An improved, non-surgical technique for recovering bovine eggs is described using modified Foley catheters. Egg recovery rates per attempt were, 3651 (71%), for normal, unsuperovulated donors and 438 (11%), for unsuperovulated donors with known fertility problems. For superovulated donors, eggs were collected in 2426 attempts (92%) averaging 6.9 per recovery.  相似文献

Carbon dioxide has been found to produce various negative consequences for a number of aquatic species and is projected to rise in the future for many aquatic ecosystems. Crayfish act as indicators of water quality and function as keystone species in aquatic food webs; however, there is a paucity of research on how crayfish may respond to elevated CO2. This study quantified how shelter-seeking behavior in freshwater crayfish (e.g., family Cambaridae), a behavior critical for survival and fitness, may change following exposure to elevated pCO2. Red Swamp crayfish (Procambarus clarkii, Girard, 1852) were exposed to one of three potential levels of dissolved CO2 that could be found in freshwater basins currently or under future climate change conditions: Control (2 levels as the respective initial exposure. The treatment aquarium contained a shelter and was divided into three equal sections based on proximity to that shelter. Crayfish proximity to the shelter (defined by the tank sections) in the treatment aquarium was monitored every 5 s for a 2-min trial. Crayfish spent differing amounts of time in differing zones of the experiment and had different levels of activity, depending on their pCO2 exposure; crayfish acclimated to High pCO2 increased their time spent hiding and decreased their overall activity when compared to the Low pCO2 and Control treatments. Augmented shelter-seeking behavior may affect crayfish social hierarchies, feeding, mating, and mortality, which could generate cascading effects on the ecology of many freshwater ecosystems.  相似文献

L-Proline, which is accumulated by Escherichia coli during growth in media of high osmolality, also induces the synthesis of the enzyme degrading it to glutamate. To determine if proline catabolism is inhibited during osmotic stress, proline utilization and the formation of proline dehydrogenase were examined in varying concentrations of NaCl and sucrose. Although the specific growth rate of E. coli with proline as the sole nitrogen source diminished as the solute osmolality increased, a comparable reduction in growth rate occurred with ammonium as the primary nitrogen source. Proline catabolism, as measured in whole cells by the conversion of [14C]proline to [14C]glutamate, was only slightly inhibited by solute osmolalities up to 1.0 osmol/kg; more than 50% of the initial activity was still found at 2.0 osmol/kg. By contrast, the specific activity of proline dehydrogenase in bacteria grown in the presence of added solutes decreased to less than 20% of the control level. This reduction was related to a lower rate of synthesis, but was independent of genes currently known to be involved in osmoregulation or proline metabolism. The specific activities of tryptophanase, beta-galactosidase, and histidinol dehydrogenase were also reduced under similar growth conditions. These results indicate that while proline catabolism is not directly inhibited by high solute concentrations, prolonged exposure to osmotic stress leads to its reduction as part of a more general metabolic response.  相似文献

Summary Umbra limi has an elaborate developement of superficial neuromasts on the head and body. The individual lateral line organs of the head form rows. Intermittant clusters of organs occur on the body. The lateral line consists of free standing organs, which are oriented alternating horizontally and vertically. A lateral line canal is wanting.The fish respond to water currents as small as 1.6 mm/sec. They also orient to fairly strong surface waves.

Die Einzelorgane sind am Kopf in Reihen angeordnet, während sie am Rumpf in Gruppen zusammen stehen. Die Organe der eigentlichen Seitenlinie sind vertikal und horizontal ausgerichtet. Ein Rumpfseitenkanal fehlt.Die Fische reagieren noch auf Wasserströme von 1,6 mm/see. Sie reagieren ebenfalls auf Oberflächenwellen gerichtet.

Salmon liver was chosen for the isolation of 6-pyruvoyl tetrahydropterin synthase, one of the enzymes involved in tetrahydrobiopterin biosynthesis. A 9500-fold purification was obtained and the purified enzyme showed two single bands of 16 and 17 kDa on SDS/PAGE. The native enzyme (68 kDa) consists of four subunits and needs free thiol groups for enzymatic activity as was shown by reacting the enzyme with the fluorescent thiol reagent N-(7-dimethylamino-4-methylcoumarinyl)-maleimide. The enzyme is heat-stable up to 80 degrees C, has an isoelectric point of 6.0-6.3, and a pH optimum at 7.5. The enzyme is Mg2+ -dependent and has a Michaelis constant for its substrate dihydroneopterin triphosphate of 2.2 microM. The turnover number of the purified salmon liver enzyme is about 50 times as high as that of the enzyme purified from human liver. It does not bind to the lectin concanavalin A, indicating that it is free of mannose and glucose residues. Polyclonal antibodies raised against the purified enzyme in Balb/c mice were able to immunoprecipitate enzyme activity. The same polyclonal serum was not able to immunoprecipitate enzyme activity of human liver 6-pyruvoyl tetrahydropterin synthase, nor was any cross-reaction in ELISA tests seen.  相似文献

Volatile anesthetics are known to have immunomodulatory effects in conditions of organ injury. A recent study in an experimental sepsis model has shown remarkably improved survival when mice were exposed to volatile anesthetics. In the present study, we show that hexafluoroisopropanol – a water-soluble primary sevoflurane metabolite – has beneficial effects on the overall survival in a murine model of cecal ligation and puncture. Seven-day survival as well as tissue damage markers including transaminases and high mobility group box protein-1 were assessed as measures of end organ damage. In animals undergoing cecal ligation and puncture procedure hexafluoroisopropanol conditioning - but not late postconditioning 24 hours after sepsis induction - significantly increased survival rate (17% vs. 77%, p = 0.037) and attenuated secretion of organ damage markers. This study shows survival benefits by administration of the metabolite of a volatile anesthetic. If successfully translated, hexafluoroisopropanol might offer interesting therapeutic opportunities in the future treatment of abdominal sepsis.  相似文献
