L6 skeletal muscle myoblasts stably overexpressing glucose transporter GLUT1 or GLUT4 with exofa- cial myc-epitope tags were characterized for their response to insulin. In clonally selected cultures, 2-deoxyglucose uptake into L6-GLUT1myc myoblasts and myotubes was linear within the time of study. In L6-GLUT1myc and L6-GLUT4myc myoblasts, 100 nmol/L insulin treatment increased the GLUT1 content of the plasma membrane by 1.58±0.01 fold and the GLUT4 content 1.96±0.11 fold, as well as the 2-deoxyglucose uptake 1.53±0.09 and 1.86±0.17 fold respectively, all by a wortmannin-inhibitable manner. The phosphorylation of Akt in these two cell lines was increased by insulin. L6-GLUT1myc myoblasts showed a dose-dependent stimulation of glucose uptake by insulin, with unaltered sensitiv- ity and maximal responsiveness compared with wild type cells. By contrast, the improved insulin re- sponsiveness and sensitivity of glucose uptake were observed in L6-GLUT4myc myoblasts. Earlier studies indicated that forskolin might affect insulin-stimulated GLUT4 translocation. A 65% decrease of insulin-stimulated 2-deoxyglucose uptake in GLUT4myc cells was not due to an effect on GLUT4 mobi- lization to the plasma membrane, but instead on direct inhibition of GLUT4. Forskolin and dipyridamole are more potent inhibitors of GLUT4 than GLUT1. Alternatively, pentobarbital inhibits GLUT1 more than GLUT4. The use of these inhibitors confirmed that the overexpressed GLUT1 or GLUT4 are the major functional glucose transporters in unstimulated and insulin-stimulated L6 myoblasts. Therefore, L6-GLUT1myc and L6-GLUT4myc cells provide a platform to screen compounds that may have differ- ential effects on GLUT isoform activity or may influence GLUT isoform mobilization to the cell surface of muscle cells. 相似文献

通过对上海市的苏州河支流绥宁河治理段与非治理段水体叶绿素a分粒级分析,探讨了生态修复对水体粒径分级叶绿素a的影响.结果表明,非治理和对照以及治理采样点微型和微微型浮游植物叶绿素a占总叶绿素a的百分比均值分别为85.232、92.402和95.205%,其中微型浮游植物叶绿素a占总叶绿素a的百分比均值分别为78.460、87.943和87.211%,对全河叶绿素a的贡献平均为84.538%,是该水体叶绿素a生物量的最大贡献者;网采浮游植物对全河叶绿素a的贡献仅为9.054%.生态修复工程试验使网采浮游植物相对生物量减少,微型浮游植物相对生物量保持稳定,而微微型浮游植物相对生物量增多,对超微型浮游植物的影响不大,微型和微微型浮游植物对工程试验的反应最敏感. 相似文献

Pardiñas, Valenzuela, and Salazar‐Bravo (2017) are concerned that eventual differences in species mean body masses and in the phylogenetic hypothesis used in Maestri et al. (2016)—compared with those available on other potential sources—could affect the results of our original article. Here, we used a new phylogenetic hypothesis to conduct the same analyses of the original article, and we randomly sampled 1000 values of body mass within approximately 35% upper and lower intervals around the mean body mass for each species included in our database. We show that our previous results and conclusions are robust and valid, and they persist despite uncertainty in mean body mass estimation. We argue that sampling variation and uncertainty in both species mean body mass estimation and phylogenetic hypothesis are to be expected and should not always be confused with inaccuracies. 相似文献

In Brazil, 4.5% of the AIDS-related opportunistic infections are caused by Cryptococcus neoformans. This pathogen is a ubiquitous environmental basidiomycetous encapsulated yeast, commonly found in soil and avian excreta. The present study investigates further the population structure of clinical and environmental C. neoformans isolates from south Brazil. One hundred five clinical and 19 environmental (pigeon excreta and Eucalyptus spp.) isolates from the Brazilian state Rio Grande do Sul were characterized based on morphological, biochemical, molecular and serological data. The majority of the clinical and environmental isolates analyzed belonged to C. neoformans var. grubii serotype A (89.5 and 52.6%, respectively), were mating type alpha (98.1 and 94.7%, respectively) and were phospholipase-positive (94.3 and 73.7%, respectively). PCR-fingerprinting with the microsatellite-specific primer M13 and the minisatellite-specific primer (GACA)(4) grouped the majority of the isolates into the molecular type VNI (89.5 of the clinical and 52.6% of the environmental isolates). Our results add considerable new information to the few available data on ecology, molecular biology and epidemiology of C. neoformans in the southern region of Brazil. 相似文献

