Ethanolic extracts of 20 medicinal plants were screened for influenza virus NA inhibition and in vitro antiviral activities using MDCK cells in an MTT assay. The vaccine proteins of influenza virus A/New Caledonia/20/99 (H1N1), mouse-adapted influenza virus A/Guizhou/54/89 (A/G)(H3N2) and mouse-adapted influenza virus B/Ibaraki/2/85 (B/I) were used in the NA inhibition assay, and mouse-adapted influenza viruses A/PR/8/34 (H1N1), A/G and B/I were used in the in vitro antiviral assay. The results of the in vitro antiviral assay indicated that the A/G virus was the most susceptible and an extract of the leaf of CS possessed the highest in vitro anti-A/G virus activity (41.98%). Therefore, the A/G virus and the CS extract were selected for studying in vivo anti-influenza virus activity. BALB/c mice were treated with CS extract (100 mg/kg per day, 5 times) orally from 4 hr before to 4 days after infection. CS extract elicited significant production of anti-influenza virus IgG1 antibody in BAW and increased mouse weight compared to oseltamivir (0.1 mg/kg per day) on day 19 or water on days 17–19 of infection. Moreover, CS extract produced a higher anti-influenza virus IgA antibody level in BAW compared to oseltamivir, and a tendency towards an increase in anti-influenza virus IgA compared to water was shown. Partial acid hydrolysis of the anti-complementary acidic heteroglycan, AAFIIb-3, isolated from the leaves of Artemisia princeps PAMP gave the oligosaccharides Gal-(1→6)-Gal, Gal-(1→6)-Gal-(1→6)-Gal, GalA-(1→4)-Rha, GalA-(1→2)-Rha, GlcA-(1→4)-Gal, GlcA-(1→4)-Rha, GlcA-(1→6)-Gal, and GlcA-(1→4)-Xyl. On methylation of AAFIIb-3 without de-esterification, 4-linked and 3,6-disubstituted galactan, 3-linked galactan, 4-linked galactan, and branched arabinan-rich fragments were obtained. The results of base-catalysed β-elimination indicated that AAFIIb-3 has a backbone consisting of 4-linked GalA and 2-linked Rha to which a highly branched arabino-3,6-galactan and arabino-4-galactan are linked at positions 4 of some 2-linked Rha units. Xyl-(1→4)-GalA, GlcA-(1→4)-Xyl-GalA, and →3)-Gal-(1→4)-GalA might also be joined to other 2-linked Rha at the same position. Some 6-linked and 4-linked Gal were terminated by GlcA. 相似文献

Abstract The ability of Lactobacillus gasseri , a dairy lactic acid bacterium, to induce interferon (IFN) was investigated in murine macrophage cultures. IFN α was substantially induced by some strais of L. gasseri and the titers were the highest at a concentration of 100 μg ml−1 of L. gasseri DSM20243T . The expression of mRNA encoding IFN α was detected in spleen-macrophages (SP-M θ ) and Peyer's patch-adherent cells stimulated with L. gasseri DSM20243T . Actinomycin D and cycloheximide added to SP-M θ cultures showed that the mRNA was synthesized by 0.5 h, and that IFN α was produced within 3 to 6 h after the stimulation with L. gasseri DSM20243T . The results support the notion that dairy products containing L. gasseri can be 'physiologically functional foods'. 相似文献

Bioactive pectic polysaccharides have been isolated from Chinese herbs, and their structure, activity and modes of action have been studied. Complement-activating pectin from Angelica acutiloba contained a variety of neutral galactosyl chains which attached to the rhamnogalacturonan core (ramified region), and these neutral galactosyl chains were essential for the expression of complement activating activity, but the polygalacturonan moiety modulated the mode of activation by the ramified region. Another pectin-like polysaccharide, bupleuran 2IIb from Bupleurum falcatum showed a potent immune complex clearance-enhancing activity. Bupleuran 2IIb may enhance Fc receptor expression on the macrophage surface via the ramified region as an active site. An anti-ulcer pectin, bupleuran 2IIc from B. falcatum consists mainly of partially branched polygalacturonan in addition to the ramified region and the rhamnogalacturonan II-like region, and the polygalacturonan region may contribute somewhat to the expression of activity. Collectively considered results have indicated that the pharmacological activity of each of the pectic polysaccharides may depend on their fine chemical structure. 相似文献

Chronic rotavirus infection of an infant with severe combined immunodeficiency (SCID) was studied by virological examinations in association with long-term observation of his symptoms and immune status. During eleven months of hospitalization, the patient was suffering from incurable severe diarrhea with persisting excretion of rotaviruses detected by electron microscopy and the reversed-passive hemagglutination (R-PHA) test and had transient hepatitis symptom despite multiple administrations of human gammaglobulin and high calorie fluids. The detected viruses were morphologically recognized as rotavirus with double capsid structure. Polyacrylamide gel electrophoretic (PAGE) analysis of their genomic RNAs showed the long electropherotype of group A virus with abnormal migration profiles changing considerably from the early to the late phase of illness: (1) The 11th segment became undetectable; (2) the molecular weight of the 6th segment slightly increased; (3) seven to fourteen extra segments appeared; and (4) PAGE patterns of viral genomic RNAs changed every three or four months. These findings suggest that chronic infection with rotavirus accompanied the generation of extra viral genomic segments and their unusual assortments in an immunodeficient host. 相似文献
