Freshwater fish behaviors have the potential to be impacted by acidification due to increases in dissolved carbon dioxide (CO2). Recent work in the marine environment suggests that increased CO2 levels due to climate change can negatively affect fishes homing to natal environments, while also hindering their ability to detect predators and perform aerobically. The potential for elevated CO2 to have similar negative impacts on freshwater communities remains understudied. The objective of our study was to quantify the effects of elevated CO2 on the behaviors of fathead minnows (Pimephales promelas) and silver carp (Hypophthalmichthys molitrix) following exposure to conspecific skin extracts (alarm cues). In fathead minnows, their response to conspecific skin extracts was significantly impaired following exposure to elevated CO2 levels for at least 96 h, while silver carp behaviors were unaltered. However, fathead minnow behaviors did return to pre-CO2 exposure in high-CO2-exposed fish following 14 days of holding at ambient CO2 levels. Overall, this study suggests there may be potential impacts to freshwater fishes alarm cue behaviors following CO2 exposure, but these responses may be species-specific and will likely be abated should the CO2 stressor be removed.  相似文献

Fine regulation of water reabsorption by the antidiuretic hormone [8-arginine]vasopressin (AVP) occurs in principal cells of the collecting duct and is largely dependent on regulation of the aquaporin-2 (AQP2) water channel. AVP-inducible long term AQP2 expression was investigated in immortalized mouse cortical collecting duct principal cells. Combined RNase protection assay, Western blot, and immunofluorescence analyses revealed that physiological concentrations of AVP added to the basal side, but not to the apical side, of cells grown on filters induced both AQP2 mRNA and apical protein expression. The stimulatory effect of AVP on AQP2 expression followed a V(2) receptor-dependent pathway because [deamino-8-d-arginine]vasopressin (dDAVP), a specific V(2) receptor agonist, produced the same effect as AVP, whereas the V(2) antagonist SR121463B antagonized action of both AVP and dDAVP. Moreover, forskolin and cyclic 8-bromo-AMP fully reproduced the effects of AVP on AQP2 expression. Analysis of protein degradation pathways showed that inhibition of proteasomal activity prevented synthesis of AVP-inducible AQP2 mRNA and protein. Once synthesized, AQP2 protein was quickly degraded, a process that involves both the proteasomal and lysosomal pathways. This is the first study that delineates induction and degradation mechanisms of AQP2 endogenously expressed by a renal collecting duct principal cell line.  相似文献

This paper proposes a general approach for handling multiple contrast tests for normally distributed data in the presence of heteroscedasticity. Three candidate procedures are described and compared by simulations. Only the procedure with both comparison-specific degrees of freedom and a correlation matrix depending on sample variances maintains the alpha-level over all situations. Other approaches may fail notably as the variances differ more. Furthermore, related approximate simultaneous confidence intervals are given. The approach will be applied to a toxicological experiment.  相似文献

Na,K-ATPase plays a crucial role in cellular ion homeostasis and is the pharmacological receptor for digitalis in man. Nine different human Na,K-ATPase isozymes, composed of 3 alpha and beta isoforms, were expressed in Xenopus oocytes and were analyzed for their transport and pharmacological properties. According to ouabain binding and K(+)-activated pump current measurements, all human isozymes are functional but differ in their turnover rates depending on the alpha isoform. On the other hand, variations in external K(+) activation are determined by a cooperative interaction mechanism between alpha and beta isoforms with alpha2-beta2 complexes having the lowest apparent K(+) affinity. alpha Isoforms influence the apparent internal Na(+) affinity in the order alpha1 > alpha2 > alpha3 and the voltage dependence in the order alpha2 > alpha1 > alpha3. All human Na,K-ATPase isozymes have a similar, high affinity for ouabain. However, alpha2-beta isozymes exhibit more rapid ouabain association as well as dissociation rate constants than alpha1-beta and alpha3-beta isozymes. Finally, isoform-specific differences exist in the K(+)/ouabain antagonism which may protect alpha1 but not alpha2 or alpha3 from digitalis inhibition at physiological K(+) levels. In conclusion, our study reveals several new functional characteristics of human Na,K-ATPase isozymes which help to better understand their role in ion homeostasis in different tissues and in digitalis action and toxicity.  相似文献

Recently, an implantable force transducer (IFT) has been introduced [Xu et al. (1992, J. Biomech. Engng 114, 170–177)] which can be used in tight spaces where force recordings with established transducers, such as the buckle-type transducers, are not possible because of impingement artifacts. The IFT is easily implanted in chronic animal preparations; however, calibration of the IFT in terminal experiments has produced unreliable results. The problems of IFT calibration are that minute movements of the transducer within the tendon, slight misalignments of the tendon, or slight errors in the line of pull cause dramatic changes in the IFT voltage output for a given applied calibration load. Here, we propose a method that eliminates the above calibration problems primarily because the target tendon is left in situ, the calibration loads are applied by the muscles which insert into the target tendon, and the transducer is implanted into the target tendon about two weeks prior to calibration. The theoretical and experimental approaches are demonstrated for the cat patellar tendon, but in principle can be performed with any tendon. The results are repeatable, lie within expected values, and reproduce some of the basic properties which have been observed in prior IFT testing.  相似文献

