Abstract A fully resolved cladogram for 19 species in the Charis cleonus group of riodinid butterflies, which have closely parapatric ranges throughout the Amazon basin, is used to derive an area cladogram for the region. This represents the first comprehensive species‐level analysis using insects and results in a hypothesis of Amazonian area relationships that is the most detailed to date. The Charis area cladogram is interpreted as supporting an historical vicariant split between the Guianas and the remainder of the Amazon and then between the upper and lower Amazon. The latter two clades can be further divided into the six most widely recognized areas of endemism and even smaller endemic centers within these, some of which, especially along the Madeira and lower Amazon Rivers, have never been previously hypothesized for butterflies. The overall pattern of historical interrelationships indicated is Guiana + ((Rondônia + (Pará+ Belém)) + (Imeri + (Napo + Inambari))). The area relationships for riodinid butterflies show substantial congruence with those presented from the literature for amphibians, reptiles, birds, primates, rodents, and marsupials, suggesting a common vicariant history for these organisms. A summary area cladogram generated by combining area cladograms for all the aforementioned groups of organisms indicated the pattern of historical interrelationships to be (Guiana + (Rondônia + (Pará+ Belém))) + (Imeri + (Napo + Inambari)). Charis cleonus group species distributions are noticeably larger around the upland periphery of Amazonia and smaller in the central and lower regions. A significant positive correlation between the proportion of range area above 100 m and total range size for each species is used to suggest that past sea‐level rises may explain smaller range sizes in low‐lying regions and that riverine barriers have been important in shaping the current distribution of C. cleonus group species.  相似文献

We have found a new kind of structural motif that appears to be highly conserved among the pyridine nucleotide-linked alpha-amino acid dehydrogenases. This feature is comprised of four atoms closely packed in a planar form. Two of the atoms are donated by the enzyme, one is provided by the substrate (or reactive intermediate), a bound water molecule constitutes the fourth. This tetrad, by virtue of its spatial connectivity, constitutes a two-dimensional machine in contrast to the one-dimensional charge-relay system commonly observed at enzyme active sites. As such, this new motif is capable of more complex behavior permitting a wide variety of possible bonding patterns. Modulation of these potentially variable patterns can lead to qualitative differences in mechanism between structurally similar enzymes, and, in the case of a given enzyme, may constitute the core of its catalytic machinery. We offer a conjecture as to how such a structure may participate in enzymatic catalysis.  相似文献

We have undertaken a systematic proteomic approach to purify and identify secreted factors that are differentially expressed in preadipocytes versus adipocytes. Using one-dimensional gel electrophoresis combined with nanoelectrospray tandem mass spectrometry, proteins that were specifically secreted by 3T3-L1 preadipocytes or adipocytes were identified. In addition to a number of previously reported molecules that are up- or down-regulated during this differentiation process (adipsin, adipocyte complement-related protein 30 kDa, complement C3, and fibronectin), we identified four secreted molecules that have not been shown previously to be expressed differentially during the process of adipogenesis. Pigment epithelium-derived factor, a soluble molecule with potent antiangiogenic properties, was found to be highly secreted by preadipocytes but not adipocytes. Conversely, we found hippocampal cholinergic neurostimulating peptide, neutrophil gelatinase-associated lipocalin, and haptoglobin to be expressed highly by mature adipocytes. We also used liquid chromatography-based separation followed by automated tandem mass spectrometry to identify proteins secreted by mature adipocytes. Several additional secreted proteins including resistin, secreted acidic cysteine-rich glycoprotein/osteonectin, stromal cell-derived factor-1, cystatin C, gelsolin, and matrix metalloprotease-2 were identified by this method. To our knowledge, this is the first study to identify several novel secreted proteins by adipocytes by a proteomic approach using mass spectrometry.  相似文献

