Recent studies have demonstrated that hydrogen sulfide (H2S) produced through the activity of l -cysteine desulfhydrase (DES1) is an important gaseous signaling molecule in plants that could participate in abscisic acid (ABA)-induced stomatal closure. However, the coupling of the DES1/H2S signaling pathways to guard cell movement has not been thoroughly elucidated. The results presented here provide genetic evidence for a physiologically relevant signaling pathway that governs guard cell in situ DES1/H2S function in stomatal closure. We discovered that ABA-activated DES1 produces H2S in guard cells. The impaired guard cell ABA phenotype of the des1 mutant can be fully complemented when DES1/H2S function has been specifically rescued in guard cells and epidermal cells, but not mesophyll cells. This research further characterized DES1/H2S function in the regulation of LONG HYPOCOTYL1 (HY1, a member of the heme oxygenase family) signaling. ABA-induced DES1 expression and H2S production are hyper-activated in the hy1 mutant, both of which can be fully abolished by the addition of H2S scavenger. Impaired guard cell ABA phenotype of des1/hy1 can be restored by H2S donors. Taken together, this research indicated that guard cell in situ DES1 function is involved in ABA-induced stomatal closure, which also acts as a pivotal hub in regulating HY1 signaling.  相似文献

Members of a family of collagen-binding microbial surface components recognizing adhesive matrix molecules (MSCRAMMs) from Gram-positive bacteria are established virulence factors in several infectious diseases models. Here, we report that these adhesins also can bind C1q and act as inhibitors of the classical complement pathway. Molecular analyses of Cna from Staphylococcus aureus suggested that this prototype MSCRAMM bound to the collagenous domain of C1q and interfered with the interactions of C1r with C1q. As a result, C1r2C1s2 was displaced from C1q, and the C1 complex was deactivated. This novel function of the Cna-like MSCRAMMs represents a potential immune evasion strategy that could be used by numerous Gram-positive pathogens.  相似文献

为进一步明确区域土地利用/覆被—景观格局对河流水质影响的空间尺度。选择新疆艾比湖区域为研究对象,以25个水质采样点为中心,建立5种尺度的河流缓冲区并提取不同尺度下的土地利用/覆被-景观格局数据。首先,通过主成分分析获得水环境的主要水质变量。其次,利用冗余分析(RDA)方法探讨研究区不同宽度缓冲区土地利用/覆被—景观格局对河流水质的影响,获得水质管理的有效缓冲区。最后,引入突变点分析方法进一步寻找导致水质变量沿景观梯度突变的特定位置。结果表明:(1)土地利用/覆被—景观格局在不同宽度缓冲区内对河流水质的影响不同。4 km缓冲区土地利用/覆被—景观格局对区域水质有较强的分异解释能力,因此4 km缓冲区的景观格局合理配置对河流水质管理尤为重要。(2)通过偏RDA分析发现4 km缓冲区中,影响区域水质的主要环境变量为景观水平斑块密度、类型水平耕地斑块密度和森林聚集度。(3)在偏RDA分析的基础上,对4 km缓冲区内的景观指数进行突变点分析研究,发现区域景观水平斑块密度为90—105 m/hm~2,类型水平耕地的斑块密度ED值在90—110 m/hm~2、林草地的AI值在70%—90%,是艾比湖区域水质保护的最佳突变值,该值为艾比湖区域水质保护的阈值。本研究从多个角度对新疆艾比区域的土地利用/覆被—景观格局对区域水质的影响进行定量分析,揭示该地区景观生态变化的规律,为区域景观格局优化和土地合理规划提供理论依据,并对改善"丝绸之路经济带"生态环境、实现资源可持续利用具有重要的现实意义和理论价值。  相似文献

Shigella dysenteriae type 1 is the causative agent of the most severe form of bacillary dysentery, which occurs as epidemics in many developing countries. We isolated a bacteriophage from surface water samples from Bangladesh that specifically lyses strains of S. dysenteriae type 1. This phage, designated SF-9, belongs to the Podoviridae family and has a 41-kb double-stranded DNA genome. Further screening of water samples for the prevalence of the phage revealed 9 of 71 (12.6%) water samples which were positive for the phage. These water samples were also positive in PCR assays for one or more S. dysenteriae type 1-specific genes, including ipaBCD and stx1, and live S. dysenteriae type 1 was isolated from three phage-positive samples. The results of this study suggest that phage SF-9 may have epidemiological applications in tracing the presence of S. dysenteriae type 1 in environmental waters.  相似文献

Novel bio-sorbent wheat bran has been successfully utilized for the removal of cadmium(II) from wastewater. The maximum removal of cadmium(II) was found to be 87.15% at pH 8.6, initial Cd(II) concentration of 12.5 mg l-1 and temperature of 20 degrees C. The effect of different parameters such as contact time, adsorbate concentration, pH of the medium and temperature were investigated. Dynamics of the sorption process were studied and the values of rate constant of adsorption, rate constant of intraparticle diffusion and mass transfer coefficient were calculated. Different thermodynamic parameters viz., changes in standard free energy, enthalpy and entropy have also been evaluated and it has been found that the reaction was spontaneous and exothermic in nature. The applicability of Langmuir isotherm showed of monolayer coverage of the adsorbate on the surface of adsorbent. A generalized empirical model was proposed for the kinetics at different initial concentrations.  相似文献

