Nicotine Effects on Dopamine Clearance in Rat Nucleus Accumbens   总被引:2,自引:0,他引:2  
Abstract: In vivo voltammetry was used to measure the clearance of exogenously applied dopamine (DA) in the nucleus accumbens following acute systemic nicotine administration in urethane-anesthetized rats. The IVEC-5 system was used for continuous in vivo electrochemical measurements. A finite amount of DA was pressure-ejected (25–100 nl, 200 µ M barrel concentration) at 5-min intervals from micropipettes (tip diameter, 10–15 µm) positioned 250 ± 50 µm from the recording electrode. The peak DA concentration after each DA ejection was significantly decreased in rats following nicotine, but not in rats given saline. In addition, when mecamylamine was administered 20 min before nicotine it clearly antagonized nicotine effects. These results suggest that nicotine may actually facilitate DA transporter systems within the nucleus accumbens.  相似文献

Species with large eggs and nonfeeding larvae have evolved many times from ancestors with smaller eggs and feeding larvae in numerous groups of aquatic invertebrates and amphibians. This change in reproductive allocation and larval form is often accompanied by dramatic changes in development. Little is known of this transformation because the intermediate form (a facultatively feeding larva) is rare. Knowledge of facultatively feeding larvae may help explain the conditions under which nonfeeding larvae evolve. Two hypotheses concerning the evolutionary loss of larval feeding are as follows: (1) large eggs evolve before modifications in larval development, and (2) the intermediate form (facultatively feeding larva) is evolutionarily short-lived. I show that larvae of a heart urchin, Brisaster latifrons, are capable of feeding but do not require food to complete larval development. Food for larvae appears to have little effect on larval growth and development. The development, form, and suspension feeding mechanism of these larvae are similar to those of obligate-feeding larvae of other echinoids. Feeding rates of Brisaster larvae are similar to cooccurring, obligate-feeding echinoid larvae but are low relative to the large size of Brisaster larvae. The comparison shows that in Brisaster large egg size, independence from larval food, and relatively low feeding rate have evolved before the heterochronies and modified developmental mechanisms common in nonfeeding echinoid larvae. If it is general, the result suggests that hypotheses concerning the origin of nonfeeding larval development should be based on ecological factors that affect natural selection for large eggs, rather than on the evolution of heterochronies and developmental novelties in particular clades. I also discuss alternative hypotheses concerning the evolutionary persistence of facultative larval feeding as a reproductive strategy. These hypotheses could be tested against a phylogenetic hypothesis.  相似文献

The odd-skipped (odd) gene, which was identified on the basis of a pair-rule segmentation phenotype in mutant embryos, is initially expressed in the Drosophila embryo in seven pair-rule stripes, but later exhibits a segment polarity-like pattern for which no phenotypic correlate is apparent. We have molecularly characterized two embryonically expressed odd-cognate genes, sob and bowel (bowl), that encode proteins with highly conserved C(2)H(2) zinc fingers. While the Sob and Bowl proteins each contain five tandem fingers, the Odd protein lacks a fifth (C-terminal) finger and is also less conserved among the four common fingers. Reminiscent of many segmentation gene paralogues, the closely linked odd and sob genes are expressed during embryogenesis in similar striped patterns; in contrast, the less-tightly linked bowl gene is expressed in a distinctly different pattern at the termini of the early embryo. Although our results indicate that odd and sob are more likely than bowl to share overlapping developmental roles, some functional divergence between the Odd and Sob proteins is suggested by the absence of homology outside the zinc fingers, and also by amino acid substitutions in the Odd zinc fingers at positions that appear to be constrained in Sob and Bowl.  相似文献

Analyses of RFLPs, isozymes, morphological markers and chromosome pairing were used to isolate 12 Triticum aestivum cv Chinese Spring (genomes A, B, and D)-T. peregrinum (genomes Sv and Uv) disomic chromosome addition lines. The evidence obtained indicates that each of the 12 lines contains an intact pair of T. peregrinum chromosomes. One monosomic addition line, believed to contain an intact 6Sv chromosome, was also isolated. A CS-7Uv chromosome addition line was not obtained. Syntenic relationships in common with the standard Triticeae arrangement were found for five of the seven Sv genome chromosomes. The exceptions were 4Sv and 7Sv. A reciprocal translocation exists between 4S1 and 7S1 in T. longissimum and evidence was obtained that the same translocation exists in T. peregrinum. In contrast, evidence for syntenic relationships in common with the standard Triticeae arrangements were found for only one Uv chromosome of T. peregrinum.; namely, chromosome 2Uv. All other Uv genome chromosomes are involved in at least one translocation, and the same translocations were found in the U genome of T. umbellulatum. Evidence was also obtained indicating that the centromeric regions of 4U and 4Uv are homoeologous to the centromeric regions of Triticeae homoeologous group-6 chromosomes, that the centromeric regions of 6U and 6Uv are homoeologous to the centromeric regions of group-4 chromosomes, and that 4U and 4Uv are more closely related overall to Triticeae homoeologous group-6 chromosomes than they are to group-4 chromosomes.  相似文献

