Cationic trialkylammonium‐substituted α‐, β‐, and γ‐cyclodextrins containing trimethyl‐, triethyl‐, and tri‐n‐propylammonium substituent groups were synthesized and analyzed for utility as water‐soluble chiral nuclear magnetic resonance (NMR) solvating agents. Racemic and enantiomerically pure (3‐chloro‐2‐hydroxypropyl)trimethyl‐, triethyl‐, and tri‐n‐propyl ammonium chloride were synthesized from the corresponding trialkyl amine hydrochloride and either racemic or enantiomerically pure epichlorohydrin. The ammonium salts were then reacted with α‐, β‐, and γ‐cyclodextrins at basic pH to provide the corresponding randomly substituted cationic cyclodextrins. The 1H NMR spectra of a range of anionic, aromatic compounds was recorded with the cationic cyclodextrins. Cyclodextrins with a single stereochemistry at the hydroxy group on the (2‐hydroxypropyl)trialkylammonium chloride substituent were often but not always more effective than the corresponding cyclodextrin in which the C‐2 position was racemic. In several cases, the larger triethyl or tri‐n‐propyl derivatives were more effective than the corresponding trimethyl derivative at causing enantiomeric differentiation. A theoretical model of the composition of the inorganic carbon pool generated in C4 leaves during steady-state photosynthesis was derived. This model gives the concentrations of CO2 and O2 in the bundle sheath cells for any given net photosynthesis rate and inorganic carbon pool size. The model predicts a bundle sheath CO2 concentration of 70 micromolar during steady state photosynthesis in a typical C4 plant, and that about 13% of the inorganic carbon generated in bundle sheath cells would leak back to the mesophyll cells, predominantly as CO2. Under these circumstances the flux of carbon through the C4 acid cycle would have to exceed the net rate of CO2 assimilation by 15.5%. With the calculated O2 concentration of 0.44 millimolar, the potential photorespiratory CO2 loss in bundle sheath cells would be about 3% of CO2 assimilation. Among the factors having a critical influence on the above values are the permeability of bundle sheath chloroplasts to HCO3, the activity of carbonic anhydrase within these chloroplasts, the assumed stromal volume, and the permeability coefficients for CO2 and O2 diffusion across the interface between bundle sheath and mesophyll cells. The model suggests that as the net photosynthesis rate changes in C4 plants, the level and distribution of the components of the inorganic carbon pool change in such a way that C4 acid overcycling is maintained in an approximately constant ratio with respect to the net photosynthesis rate.  相似文献

A method is described which is suitable for assessing the effects of relative humidity (RH) on the viability of two unicellular algae in experimental aerosols. Viable cells of Nannochloris atomus collected from the airborne state were detected by plating onto agar surfaces of an appropriate growth medium, whereas viable airborne cells of Synechococcus sp., because of unreliable growth on solid media, were determined by a liquid assay system. The assays were performed at intervals during short-term and prolonged storage of algal aerosols in chambers preconditioned to a selected RH and temperature. Both species showed the greatest loss in viability during the first minute after atomization, and the extent of this inactivation, as a function of RH, reflected the subsequent long-term survival. The airborne eukaryotic alga was unable to survive at an RH below 91%, whereas the airborne prokaryotic alga was comparatively stable over a wide humidity range. Initial inactivation was least and long-term survival best, for both species, at 94% RH.  相似文献

