Trichoderma reesei strains were constructed for production of elevated amounts of endoglucanase II (EGII) with or without cellobiohydrolase I (CBHI). The endoglucanase activity produced by the EGII transformants correlated with the copy number of the egl2 expression cassette. One copy of the egl2 expression cassette in which the egl2 was under the cbh1 promoter increased production of endoglucanase activity 2.3-fold, and two copies increased production about 3-fold above that of the parent strain. When the enzyme with elevated EGII content was used, an improved stonewashing effect on denim fabric was achieved. A T. reesei strain producing high amounts of EGI and -II activities without CBHI and -II was constructed by replacing the cbh2 locus with the coding region of the egl2 gene in the EGI-overproducing CBHI-negative strain. Production of endoglucanase activity by the EG-transformant strain was increased fourfold above that of the host strain. The filter paper-degrading activity of the endoglucanase-overproducing strain was lowered to below detection, presumably because of the lack of cellobiohydrolases.  相似文献

(1) We measured cooling rate for neonatal mink during a 10min coldroom (3.9 degrees C) exposure and subsequent warming rate during a 20min incubator (37.2 degrees C) exposure, the behaviour of the kits and the changes in their pelage between 1 and 46d of age, in an attempt to monitor the ontogeny of their thermoregulatory capacity. (2) Body weight of the 1d old kits averaged only 12.8+/-2.3g (n=4), but they gained weight rapidly reaching 226.1+/-28.3g (males, n=4) and 207.6+/-16.1g (females, n=4) at 30-31d of age, and 562.3+/-43.2g (males, n=3) and 435.7+/-35.5g (females, n=4) at 45-46d of age. (3) Body cooling rate (C(rate) ( degrees C/min); n=80) was affected by the age (between 1 and 31d), BW, initial rectal temperature (T(r0)), and sex of the kits, in addition to their body posture (P(cold), 1=extended, 2=curled-up) during coldroom exposure. C(rate) ( degrees C/min)=-0.34-0.02age-0.002BW+0.05T(r0)-0.06sex-0.20P(cold) (R(2)=0.75). (4) Body warming rate (W(rate) ( degrees C/min); n=80) was influenced by the age(2) and rectal temperature of the kit after the coldroom exposure (T(r10)). W(rate)( degrees C/min)=1.24+0.0002age(2)-0.04T(r10) (R(2)=0.76). (5) Kit fur fibre length increased from 5.45+/-0.63mm (males, n=2) and 6.20+/-0.20mm (females, n=3) at 22-23d of age to 9.43+/-1.44mm (males, n=4) and 8.70+/-1.89mm (females, n=4) at 30-31d of age, and to 12.93+/-0.47mm (males, n=3) and 11.38+/-0.41mm (females, n=4) at 45-46d of age, the growth averaging about 0.26mm per day. (6) Under normal circumstances newborn mink kits are hypothermic.Their thermoregulation develops only gradually and is dependent on increase in body mass, insulation and behavioural thermoregulation. Their strategy of survival is based on the ability to withstand hypothermia and on the nutrition and warmth provided by the dam.  相似文献

The loss and fragmentation of forest habitats have been considered to pose a worldwide threat to the viability of forest-dwelling animals, especially to species that occupy old forests. We investigated whether the annual survival of sedentary male Tengmalm’s owls Aegolius funereus was associated with the cover of old coniferous forests in Finland. Survival and recapture probabilities varied annually with density changes in populations of the main prey (Microtus voles). When this variation was controlled for, and relationships between survival and proportions of the three different forest age classes (old-growth, middle-aged, and young) were modeled separately, the old-growth model was the most parsimonious. Survival increased with the cover of old forest, although the extent of old forest within owl territories was relatively small (mean ∼12%, range 2–37%). This association, however, varied among years and appeared especially in years of increasing vole abundance. At such times, old forests may sustain high populations of bank voles Clethrionomys glareolus, shrews and small passerines. In addition, old forests may serve as refuges against large avian predator species, such as Ural owls Strix uralensis and goshawks Accipiter gentilis. Our results suggest that changes in habitat quality created by agriculture and forestry may have the potential to reduce adult survival, an essential component of fitness and population viability.  相似文献