Updated locality records of species of Metadiaptomus and Tropodiaptomus on the African continent confirm the generally disjunct distribution of these two taxa as recognised by Dumont (1980) in North Africa. Distributional data for southern Africa reveal little range overlap between these two genera. Apart from two south western Cape taxa, species of Metadiaptomus are largely confined to upland, higher latitude, semi-arid or arid warm subtemperate regions, while species of Tropodiaptomus generally occupy moist, lower-lying, lower latitude subtropical regions. Separation along latitudinal and/or altitudinal axes implicates temperature as a controlling factor, while separation on the precipitation axis suggests the importance of habitat permanence. Using a multiple regression equation derived for African waters to predict water temperature from latitude and altitude, it is shown that the two genera tend to separate around the 20 °C mean annual temperature isotherm. Additional factors influencing distribution (habitat permanence, water quality, competition and predation) are discussed.  相似文献

The temperature-dependence of development was studied in two ecologically divergent freshwater calanoids, Metadiaptomus meridianus (Douwe) and Tropodiaptomus spectabilis (Kiefer). Egg durations were determined between 10 and 35 °C, and food satiated post-embryonic development times between 12 and 32 °C. All responses were basically inverse monotonic functions of temperature, adequately described by Blehrádek's equation. M. meridianus generally developed faster than T. spectabilis. Its egg development was faster at all temperatures, and while its naupliar durations were shorter only up to ± 15 °C, its overall post-embryonic development was more rapid up to ± 24 °C in females and ± 28 °C in males. Relatively speaking, however, T. spectabilis is clearly more warm-adapted than M. meridianus. The respective distributions (warm subtropical lowlands vs cooler uplands) of these copepods in the southern African subcontinent, as well as reversible switches between these species observed in two Natal impoundments are consistent with their contrasting thermal responses, although additional considerations apply in respect of the species alternations. T. spectabilis was replaced by M. meridianus in L. Midmar in spring 1981 and 1989, and in L. Albert Falls in spring 1990. Reciprocal replacements occurred in Midmar in autumn 1984, and in Albert Falls in late summer 1991. Both spring switches in Midmar coincided with cool spring temperatures, although the consequent shifts in growth rate advantage predicted from the measured temperature-duration responses only partly explain the switches in this warm-temperate reservoir. Parasitism of T. spectabilis by an ellobiopsid was observed during both switching events in Midmar, and perhaps augmented the change, although its effects on the host are indeterminate. Both species showed exactly parallel temporal changes in fecundity during the recent switches in both reservoirs, indicating closely similar trophic niches in the adults at least, and mitigating the possibility of trophic influences as determinants of the replacement. A dramatic but inexplicable increase (around 50% at 20 °C) in the development time of T. spectabilis was noted between 1988 and 1990, and perhaps contributed too.The protracted historical dominance of T. spectabilis in thermally suboptimal conditions in Midmar is ascribed to a general competitive superiority presumed from its K-selected attributes, in contrast to the r-selection evident in M. meridianus. This alternation between species with contrasting life styles is of fundamental ecological interest. Studies on Albert Falls, commenced in 1989, suggest an even greater competitive superiority of T. spectabilis, in keeping with the warmer conditions in this larger sister reservoir below Midmar.  相似文献

The mechanism of CD4-mediated fusion via activated human immunodeficiency virus type 1 (HIV-1) gp41 and the biological significance of soluble CD4 (sCD4)-induced shedding of gp120 are poorly understood. The purpose of these investigations was to determine whether shedding of gp120 led to fusion activation or inactivation. BJAB cells (TF228.1.16) stably expressing HIV-1 envelope glycoproteins (the gp120-gp41 complex) were used to examine the effects of pH and temperature on sCD4-induced shedding of gp120 and on cell-to-cell fusion (syncytium formation) with CD4+ SupT1 cells. sCD4-induced shedding of gp120 was maximal at pH 4.5 to 5.5 and did not occur at pH 8.5. At physiologic pH, sCD4-induced shedding of gp120 occurred at 22, 37, and 40 degrees C but neither at 16 nor 4 degrees C. In contrast, syncytia formed at pH 8.5 (maximally at pH 7.5) but not at pH 4.5 to 5.5. At pH 7.5, syncytia formed at 37 and 40 degrees C but not at 22, 16, or 4 degrees C. Preincubation of cocultures of TF228.1.16 and SupT1 cells at 4, 16, or 22 degrees C before the shift to 37 degrees C resulted in similar, increased, or decreased syncytium formation, respectively, compared with the control. Furthermore, an activated intermediate of CD4-gp120-gp41 ternary complex may form at 16 degrees C; this intermediate rapidly executes fusion upon a shift to 37 degrees C but readily decays upon a shift to the shedding-permissive but fusion-nonpermissive temperature of 22 degrees C. These physicochemical data indicate that shedding of HIV-1 gp120 is not an integral step in the fusion cascade and that CD4 may inactivate the fusion complex in a process analogous to sCD4-induced shedding of gp120.  相似文献
