Variation in heat-shock proteins among species of desert fishes (Poeciliidae, Poeciliopsis) 总被引:4,自引:0,他引:4
Analysis of the heat-shock proteins (hsps) of six closely related species
of Poeciliopsis demonstrated the existence of biochemical diversity in the
hsp100, hsp70, hsp60, and hsp30 protein families among species. Each
species expressed five to seven hsp70-related isoforms. Constitutive 70-kD
isoforms were identical among species, but four different patterns of
heat-inducible isoforms were seen in these six species. Members of the
hsp70 family of molecular chaperones are included among the most highly
conserved proteins known, and the possibility of variation in hsp70 among
closely related species has rarely been addressed. The hsp30 family is
known to be less conserved than the hsp70 family, and, as expected, the
Poeciliopsis hsp30 patterns showed more variation. Most of the hsp30
isoforms characteristic of a particular species were unique to that
species. Hsp100 and hsp60 were identical in five of the species, but
alternate isoforms were found in P. monacha. The small size and limited
geographical distribution of the P. monacha population have probably
contributed to the uniqueness of the monacha pattern. Two of the species
were shown to acquire thermotolerance, the ability to withstand normally
lethal temperatures when subjected to a gradual temperature increase.
Rapid-heating protocols commonly used to establish critical thermal maxima
of organisms do not include this inducible component of thermoresistance
and therefore do not adequately assess an organism's capacity to withstand
thermal stress.

The role of oligomeric procyanidins in the ageing of wine has been examined by the addition of these substances to red and white wines. Observations show that in wines the acid-catalysed C C bond-breaking and bond-making process, characteristic of procyanidin chemistry, is set up and leads to the eventual precipitation of polymers. Secondary metabolism and theories of plant-animal co-evolution are briefly discussed. Current ideas concerning the role of polyphenols as mediators of a general form of chemical defence in plants are outlined. Studies are described of the association of a group of biosynthetically inter-related polyphenols based on gallic acid and D-glucose with the protein bovine serum albumin. The results are interpreted in terms of structure-activity relationships in protein-polyphenol complexation and are used to comment on the co-evolution theory and the position of plant polyphenols in higher plant secondary metabolism and defence. 相似文献

Man, increasingly the limiting element in the military man-machine system, must often operate for several days in a high-risk environment with little or no sleep. It is necessary, therefore, to have some knowledge of the likely effects of sleep deprivation to predict his behaviour and minimize the adverse effects of sleep loss. The early work of the Army Personnel Research Establishment (APRE) concentrated on studying the infantryman in field trials, characterized by more realism and of greater length than previously attempted. Although measures of cognitive functioning were included in these trials, continuous cognitive performance was not assessed, nor was performance on complex tasks. An opportunity to remedy this situation arose because of a newer study concerned with controlling a removely-piloted air vehicle from a ground control station (GCS). A 65-hour experiment was designed during which subjects performed continuously either on the GCS simulator or on a battery of cognitive tests, mood scales, and physiological assessments. Results showed that whereas performance showed the usual deterioration in the test battery, it held up remarkably well on the simulator. Several reasons for this difference are suggested. 相似文献

Murine sperm initiate fertilization by binding to the zona pellucida (mZP), the specialized extracellular matrix of their homologous eggs. O-Glycans occupying two highly conserved vicinal glycosylation sites (Ser-332 and Ser-334) on the mZP glycoprotein designated mZP3 were previously implicated in this interaction. However, recent biophysical analyses confirm that neither site is occupied, implying that an alternate O-glycosylation domain may be operational in native mZP3. Since human ZP3 (huZP3) can substitute for mZP3 in rescue mice to mediate sperm binding, the site specificity of O-glycosylation in both native mZP3 and huZP3 was analyzed using ultrasensitive mass spectrometric techniques. Two O-glycosylation sites in native mZP3, one at Thr-155 and the other within the glycopeptide at positions 161-168 (ATVSSEEK), are conserved in huZP3 derived from transgenic mice. Thus, there is a specific O-glycosylation domain within native mZP3 expressing two closely spaced O-glycans that is very well conserved in an evolutionarily related glycoprotein. In native mZP3, core 2 O-glycans predominate at both sites. However, in huZP3 derived from rescue mice, the O-glycans associated with Thr-156 (analogous to Thr-155 in mZP3) are exclusively core 1 and related Tn sequences, whereas core 2 O-glycans predominate at the other conserved site. This unique restriction of O-glycan expression suggests that sequence differences in the conserved O-glycosylation domains of mZP3 and huZP3 affect the ability of core 2 N-acetylglucosaminyltransferase(s) to extend the core 1 sequence. However, this difference in O-glycosylation at Thr-156 does not affect the fertility or the sperm binding phenotype of eggs derived from female huZP3 rescue mice. 相似文献