The present study investigated the effects of 902.4 MHz Global System for Mobile Communications (GSM) mobile phone radiation on cerebral blood flow using positron emission tomography (PET) with the 15O‐water tracer. Fifteen young, healthy, right‐handed male subjects were exposed to phone radiation from three different locations (left ear, right ear, forehead) and to sham exposure to test for possible exposure effects on brain regions close to the exposure source. Whole‐brain [15O]H2O–PET images were acquired 12 times, 3 for each condition, in a counterbalanced order. Subjects were exposed for 5 min in each scan while performing a simple visual vigilance task. Temperature was also measured in the head region (forehead, eyes, cheeks, ear canals) during exposure. The exposure induced a slight temperature rise in the ear canals but did not affect brain hemodynamics and task performance. Microorganisms attached to particles have been shown to be different from free-living microbes and to display diverse metabolic activities. However, little is known about the ecotypes associated with particles and their substrate preference in anoxic marine waters. Here, we investigate the microbial community colonizing particles in the anoxic and sulfide-rich waters of the Black Sea. We incubated beads coated with different substrates in situ at 1000 and 2000 m depth. After 6 h, the particle-attached microbes were dominated by Gamma- and Alpha-proteobacteria, and groups related to the phyla Latescibacteria, Bacteroidetes, Planctomycetes and Firmicutes, with substantial variation across the bead types, indicating that the attaching communities were selected by the substrate. Further laboratory incubations for 7 days suggested the presence of a community of highly specialized taxa. After incubation for 35 days, the microbial composition across all beads and depths was similar and primarily composed of putative sulfur cycling microbes. In addition to the major shared microbial groups, subdominant taxa on chitin and protein-coated beads were detected pointing to specialized microbial degraders. These results highlight the role of particles as sites for attachment and biofilm formation, while the composition of organic matter defined a secondary part of the microbial community.  相似文献

Copper(II) and zinc(II) complexes of a polyamino-polyol ligand 1,3,5-trideoxy-1,3,5-tris(methylamino)-cis-inositol (tmci) have been investigated as potential candidates for the selective elimination of the 5'-cap structure of mRNA. A cap-model compound ApppA has been utilised as substrate for studying the effect of the different metal ion complex catalysts on the hydrolysis of the triphosphate bridge. Kinetic experiments have been performed by the variation of pH, metal-to-ligand ratio and total concentrations of the metal ion and ligand. The zinc(II) complexes of tmci have been proved to possess a remarkable activity for the hydrolysis of ApppA. The observed rate enhancement compared to the uncatalysed reaction was found to be 12,000-fold, in the presence of 4.5mM zinc(II) and 1.5mM tmci at pH approximately 7.5. In contrast with the copper(II) containing systems, an extra product has also been formed during the cleavage process, beside the expected AMP and ADP. According to the ESI-MS characterisation of the samples, the additional product is a covalent phosphoester adduct of AMP and the ligand. The formation of this species is initiated by a nucleophilic attack of a zinc(II)-bound alcohol or alkoxo group on one of the alpha phosphate groups of ApppA, which leads to the formation of a phosphodiester bond. In an alternative pathway, the substrate is cleaved into AMP and ADP. According to the pH-potentiometric studies, performed with the tmci-zinc(II) system, di- and trinuclear complexes are responsible for the accelerated ApppA hydrolysis. The copper(II)-tmci 2:1 system showed only a modest kinetic activity. The rate acceleration significantly increased when threefold excess of copper(II) was applied. Although, the detailed investigations above pH approximately 6.6 have been prevented by precipitate formation during the addition of the substrate into the reaction solution, the activity of the copper(II)-tmci 3:1 system does not exceed that of the zinc(II) complexes. Due to the specific mechanism leading to the covalent extra product, the zinc(II) complexes of tmci provide a comparable rate enhancement for ApppA hydrolysis to the widely studied lanthanide or copper(II) species, in spite of the fact that they are stronger Lewis acids.  相似文献

The potential association of CD2AP with the adherens junction protein E-cadherin, co-localization with the actin cytoskeleton, and involvement in cell migration was investigated in cultured rat gastric mucosal cells. In stationary cells, CD2AP was localized perinuclearly while E-cadherin was expressed along cell-cell contacts and F-actin formed a branched network and adhesion belts. In migrating cells, CD2AP appeared as thread-like accumulations in the leading edges, colocalizing with F-actin and occasionally with E-cadherin. Intracellular injection of anti-CD2AP significantly retarded the migration speed of the cells suggesting a crucial role for CD2AP in mucosal cell migration, possibly as a scaffolding protein between cell membrane proteins and actin cytoskeleton. Co-immunoprecipitation assays revealed that CD2AP and E-cadherin are in a complex in HGF stimulated cells. It is concluded that CD2AP interacts with E-cadherin and co-localizes with F-actin in the leading edge of migrating cells, and significantly contributes to cell migration in restituting gastric epithelium.  相似文献