In this communication, light harvesting and photoelectrochemical (PEC) hydrogen generation beyond the visible region are realized by an anisotropic plasmonic metal/semiconductor hybrid photocatalyst with precise control of their topology and heterointerface. Controlling the intended configuration of the photocatalytic semiconductor to anisotropic Au nanorods' plasmonic hot spots, through a water phase cation exchange strategy, the site‐selective overgrowth of a CdSe shell evolving from a core/shell to a nanodumbbell is realized successfully. Using this strategy, tip‐preferred efficient photoinduced electron/hole separation and plasmon enhancement can be realized. Thus, the PEC hydrogen generation activity of the Au/CdSe nanodumbbell is 45.29 µmol cm?2 h?1 (nearly 4 times than the core/shell structure) beyond vis (λ > 700 nm) illumination and exhibits a high faradic efficiency of 96% and excellent stability with a constant photocurrent for 5 days. Using surface photovoltage microscopy, it is further demonstrated that the efficient plasmonic hot charge spatial separation, which hot electrons can inject into CdSe semiconductors, leads to excellent performance in the Au/CdSe nanodumbbell.  相似文献

为了解益智(Alpinia oxyphylla)多糖生物合成途径关键酶功能,对其茎、叶、果实中的多糖含量及其单糖组成进行了研究,并采用Real-Time qPCR分析了益智多糖生物合成关键酶基因的表达模式。结果表明,益智多糖含量依次为果实 > 叶 > 茎,主要由葡萄糖、木糖、甘露糖、半乳糖和阿拉伯糖组成;利用益智转录组数据共获得47 690条unigenes,其中31 892条在NR、Swiss-Prot、KEGG、COG、KOG、GO和Pfam数据库获得注释,其中208个unigenes参与益智多糖的生物合成,涉及15个酶。表达分析表明,所筛选的18个基因在茎、叶、果实中均有表达,14个基因在果实中的表达量最高,以糖基转移酶基因和UDP-葡萄糖焦磷酸化酶基因的表达量最高,且其表达模式与不同组织中葡萄糖含量的变化一致。  相似文献

通过研究葛根资源的遗传多样性和葛根表型性状与分子标记的关联分析,为葛根的分子育种和指纹图谱构建提供理论依据。鉴定分析了127份不同来源葛根资源的10个表型性状,并用ISSR标记研究127份葛根资源的遗传多样性,对ISSR标记与表型性状进行关联分析。表型性状鉴定结果显示葛根资源的10个性状变异大、多样性较好。利用ISSR标记获得多态性条带109条,平均每个引物扩增5.73条、平均Nei's基因多样性0.2085、平均Shannon's指数0.3378,最远遗传距离为0.46。ISSR分子标记聚类分析将127份资源聚为两大类,群体结构分析和PCo A分析结果类似将127份资源分为2个亚群。GLM分析发现3个与茸毛性状关联标记,MLM分析未发现与表型性状关联的标记。本研究收集的资源遗传多样性较好,葛根分子标记聚类结果与地域关系不大,综合GLM和MLM关联分析结果,本试验未发现与表型关联位点。  相似文献

Introduction

The occurrence of skeletal metastases in cancer, e.g. breast cancer (BC), deteriorates patient life expectancy and quality-of-life. Current treatment options against tumor-associated bone disease are limited to anti-resorptive therapies and aimed towards palliation. There remains a lack of therapeutic approaches, which reverse or even prevent the development of bone metastases. Recent studies demonstrate that not only osteoclasts (OCs), but also osteoblasts (OBs) play a central role in the pathogenesis of skeletal metastases, partly by producing hepatocyte growth factor (HGF), which promotes tumor cell migration and seeding into the bone. OBs consist of a heterogeneous cell pool with respect to their maturation stage and function. Recent studies highlight the critical role of pre-OBs in hematopoiesis. Whether the development of bone metastases can be attributed to a particular OB maturation stage is currently unknown.

Methods and Results

Pre-OBs were generated from healthy donor (HD)-derived bone marrow stromal cells (BMSC) as well as the BMSC line KM105 and defined as ALPlow OPNlow RUNX2high OSX high CD166high. Conditioned media (CM) of pre-OBs, but not of undifferentiated cells or mature OBs, enhanced migration of metastatic BC cells. Importantly, HGF mRNA was significantly up-regulated in pre-OBs versus mature OBs, and CM of pre-OBs activated the MET signaling pathway. Highlighting a key role for HGF, CM from HGF-negative pre-OBs derived from the BMSC line HS27A did not support migration of BC cells. Genetically (siMET) or pharmacologically (INCB28060) targeting MET inhibited both HGF- and pre-OB CM- mediated BC cell migration.

Conclusions

Our data demonstrate for the first time a role for pre-OBs in mediating HGF/MET- dependent migration of BC cells and strongly support the clinical evaluation of INCB28060 and other MET inhibitors to limit and/or prevent BC-associated bone metastases.  相似文献

Here, we study the evolution of specialization using realistic computer simulations of bacteria that secrete two public goods in a dynamic fluid. Through this first‐principles approach, we find physical factors such as diffusion, flow patterns and decay rates are as influential as fitness economics in governing the evolution of community structure, to the extent that when mechanical factors are taken into account, (a) generalist communities can resist becoming specialists despite the invasion fitness of specialization; (b) generalist and specialists can both resist cheaters despite the invasion fitness of free‐riding; and (c) multiple community structures can coexist despite the opposing force of competitive exclusion. Our results emphasize the role of spatial assortment and physical forces on niche partitioning and the evolution of diverse community structures.  相似文献
