Several non-Ewald methods for calculating electrostatic interactions have recently been developed, such as the Wolf method, the reaction field method, the pre-averaging method, and the zero-dipole summation method, for molecular dynamics simulations of various physical systems, including biomolecular systems. Sequence characterized amplified region markers tightly linked to the mating factors of Lentinula edodes. 总被引:1,自引:0,他引:1
Detecting the mating types in shiitake, Lentinula edodes (Berk.) Pegler, is important for making progress in the breeding of this mushroom and determining the compatibility of the pair to cross. Shiitake is a tetrapolar fungus with two unlinking mating factors, A factor and B factor. We screened molecular markers linked to the mating factors using the randomly amplified polymorphic DNA (RAPD) method to develop the mating type identification procedure. Using 147 oligonucleotide primers, a total of 6 linkage markers for the shiitake mating factors, 4 markers for the A factor and 2 markers for the B factor, were discovered with a logarithm of the odds threshold of 3.0 for linkage. Two RAPDs that perfectly segregated with each mating factor among 72 basidiospore strains were detected. Both of these RAPDs were cloned and sequenced to convert them to the sequence characterized amplified region (SCAR) markers. Four primers, two sets of primers, were designed according to the internal sequences of two RAPDs tightly linking to the A factor or B factor. Consequently, we determined the polymerase chain reaction condition for multiplex analyses of these SCAR markers. 相似文献

Two types of experiment were carried out to examine whetheror not the inactivation of photosynthesis is related to lossof chloroplasts during foliar senescence of rice seedlings.Levels of both soluble and insoluble leaf proteins decreasedduring senescence, the loss of the soluble proteins being fasterthan that of the insoluble ones. There was a good positive correlationbetween the rate of oxygen evolution and the level of solubleproteins. The inactivation of photosynthesis was also linearlyrelated to the loss of a major fraction of insoluble proteins.Thus, the loss of photosynthetic ability is ascribable to thedegradation of relevant proteins and enzymes during leaf senescence.Electron microscopy revealed that senescence caused the disorientationof the grana and stroma thylakoids, a decrease in the numberof starch granules, and an increase in the size and number ofplastoglobuli. Large grana consisting 20 to 30 thylakoids appearedin aged leaves. In addition to these changes in ultrastructure,there was a significant decrease in the size of chloroplasts.Furthermore, the number of chloroplasts in mesophyll cells wasalso notably reduced during senescence. Thus, the loss of leafproteins and inactivation of photosynthesis are both relatedto the decrease in the total mass of chloroplasts during senescenceof rice seedlings.

Summary The plastokinesis (kinesis of chloroplasts) of a raphidophyte alga,Heterosigma akashiwo, was studied by electron microscopy using rapid freezing and freeze-substitution techniques. The chloroplasts are enveloped by two pairs of tightly appressed double membranes, the inner and the cytoplasmic outer pair. The inner pair constricts to divide in advance of the outer pair. By observation of serial sections an electron-opaque, annular structure (plastid-dividing ring) was observed at the isthmus of constricting chloroplasts, girdling the periplastidal outer surface of the inner pair of the four surrounding membranes. These observations suggest that the mechanisms underlying the constriction of the inner and outer pair may differ from each other. The localization of the annular structure (plastid-dividing ring) suggests that the inner pair of the surrounding membranes may be homologous to the double envelope membranes of the chloroplasts of Chlorophyta and Rhodophyta. In addition these findings provide a new evidence supporting the secondary endosymbiosis hypothesis for the origin of the chloroplasts in chromophyte algae. 相似文献

Thirteen polysaccharides isolated from an extract of the aerial portions of Astragalus mongholics Bunge demonstrated immunomodulating activity against Peyer’s patch immunocompetent cells. Nine of the active polysaccharide fractions were composed of either arabinogalactans, pectic arabinogalactans or pectins. The activities of the arabinogalactans and pectic arabinogalactans were associated with β-d-(1 → 3)-galactan moieties branched with β-d-(1 → 6)-galactooligosaccharide side-chains having degrees of polymerization of 8 or less. Degradation of the β-d-(1 → 3)-galactan or β-d-(1 → 6)-galactosyl side-chains in the arabinogalactans significantly decreased immunomodulating activity. Rhamnogalacturonan I (RG-I) with β-d-(1 → 3,6)-galactosyl side-chains having terminal β-d-GlcA showed activity in the pectin-enriched fractions. Interestingly, the terminal GlcA was not required for activity of the arabinogalactan-enriched fractions, suggesting at least two different immunomodulating structures. 相似文献

The amount of estradiol and progesterone in the systemic blood plasma of six adult female Japanese monkeys was measured by
radioimmunoassay. Data on heterosexual pairing tests was also collected and examined for correlation with these hormones.
The relationship between ovarian hormones and frequency of ejaculation varied with each female. In nine normal menstrual cycles,
the frequencies of invitation and approach by the female reached peaks on the day of the estradiol surge, after which they
rapidly decreased. Frequencies of male behaviors including approach, leaving, invitation, and yawning significantly decreased
during the luteal phase, as compared with those at the midcycle. Grooming by both sexes of the partner reached low points
during the late follicular phase. The number of ejaculations per test increased significantly at the midcycle and reached
a maximum a few days after the estradiol surge. The present results suggest that female proceptivity is highly correlated
with the amount of estradiol, but her attractiveness is little correlated with estradiol and is lowered by progesterone. 相似文献

Three species of Agaricales and Boletales are fully described and illustrated from Japan: (1) Clitopilus
vernalis sp. nov. produces collybioid basidiomata with pruinose, greyish-yellow pileus and stipe, obscurely undulate basidiospores,
and has a lignicolous habit fruiting in spring on dead decorticated logs of Pinus densiflora; (2) Favolaschia gelatina sp. nov. (section Anechinus subsection Rubrinae) has pleurotoid, astipitate basidiomata with transparent, elastic, jelly-like flesh, and truly poroid hymenophore; (3) Boletellus betula is a new record for Japan, growing in Quercus crispula and Pinus densiflora forests. 相似文献

Synopsis In the main habitat of the anemonefishes Amphiprion, their movements between host sea anemones are generally restricted because of the low population density of hosts and high predation pressures. On the contrary, movements of A. clarkii between hosts were usual in the present study area (temperate waters of southern Japan), where host anemones are abundant. The general social unit of the anemonefishes is an isolated group consisting of a monogamous breeding pair and a varying number of nonbreeders. In the present study area, however, monogamous pairs established territories almost contiguous to others and nonbreeders had home ranges on the outskirts of the pairs' territories. The high host population density allows A. clarkii to move between hosts for searching for mates and acquiring additional mates. Most pair bonds lasted for more than 6 months, but 13% of the pairs separated because of migration of a mate to another territory. Bigamy occasionally originated from a penetration into a territory of a breeder by a mated neighbor of the opposite sex after the former's mate loss. Among 18 males who had lost their mates, only 3 changed sex and others re-paired with immigrant females, migrated to unmated females' territories or invaded pairs' territories. In the present study area, sex change to female is not the best way for an unmated male to increase his future reproductive success because of a loss of time spent on sex change and an opportunity to re-pair with new mates larger than himself, but is adaptively maintained as the best of a bad situation for the unmated male. 相似文献
