The electrochemistry of a water-soluble fragment from the CuA domain of Thermus thermophilus cytochrome ba

has been investigated. At 25 °C, CuA exhibits a reversible reduction at a pyridine-4-aldehydesemicarbazone-modified gold electrode (0.1 M Tris, pH 8) with E° = 0.24 V vs NHE. Thermodynamic parameters for the [Cu(Cys)2Cu]+/0 electrode reaction were determined by variable-temperature electrochemistry (ΔS°rc = –5.4(12) eu, ΔS° = –21.0(12) eu, ΔH° = –11.9(4) kcal/mol;ΔG° = –5.6 (11) kcal/mol). The relatively small reaction entropy is consistent with a low reorganization energy for [Cu(Cys)2Cu]+/0 electron transfer. An irreversible oxidation of [Cu(Cys)2Cu]+ at 1 V vs NHE confirms that the CuII:CuII state of CuA is significantly destabilized relative to the CuII state of analogous blue-copper proteins.

Analysis of in situ gut contents of the ctenophore Mnemiopsisleidyi collected during summer months of 1998 at Woods Hole,MA, USA indicates that dietary changes accompanied developmentof this zooplankter. The transition from cydippid to lobatemorphologies involved a shift from a microplanktonic diet dominatedby protists to a metazoan-based diet. Dietary diversity increasedrapidly during the transition from cydippid to lobate stagebut remained essentially similar through most (>3.0 cm totallength) of the lobate stage. Localization and coordinate phosphorylation/dephosphorylation of endothelial nitric oxide synthase (eNOS) are critical determinants for the basal and stimulated production of nitric oxide. Several phosphorylation sites in eNOS have been identified as targets of the cAMP-dependent protein kinase A (PKA). Basal eNOS activity is also regulated by interaction with caveolin-1, the major coat protein of caveolae. In the present study we have examined in rat aorta endothelium the subcellular steady-state distribution of eNOS, the catalytic subunit of PKA (PKA-c), and caveolin-1. Basal eNOS expression was found in two distinct locations, the endothelial cell surface and the Golgi complex. Cell surface eNOS was equally distributed over caveolar and non-caveolar membranes but was 2.5-fold enriched on luminal lamellipodia located at endothelial cell contacts. PKA-c colocalized with eNOS in the lamellipodia, whereas caveolin-1 was absent from these membrane domains. PKA-c was also found associated with cell surface caveolae and with tubulovesicular membranes of Golgi complex and endosomes. The topological proximity of eNOS with the catalytic subunit of PKA in restricted intracellular locations may provide mechanisms for differential PKA-mediated eNOS regulation. 相似文献

Aquaculture – quo vadis? Aquaculture is more than just fish production; it is a globally active sector of the food production industry. The production methods interact in different degrees with the environment, resulting in possible negative impact. Through enhancement of the production systems as well as utilized resources, such influence can be controlled and minimized. This includes the amount of water consumption, nutrient efficiency and thus nutrient excretion from the production systems. An optimization of the culture conditions has direct benefits for animal welfare, diseases resistance, product quality and consequently supporting the productivity of the aquaculture enterprises. Understanding foraging decisions made by wildlife at different spatio‐temporal scales is important for wildlife management and conservation. We tested whether foraging decisions by Masai giraffe (Giraffa camelopardalis tippelskirchi Matschie) differed with scales; habitat, patch and tree in a heterogeneous savannah. We collected data from Arusha National Park, Tanzania, in March–May and August–October 2013. Visual observations were used to collect data on foraging behaviour. Measurements of tree height and stem height and scoring of accumulated browsing were made in 133 patches around trees where the giraffes had been seen browsing, and in a corresponding number of available patches. Giraffes selected Acacia shrub and Dodonea shrub habitats but not for water availability or predator avoidance. For patch use, giraffe selected high quantity of Acacia xanthophloea and Dodonea viscosa. Between plant species, A. xanthophloea was the most preferred and within plant species, tree quality was enhanced by tree height and high score of accumulated browsing. Epithelial-mesenchymal transition (EMT) is one mechanism of acquired resistance to inhibitors of the epidermal growth factor receptor-tyrosine kinases (EGFR-TKIs) in non-small cell lung cancer (NSCLC). The precise mechanisms of EMT-related acquired resistance to EGFR-TKIs in NSCLC remain unclear. We generated erlotinib-resistant HCC4006 cells (HCC4006ER) by chronic exposure of EGFR-mutant HCC4006 cells to increasing concentrations of erlotinib. HCC4006ER cells acquired an EMT phenotype and activation of the TGF-β/SMAD pathway, while lacking both T790M secondary EGFR mutation and MET gene amplification. We employed gene expression microarrays in HCC4006 and HCC4006ER cells to better understand the mechanism of acquired EGFR-TKI resistance with EMT. At the mRNA level, ZEB1 (TCF8), a known regulator of EMT, was >20-fold higher in HCC4006ER cells than in HCC4006 cells, and increased ZEB1 protein level was also detected. Furthermore, numerous ZEB1 responsive genes, such as CDH1 (E-cadherin), ST14, and vimentin, were coordinately regulated along with increased ZEB1 in HCC4006ER cells. We also identified ZEB1 overexpression and an EMT phenotype in several NSCLC cells and human NSCLC samples with acquired EGFR-TKI resistance. Short-interfering RNA against ZEB1 reversed the EMT phenotype and, importantly, restored erlotinib sensitivity in HCC4006ER cells. The level of micro-RNA-200c, which can negatively regulate ZEB1, was significantly reduced in HCC4006ER cells. Our results suggest that increased ZEB1 can drive EMT-related acquired resistance to EGFR-TKIs in NSCLC. Attempts should be made to explore targeting ZEB1 to resensitize TKI-resistant tumors. 相似文献