Human antisera against Australia (Au) antigen have been characterized by liquid-phase radioimmunoassay (RIA) for their precipitation of (125)I-labeled Au antigen. The end-point dilutions of sera (anti-Au) which precipitated 50% of (125)I-Au antigen by RIA correlated well with complement fixation titers but had a much wider range, indicating a greater precision and perhaps a better sensitivity of assay. Anti-Au serum diluted to precipitate 50% of (125)I-labeled Au antigen was used as standard antibody in RIA tests to detect either inhibition or enhancement of the reaction by preincubated mixtures of Au antigen and antibody specimens. Without free Au antigen or antibody in the resultant mixtures there was no inhibition or enhancement; the mixtures presumably contained immunoreactively equivalent proportions of Au antigen and antibody. RIA data for diagnostic specimens indicated an end-point sensitivity which was proportional to the dilution of the standard anti-Au sera used in the test. High concentrations of the standard antibody permitted detectable inhibition of (125)I-Au antigen precipitation at lower antigen specimen concentrations. Similarly, low concentrations of the standard antibody permitted detectable enhancement of (125)I-Au antigen precipitation at lower antibody specimen concentrations. Omitting the standard antibody altogether resulted in a more sensitive RIA for Au antibody in test sera.  相似文献

Properties of leaf NAD malic enzyme from plants with C4 pathway photosynthesis   总被引:11,自引:0,他引:11  
C4 acid decarboxylation in one group of C4-pathway species is mediated by an NAD malic enzyme. This paper reports on the partial purification and properties of this enzyme from three species of this group, Atriplex spongiosa, Amaranthus edulis, and Panicum miliaceum. Depending upon the conditions, the Atriplex spongiosa enzyme was 5–30% as active with NADP compared with NAD but the enzyme from the other species was specific for NAD. The enzyme from each species had an absolute requirement for Mn2+ that could not be replaced by Mg2+, and activity was increased several fold by low concentrations of either CoA or acetyl CoA. For the enzyme from Atriplex spongiosa and Amaranthus edulis, there was cooperativity for malate binding and the activators CoA and acetyl CoA functioned to increase the affinity of malate for the enzyme. The Hill coefficients for malate binding were approximately 2 and 4, respectively. However, with the enzyme from Panicum miliaceum, cooperative binding of malate was not apparent and activators operated by increasing V rather than the affinity for malate. Bicarbonate inhibited the enzyme from Atriplex spongiosa and Amaranthus edulis and its effect was inversely related to the concentrations of malate, NAD, and activators. The possible significance of these various allosteric effects on the regulation of the enzyme in vivo is discussed. Reactant concentrations and other conditions required for maximum activity are reported.  相似文献

The effect of an abrupt change in the relative humidity on the viability of airborne Mycoplasma pneumoniae has been examined. When the microbial aerosols were permitted to equilibrate in air held at either low or high humidities and were then subjected to a sudden shift to a mid-range humidity, a significant loss (>90%) of the colony-forming units per liter of aerosol occurred within 8 min. In contrast, a change in the relative humidity of more than 18% in either direction from a lethal mid-range humidity noticeably decreased the rate of biological decay. Double humidity shifts (i.e., from dry to a mid-range level and then to a high humidity range) were very detrimental, with very few survivors after 8 min. These results indicate that the biological stability of airborne M. pneumoniae may be easily modified by a sudden change in the relative humidity, such as occurs in natural atmospheres. This increased sensitivity brought about by producing changes in relative humidity through the lethal humidity range may provide a method whereby the control of these organisms in naturally contaminated indoor air environments may be eventually achieved.  相似文献

Fluorescent-antibody (FA) reagents were prepared from sera of guinea pigs immunized with either native infectious poliovirus type 1 or poliovirus type 1 which had been heated at 56 C for 30 min. Conjugates made from sera of animals immunized with heated virus gave higher direct FA staining titers on air-dried, acetone-fixed, infected cells than conjugates made from sera of animals immunized with native infectious virus. Evidence was obtained that complement-fixing antibody reactive with heated antigen was responsible for the FA staining. Two conjugates prepared from sera of guinea pigs immunized with heated poliovirus type 1 were successfully used to identify 21 type 1 viruses isolated from a group of 44 stool suspensions studied as unknowns. These conjugates did not stain any of 23 heterologous enteroviruses present in the remainder of the stools and gave minimal non-specific staining.  相似文献