Despite enormous progress during the last twenty years in understandingthe mechanistic basis of aquatic animal propulsion—a taskinvolving the construction of a substantial data base on patternsof fin and body kinematics and locomotor muscle function—thereremains a key area in which biologists have little information:the relationship between propulsor activity and water movementin the wake. How is internal muscular force translated intoexternal force exerted on the water? What is the pattern offluid force production by different fish fins (e.g., pectoral,caudal, dorsal) and how does swimming force vary with speedand among species? These types of questions have received considerableattention in analyses of terrestrial locomotion where forceoutput by limbs can be measured directly with force plates.But how can forces exerted by animals moving through fluid bemeasured? The advent of digital particle image velocimetry (DPIV)has provided an experimental hydrodynamic approach for quantifyingthe locomotor forces of freely moving animals in fluids, andhas resulted in significant new insights into the mechanismsof fish propulsion. In this paper we present ten "lessons learned"from the application of DPIV to problems of fish locomotionover the last five years. (1) Three-dimensional DPIV analysisis critical for reconstructing wake geometry. (2) DPIV analysisreveals the orientation of locomotor reaction forces. (3) DPIVanalysis allows calculation of the magnitude of locomotor forces.(4) Swimming speed can have a major impact on wake structure.(5) DPIV can reveal interspecific differences in vortex wakemorphology. (6) DPIV analysis can provide new insights intothe limits to locomotor performance. (7) DPIV demonstrates thefunctional versatility of fish fins. (8) DPIV reveals hydrodynamicforce partitioning among fins. (9) DPIV shows that wake interactionamong fins may enhance thrust production. (10) Experimentalhydrodynamic analysis can provide insight into the functionalsignificance of evolutionary variation in fin design.  相似文献

Recombination occurs at high frequencies in all examined retroviruses. The previously determined homologous recombination rate in one retroviral replication cycle is 4% for markers 1.0 kb apart in spleen necrosis virus (SNV). This has often been used to suggest that approximately 30 to 40% of the replication-competent viruses with 7- to 10-kb genomes undergo recombination. These estimates were based on the untested assumption that a linear relationship exists between recombination rates and marker distances. To delineate this relationship, we constructed three sets of murine leukemia virus (MLV)-based vectors containing the neomycin phosphotransferase gene (neo) and the hygromycin phosphotransferase B gene (hygro). Each set contained one vector with a functional neo and an inactivated hygro and one vector with a functional hygro and an inactivated neo. The two inactivating mutations in the three sets of vectors were separated by 1.0, 1.9, and 7.1 kb. Recombination rates after one round of replication were 4.7, 7.4, and 8.2% with markers 1.0, 1.9, and 7.1 kb apart, respectively. Thus, the rate of homologous recombination with 1.0 kb of marker distance is similar in MLV and SNV. The recombination rate increases when the marker distance increases from 1.0 to 1.9 kb; however, the recombination rates with marker distances of 1.9 and 7.1 kb are not significantly different. These data refute the previous assumption that recombination is proportional to marker distance and define the maximum recombining population in retroviruses.  相似文献

Physical environment and physiological characteristics of marine mammals potentially affect the duration and depth of diving. Härkönen (1987b) proposed a hypothesis that the harbor seal would gain maximum energy by foraging at intermediate depths. To investigate this hypothesis, we studied diving behavior of the Pacific harbor seal (Phoca vitulina ricbardii) during 1995 through 1997 in Monterey Bay, California. Dive depths (n = 13,063 dives) were recorded via time‐depth recorders. Approximately 80% of recorded dives were classified as square dives (type I), which typically were associated with foraging in pinnipeds. Approximately 11% of dives were V dives (type II; 1,402 dives), and the remainder (1,225 dives) were skewed dives (type III and IV). The deepest recorded dive was 481 m, while the greatest duration was 35.25 min. Body mass explained the variability of durations of long dives for females (95th percentile; D95♂=‐5.47 + 0.18 × (mass♀), r2= 0.91, 95% CI for slope = [0.08, 0.28], n= 5) and for males (D95♂=‐5.86 + 0.18 × (mass♀), r2= 0.83, 95% CI for slope = [0.12, 0.24], n= 11). The large proportion of variability in deep dives, however, was explained by body mass only for males (95th percentile; Z95♂=‐363.9 + 6.05 × (mass♀), r2= 0.83, 95% CI for slope = [3.93, 8.17], n= 11) and not for females (Z95,♂=?148.1 +3.11 × (mass♀), r2= 0.58, 95% CI for slope = [‐1.7, 7.9], n= 5, 95% CI for slope= [?1.7, 7.9]). Median depths of presumed foraging dives of harbor seals in the Monterey Bay area were between 5 and 100 m, which were within the range of the previously reported depths for other areas (< 100 m). Our findings generally supported Härkönen's hypothesis that harbor seals forage in the intermediate depth in their environment.  相似文献

We report the finding of two species of Mantoniella from the Weddell Sen and Prydz Bay, Antarctica. As well as M. squamata, a cosmopolitan species, we describe M. antarctica sp. nov., the second species in this genus. M. antarctica is covered with two distinctive types of discoid scales. The flagella bear only the smaller of the scale types whereas the cell body has both types, the larger overlying the smaller.  相似文献