Mesodermal differentiation of dorsal marginal zone (DMZ) before and after invagination was analyzed by a series of combination experiments with different kinds of ectoderm.
Lower DMZ of early gastrula didn't show any axial-mesoderm (notochord and somitic mesoderm) but lateral mesoderm (mesenchyme, mesothelium, or blood cells) in combinant with non-competent ventral ectoderm, while combinant with competent ectoderm was found to have well-differentiated axial-mesoderm with deutero-spinocaudal neurals. The axial-mesoderms have origin in the ectoderm. Uninvaginated DMZ of middle gastrula also showed difference in mesodermal differentiation between competent and non-competent ectoderms; axial-mesoderm differentiation was much better in competent than in non-competent. The axial-mesoderm originated from the uninvaginated DMZ. Archenteron roof of late gastrula showed regional difference in mesodermal differentiation in both combinants with competent and non-competent. The present study further demonstrated that there was regionality in promoting effect of induced neurectoderm on axial-mesoderm differentiation of invaginated archenteron roof.
The present experiments suggest that the cranio-caudal and dorso-ventral axis formations of amphibian mesoderm are finally determined by sequential and reciprocal interactions between the mesodermal anlage and the overlying ectoderm. It should be also shown that lower DMZ acts to trigger a series of the sequential interactions during primary embryonic induction. 相似文献

Nitric oxide (NO), a vital cell‐signalling molecule, has been reported to regulate toxic metal responses in plants. This work investigated the effects of NO and the relationship between NO and mitogen‐activated protein kinase (MAPK) in Arabidopsis (Arabidopsis thaliana) programmed cell death (PCD) induced by cadmium (Cd2+) exposure. With fluorescence resonance energy transfer (FRET) analysis, caspase‐3‐like protease activation was detected after Cd2+ treatment. This was further confirmed with a caspase‐3 substrate assay. Cd2+‐induced caspase‐3‐like activity was inhibited in the presence of the NO‐specific scavenger 2‐(4‐carboxyphenyl)‐4,4,5,5‐tetramethylimidazoline‐1‐oxyl‐3‐oxide (cPTIO), suggesting that NO mediated caspase‐3‐like protease activation under Cd2+ stress conditions. Pretreatment with cPTIO effectively inhibited Cd2+‐induced MAPK activation, indicating that NO also affected the MAPK pathway. Interestingly, Cd2+‐induced caspase‐3‐like activity was significantly suppressed in the mpk6 mutant, suggesting that MPK6 was required for caspase‐3‐like protease activation. To our knowledge, this is the first demonstration that NO promotes Cd2+‐induced Arabidopsis PCD by promoting MPK6‐mediated caspase‐3‐like activation. 相似文献

为了了解栲树的更新方式和更新动态,研究了栲树的生长特征和种群年龄结构.结果表明:栲树种群的年龄结构呈“间歇型”,经历了两个死亡高峰,并存在一个长达30年的断层;栲树的生长受光照的影响,具有很强的可塑性;由于林下光照弱且在垂直空间上不存在差异,栲树生长5~8年后进入生长的第1个抑制期,其年高生长速度可小于0.1 m,并可维持10年;栲树生长的第1个抑制期的起始时间对应着种群第1个死亡高峰期的结束时间,而其结束时间对应着种群第2个死亡高峰期的起始时间,表明栲树生长特征是影响其种群年龄结构的关键因素. 相似文献