Approximately 1000 registered cows and heifers were superovulated one to 10 times. Nonsurgical embryo recoveries were performed on all donors which exhibited estrus. Healthy donors produced more total ova and cleaving embryos and had a higher ovum recovery rate, fertilization rate and pregnancy rate from embryos transferred than did cows classified as infertile. While ovum number was not affected during 10 repeated superovulations, fertilization rate and embryo number decreased. The number of ova recovered from healthy cows was affected by season, and from infertile cows by the day of the estrous cycle on which FSH was started and by the number of days since calving. More ova were recovered from infertile cows synchronized with prostaglandins prior to superovulation than following a natural estrous cycle. The number of embryos recovered from infertile cows was affected by age and from healthy cows by daily milk production. Fertilization rates in both healthy and infertile cows were affected by age, time since calving, daily milk production, day of cycle FSH was injected and season. There was no effect of the day of recovery on the number of ova or embryos recovered from healthy or infertile cows.  相似文献

Endothelial cell CD36 (glycoprotein IV) has been purified from bovine heart tissue by detergent partitioning and immunoaffinity chromatography. Bovine CD36 differs from human CD36 in its apparent mass (85 versus 88 kDa), primary structure, and immunological cross-reactivity. Of the 18 N-terminal residues identified, 17 conformed to the human CD36 sequence. Mouse monoclonal antibodies E-1 and 8A6 defined bovine- and human-specific epitopes, respectively. Because human CD36 has been identified as a receptor for erythrocytes infected with the malaria parasite Plasmodium falciparum, we examined the ability of bovine CD36 to bind infected erythrocytes. Bovine CD36, unlike human CD36, did not bind infected erythrocytes, suggesting that human CD36-specific structural features are responsible for recognition of the infected erythrocyte ligand.  相似文献

The effect of photoperiod on sexual function and growth in weanling male and female collared lemmings (Dicrostonyx groenlandicus) was studied. Males and females maintained in 6 hr light:18 hr dark after weaning were larger at the end of 70 days than their siblings kept in 20L:4D. Males in 6L:18D were longer and overall body growth was greater. Males in 6L:18D had heavier adrenals but there were no differences in the weights of testes, preputial glands or seminal vesicles or in the mean testosterone levels of the two groups. Females in 6L:18D had heavier preputial glands than the 20L:4D females. There were no differences between the two groups in uterine, ovarian or adrenal weights or in the number or mean size of atretic or healthy Graafian follicles. Adult males kept in 6L:18D increased more in body weight than those in 20L:4D, but there was no detectable influence of the short photoperiod on reproductive function.  相似文献

Morphologically normal embryos were transferred surgically into uteri of normal and repeat-breeder cows at seven days post-estrus to compare embryo survival rates in the two kinds of cows. All cows were less than ten years of age and had no abnormal genital discharges, cystic ovarian follicles, or anatomical abnormalities of the reproductive tract. Normal cows had not been inseminated after last calving. Repeat-breeders had at least four infertile services within the past six months (average of 6.2 services after calving). To test fertility of repeat-breeders at synchronized estrus, 22 anatomically-normal repeat-breeders were treated by intramuscular (i.m.) injection with prostaglandin F(2)alpha (PGF(2)alpha) on day 11 of an estrous cycle (estrus = day 0) and inseminated at induced estrus; 11 cows (50%) had a normal fetus at necropsy on day 60. Twenty-three repeat-breeders and 23 normal cows were assigned as embryo recipients and treated i.m. with PGF(2)alpha to synchronize estrus. All embryo donors were normal cows. Donors were treated with FSH and PGF(2)alpha and inseminated at estrus. On day 7 after estrus, embryos were recovered nonsurgically from donors and one embryo was transferred through a flank incision to the anterior end of the uterine horn adjacent to the corpus luteum of each recipient. Recipients that did not return to estrus were necropsied at day 60. Of 28 normal and 23 repeat-breeder recipients, 23 normal cows (82%) and 16 repeat breeders (70%) were pregnant at day 60 (P=0.235). Thus, at seven days post-estrus, the maternal environment of most of these repeat-breeders was satisfactory for maintaining pregnancy.  相似文献

A family is reported with a variant of von Willebrand's disease. The members of this family showed a qualitative defect of the factor VII/von Willebrand factor protein. The qualitative defect was characterized by an abnormal electrophoretical mobility of factor VIII-related antigen and an abnormal elution pattern as demonstrated by gelfiltration on Sepharose 4 B. Factor VIII-subunits in these patients were found to be normal by polyacrylamidgelelektrophoresis.  相似文献
