, did not affect the vacant postsynaptic densities. Analysis of semiserial thin sections indicated that presynaptic elements facing the incompletely covered postsynaptic densities contain predominantly clear round vesicles and also that apparently free postsynaptic densities were usually at least partially contacted by a presynaptic ending in adjacent sections.  相似文献

Samples of human apotransferrin (apo . HTr) were saturated with Fe(III) by two different techniques, a method employing excess trisodium citrate to chelate Fe(III) and a nonchelating approach which involves the ferroxidase activity of ceruloplasmin to convert Fe(II)----Fe(III). The samples were radiolabelled with either 55Fe or 3H. Using an initial molar Fe/apo . HTr ratio of 2.0-2.1, preparations of human transferrin with bound Fe (Fe . HTr) using the citrate method invariably contained 2.2-2.4 atoms Fe/molecule, whereas Fe . HTr (ceruloplasmin method) contained 2.0 atoms/molecule as shown by spectrophotometric and radioactivity measurements. Uptake of Fe from these Fe . HTr preparations by K-562 cells grown in a serum-free medium was marginally, but consistently, more rapid from 55Fe . HTr (citrate) than from 55Fe . HTr (ceruloplasmin). Taking account of the different Fe contents of the Fe . HTr preparations, the rate measured over a 2-h period amounted to approximately 12,700 and 16,100 Fe atoms/(cell . min) for Fe . HTr (ceruloplasmin) and Fe . HTr (citrate), respectively. However, cell binding by the two Fe . [3H]HTr preparations did not differ significantly over the 8-h incubation period. Furthermore, from the 3H distribution, the quantities of Fe . HTr bound reversibly at the cell surface and contained within the cell were similar for the two Fe . HTr preparations. The results indicate that apo . HTr may bind Fe in different ways depending on the method of Fe presentation and that the Fe . HTr product can donate Fe to K-562 cells at a rate which may reflect the method used for Fe-complex formation.  相似文献

This field study examined the ecophysiological responses of three tree species to salinity in the Austin Bay Nature Reserve, adjacent to the Peel-Harvey Estuary in Western Australia (115°46' E 32°37' S). The area is at increased risk of flooding with saline water during storm surges due to the construction of a channel between the estuary and Indian Ocean in 1994. Banksia attenuata R.Br. occurs on small sandy ridges adjacent to a seasonal wetland, while Melaleuca cuticularis Labill. and Casuarina obesa Miq. occur in a seasonally flooded wetland. Landscape position determined exposure to salinity, with M. cuticularis and C. obesa experiencing high soil and groundwater salinity during summer (electrical conductivity, EC, up to 70 dS m−1) while B. attenuata was not exposed to soil or groundwater with EC greater than 20 dS m−1. B. attenuata had relatively stable leaf water status throughout the year and did not osmotically adjust as root-zone salinity increased. By contrast, M. cuticularis and C. obesa had large variation in stem water potential and exhibited osmotic adjustment during summer. Whereas the sap flow rates of M. cuticularis and C. obesa remained high throughout the year, sap flow of B. attenuata decreased during summer which may have limited uptake of salt. The three species also exhibited differences in traits associated with tissue-level salt tolerance, as M. cuticularis and C. obesa produced compatible organic solutes (methyl proline in M. cuticularis and proline in C. obesa ), whereas B. attenuata did not. The distributions of these species within the Austin Bay Nature Reserve are determined in part by their tolerance to salinity, which will influence their responses to hydrological disturbance.  相似文献

To date, most studies of white matter changes in Bipolar Disorder (BD) have been conducted in older subjects and with well-established disorders. Studies of young people who are closer to their illness onset may help to identify core neurobiological characteristics and separate these from consequences of repeated illness episodes or prolonged treatment. Diffusion tensor imaging (DTI) was used to examine white matter microstructural changes in 58 young patients with BD (mean age 23 years; range 16–30 years) and 40 controls. Whole brain voxelwise measures of fractional anisotropy (FA), parallel diffusivity (λ//) and radial diffusivity (λ⊥) were calculated for all subjects. White matter microstructure differences (decreased FA corrected p<.05) were found between the patients with BD and controls in the genu, body and splenium of the corpus callosum as well as the superior and anterior corona radiata. In addition, significantly increased radial diffusivity (p<.01) was found in the BD group. Neuroimaging studies of young patients with BD may help to clarify neurodevelopmental aspects of the illness and for identifying biomarkers of disease onset and progression. Our findings provide evidence of microstructural white matter changes early in the course of illness within the corpus callosum and the nature of these changes suggest they are associated with abnormalities in the myelination of axons.  相似文献

Alternatively spliced Ultrabithorax mRNAs differ by the presence of internal exons mI and mII. Two approaches were used to identify trans-acting factors required for inclusion of these cassette exons. First, mutations in a set of genes implicated in the control of other alternative splicing decisions were tested for dominant effects on the Ubx alternative splicing pattern. To identify additional genes involved in regulation of Ubx splicing, a large collection of deficiencies was tested first for dominant enhancement of the haploinsufficient Ubx haltere phenotype and second for effects on the splicing pattern. Inclusion of the cassette exons in Ubx mRNAs was reduced strongly in heterozygotes for hypomorphic alleles of hrp48, which encodes a member of the hnRNP A/B family and is implicated in control of P-element splicing. Significant reductions of mI and mII inclusion were also observed in heterozygotes for loss-of-function alleles of virilizer, fl(2)d, and crooked neck. The products of virilizer and fl(2)d are also required for Sxl autoregulation at the level of splicing; crooked neck encodes a protein with structural similarities to yeast-splicing factors Prp39p and Prp42p. Deletion of at least five other loci caused significant reductions in the inclusion of mI and/or mII. Possible roles of identified factors are discussed in the context of the resplicing strategy for generation of alternative Ubx mRNAs.  相似文献

Changes in coastline configuration and sand movements on Portuguese Island, a small island lying close to Inhaca Island 35 km east of Maputo, Mozambique have resulted in large areas of the Island being eroded away and consequent restricted tidal recharge of the mangrove. Measurements made during 1989 show that flooding of the mangrove occurred on only three occasions. Percentage mortality of the dominant species,Ceriops tagal (Perr.) C. B. Robinson., is high. However, at the time of the study, the co-dominant,Bruguiera gymnorrhiza (L.) Lam., showed no signs of stress. Halophytic conditions are indicated by an invasive spread of salt-tolerant herbs. All species of crustaceans and molluscs have disappeared from the mangrove.Although mortality ofC. tagal is high, the regeneration status of this species is also high, comparable to that in a regularly recharged mangrove on Inhaca Island. It is suggested thatC. tagal propagules were shed under conditions when seedling establishment was favorable i.e. flooding. These seedlings may represent the last cohort which may also suffer stress and death unless tidal recharge is re-established.  相似文献

Construction of a hydropower project on the Kihansi river in Southern Tanzania will substantially alter the river's flow. On the basis of both qualitative and quantitative botanical surveys, forest in the Kihansi gorge below the dam site was found to be mostly typical species and endemic-rich Eastern Arc forest. An unusual forest type, dominated almost entirely by Filicium decipiens was also present. Eastern Arc forests are of both global and national importance for biodiversity conservation, and measures to mitigate impact of the dam on the forest are suggested. Further environmental monitoring is recommended in order to evaluate the choice of mitigation measures.  相似文献
