The effects of ovariectomy (Ovx) and of ovariectomy followed by chronic estradiol dipropionate administration (Ovx EDP) on the structure and function of the pituitary-thyroid axis were examined in the rat. Pituitary TSH cells and thyroid tissue were histologically, immunohistochemically and stereologically investigated. Serum TSH and T(4) levels were determined by RIA. Ovx did not affect pituitary weight, but subsequent treatment with EDP led to its more than two-fold increase (p<0.05). After ovariectomy, the cellular volume of pituitary TSH-immunoreactive cells increased by 28%, p<0.05 compared to sham-operated animals (SO). Treatment of Ovx rats with EDP partially reversed this change. However, the relative volume density of thyrotrophs decreased in comparison to the Ovx and SO groups (by 18% and 23%, p<0.05, respectively). No statistically significant differences in serum TSH levels were observed between the experimental groups. In thyroid tissue both peripheral and central follicles responded to Ovx and EDP treatments. Compared to SO rats, the relative volume densities of the follicles and colloid were increased (by 14% and 30%, p<0.05, respectively) in Ovx rats. Chronic EDP treatment of Ovx rats reversed these changes to the pre-ovariectomy state. Hyperplasia of thyroid follicular cells and a significant reduction (by 21%, p<0.05) of the serum level of T(4) were detected. In conclusion, estradiol deficiency and chronic treatment affected pituitary TSH cells and thyroid tissue. The sum effect of Ovx on the pituitary-thyroid axis was slightly stimulatory. Subsequent EDP treatment decreased thyroid functioning but at the same time preserved serum TSH at the control level.  相似文献

Gallic acid-based indanone derivatives have been synthesised. Some of the indanones showed very good anticancer activity in MTT assay. Compounds 10, 11, 12 and 14 possessed potent anticancer activity against various human cancer cell lines. The most potent indanone (10, IC50 = 2.2 μM), against MCF-7, that is, hormone-dependent breast cancer cell line, showed no toxicity to human erythrocytes even at higher concentrations (100 μg/ml, 258 μM). While, indanones 11, 12 and 14 showed toxicities to erythrocytes at higher concentrations.  相似文献

Polyploidization is an event that developed in plants during evolutionary path which confer them better survivorship than diploids towards the harsh environmental conditions. Regarding this approach polyploids acquire most of appealing facts in it which are the special concern of biologists. Present study is an attempt to induce autotetraploids in an important medicinal plant, Trachyspermum ammi (L.) Sprague through colchicine. The seedlings of Trachyspermum ammi (L.) Sprague were treated with 3 different concentrations of colchicine (0.2, 0.4 and 0.5%, w/v) for 3 different durations. The autotetraploidy in plants have been confirmed on the basis of cytological, morphological and palynological observations. Total six autotetraploid plants (4n = 36) were induced. On the basis of the number of induced autotetraploids, 36 hours duration of 0.2% concentration of colchicine was found to be more effective. The morphological parameters such as increased flower size, reproductive organs, stomata size, pollen length and diameter, plant height, leaf length and width, number of umbel/plant, number of umbellate/umbel and stomatal frequency etc. were considered for the detection of autotetraploids. Various chromosomal configurations have been observed during meiotic analysis. Pollen fertility was decreased in autotetraploids than diploids.  相似文献

Leishmania donovani, a protozoan parasite, causes visceral disease in humans. To identify genes that control growth, we have isolated for the first time in the order Kinetoplastida a gene encoding for centrin from L. donovani. Centrin is a calcium-binding cytoskeletal protein essential for centrosome duplication or segregation. Protein sequence similarity and immunoreactivity confirmed that Leishmania centrin is a homolog of human centrin 2. Immunofluorescence analysis localized the protein in the basal body. Calcium binding analysis revealed that its C-terminal Ca(2+) binding domain binds 16-fold more calcium than the N-terminal domain. Electrophoretic mobility shift of centrin treated with EGTA and abrogation of the shift in its mutants lacking a Ca(2+) binding site suggest that Ca(2+) binding to these regions may have a role in the protein conformation. The levels of centrin mRNA and protein were high during the exponential growth of the parasite in culture and declined to a low level in the stationary phase. Expression of N-terminal-deleted centrin in the parasite significantly reduces its growth rate, and it was found that significantly more cells are arrested in the G(2)/M stage than in control cells. These studies indicate that centrin may have a functional role in Leishmania growth.  相似文献

Identification of a novel autoantigen UACA in patients with panuveitis   总被引:2,自引:0,他引:2  
To identify the target autoantigens in Vogt-Koyanagi-Harada disease, we made use of an immunoscreening of a bovine uveal cDNA expression library with serum samples obtained from patients with Vogt-Koyanagi-Harada disease. We identified a novel bovine antigen and homologous human autoantigen and designated it as UACA (uveal autoantigen with coiled coil domains and ankyrin repeats). mRNA of human UACA is expressed most abundantly in skeletal muscles and in various human tissues, including choroid, retina, and epidermal melanocytes. IgG autoantibodies were quantitated in an ELISA, using recombinant C-terminal 18.0% fragment of human UACA. The prevalence of IgG anti-UACA autoantibodies in patients with panuveitis (Vogt-Koyanagi-Harada disease, Beh?et's disease, sarcoidosis) was significantly higher than that in healthy controls (19.6-28.1% vs 0%, P < 0.05) indicating that autoimmunity directed against UACA is a common phenomenon in these diseases.  相似文献

Abstract. Seasonal patterns of stem diameter changes in evergreen and deciduous species of a tropical montane forest in the Central Himalayas (300–2250 m a.s.l.) were investigated in relation to leaf development. Ca. 75 % of the annual rainfall in this region occurs in a short period, from mid-June to mid-September and the remaining months are dry. It was assumed that changes in stem diameter are correlated with changes in water stress. Each evergreen species could be characterized by leaf longevity of about one year; each species showed pronounced summer leaf drop and simultaneous new leaf formation. Winter stem shrinkage was more pronounced in deciduous species than in evergreen ones. The deciduous species also showed a greater proportional loss of leaf mass (before abscission) than the evergreen species. Winter leaf fall in deciduous species was related to the pronounced stem shrinkage. The leaf fall enabled these species to control further water loss. Being more resistant to desiccation, the evergreen species retained their leaves throughout the winter but showed gradual loss of leaf mass, presumably in order to control water loss. In all species, leaf expansion was completed before the onset of the rainy season, when water stress was high. This strategy has definite advantages in a climate with a monsoon pattern of rainfall. Evergreen species, showing pronounced leaf drop in summer, have advantages over deciduous species; hence their preponderance in the region.  相似文献

A novel species of non-histone protein having a molecular weight of approximately 20,000 (20K), abundantly localized in the dorsolateral prostate, was found to be decreased in the content by castration and to be restored by replacement of androgen, in addition to non-histone proteins of molecular weights ≥ 34,000. The content of 20KDNA was more rapidly decreased by castration, but more slowly restored by replacement of androgen, with the dorsolateral prostate than the ventral prostate. Of other nuclear proteins, non-histone proteins of molecular weights ≥ 90,000 in the dorsolateral prostate were more susceptible to the decrease by castration, whereas those of all kinds of histones were hardly dependent on the androgen level.  相似文献