Abstract Araucaria Forest expansion over grassland takes place under wet climate conditions and low disturbance and it is hypothesized that isolated trees established on grassland facilitate the establishment of forest woody species beneath their canopies. Forest with Araucaria angustifolia is a particular type of Brazilian Atlantic Forest and the main forest type on the highland plateau in south Brazil, often forming mosaics with natural Campos grassland. The objectives of this paper were to evaluate the role of isolated shrubs and trees as colonization sites for seedlings of Araucaria Forest woody species on grassland, to determine which species function as preferential nurse plants in the process and the importance of vertebrate diaspore dispersal on the structure of seedling communities beneath nurse plants. The study was conducted in São Francisco de Paula, Rio Grande do Sul State, where we sampled isolated shrubs and trees in natural grassland near Araucaria Forest edges. Seedlings were counted and identified, and seedling diaspore dispersal syndromes, size and colour were registered. We detected 11 woody species with a potential role in nucleating grassland colonization by forest species. Beneath the canopies of nurse plants more forest species seedlings were found compared with open field grassland and the seedlings had diaspores mostly dispersed by vertebrates. Also, more seedlings were found under the canopy of A. angustifolia than beneath other nurse plant species. We conclude that A. angustifolia trees established on grassland act as nurse plants, by attracting disperser birds that promote colonization of the site by other forest species seedlings, and that under low level of grassland disturbance, conservation of frugivorous vertebrate assemblages may increase forest expansion over natural grassland and also facilitate the regeneration of degraded forest areas. 相似文献

An analysis of the partitioning of carbohydrates in annual andperennial cotton was made to ascertain the distribution of assimilatesand constitution of reserves. Root/shoot dry matter ratio ishigh in perennial cotton and this plant shows a preferentialaccumulation of dry matter in roots corresponding to its adaptationto drought. Starch content is also higher in perennial cottonroots than in annual. It can be said that the earlier maturingthe cultivar, the lower the root/shoot ratio and the lower thestarch content. Nevertheless, at the whole plant level in annualcotton the starch content is highest in leaves where it is accumulatedbefore migration, and stem wood, and lowest in root and bark.While starch content in roots of annuals declines after 3 months,it is still increasing in perennials. Accumulation of carbohydratesas reserve material can be modified by selection and such selectionis accompanied by an increase in the activities of ß-amylasein exporting organs: leaves, woody tissue of the stem, and barkbut not in roots. Invertase activities were highest in leavesbut did not respond to selection. Non-irrigated cotton had ahigher activity of ß-amylase in leaves and stem woodcorresponding to the mobilization of reserve assimilates. Smallerincreases were observed in the activity of invertase. High yieldingannual cottons show a higher activity of ß-amylaseand invertase in leaves corresponding to a higher capacity ofassimilate transfer. Also a comparison was made from emergenceto 4 months of the partitioning of carbohydrates between leaf,stem and roots in annual and perennial cotton. In conclusionperennial cotton apparently owes its drought resistance to apartitioning of assimilates that favours the growth of the rootsystem and the accumulation of starch reserves in roots. Indoor and outdoor winter activity of Culicoides spp. (Diptera: Ceratopogonidae) in central Italy was investigated in order to evaluate whether indoor activity might account for the overwintering of bluetongue virus, as has been hypothesized by some authors. Weekly Culicoides collections were performed at three farms over three consecutive winter seasons. At each farm, two black‐light traps were operated simultaneously, indoors and outdoors. Culicoides were identified using both morphological and molecular means. The Culicoides obsoletus group accounted for 98.2% of sampled specimens. Within this group, C. obsoletus s.s. accounted for 56.8% and Culicoides scoticus for 43.2% of samples. Nulliparous, parous and engorged females were caught throughout the entire winter, both indoors and outdoors. At times, indoor catch sizes outnumbered outdoor collections. A significant inverse correlation was found between minimum temperature and the proportion of indoor Culicoides of the total midge catch, thus indicating that lower outdoor temperatures drive Culicoides midges indoors. High rates of engorged females were recorded indoors, possibly as the result of the propensity of C. obsoletus females to feed indoors. Higher proportions of parous females were found in indoor than in outdoor catches, indicating higher survival rates indoors and, consequently, higher vectorial capacities of midges sheltering indoors compared with those remaining outdoors. 相似文献
