A genetic analysis of age of onset of alcoholism was performed on the Collaborative Study on the Genetics of Alcoholism data released for Genetic Analysis Workshop 14. Our study illustrates an application of the log-normal age of onset model in our software Genetic Epidemiology Models (GEMs). The phenotype ALDX1 of alcoholism was studied. The analysis strategy was to first find the markers of the Affymetrix SNP dataset with significant association with age of onset, and then to perform linkage analysis on them. ALDX1 revealed strong evidence of linkage for marker tsc0041591 on chromosome 2 and suggestive linkage for marker tsc0894042 on chromosome 3. The largest separation in mean ages of onset of ALDX1 was 19.76 and 24.41 between male smokers who are carriers of the risk allele of tsc0041591 and the non-carriers, respectively. Hence, male smokers who are carriers of marker tsc0041591 on chromosome 2 have an average onset of ALDX1 almost 5 years earlier than non-carriers.  相似文献

DNA adjuvants and whole bacterial cell vaccines against bacterial kidney disease (BKD) were tested in juvenile chinook salmon. Whole cell vaccines of either a nonpathogenic Arthrobacter spp. or an attenuated Renibacterium salmoninarum strain provided limited prophylactic protection against acute intraperitoneal challenge with virulent R. salmoninarum, and the addition of either synthetic oligodeoxynucleotides or purified R. salmoninarum genomic DNA as adjuvants did not increase protection. However, a combination of both whole cell vaccines significantly increased survival among fish naturally infected with R. salmoninarum, and the surviving fish treated with the combination vaccine exhibited reduced levels of bacterial antigens in the kidney. This is the first demonstration of a potential therapeutic effect of a whole cell vaccine against BKD.  相似文献

Background  

Evolutionary analyses of the largest subunit of RNA polymerase II (RPB1) have yielded important and at times provocative results. One particularly troublesome outcome is the consistent inference of independent origins of red algae and green plants, at odds with the more widely accepted view of a monophyletic Plantae comprising all eukaryotes with primary plastids. If the hypothesis of a broader kingdom Plantae is correct, then RPB1 trees likely reflect a persistent phylogenetic artifact. To gain a better understanding of RNAP II evolution, and the presumed artifact relating to green plants and red algae, we isolated and analyzed RPB 1 from representatives of Glaucocystophyta, the third eukaryotic group with primary plastids.  相似文献

Genetic transformation of most insect systems requires dominant-acting markers that do not depend on reverting a mutant phenotype in a host strain, andfor this purpose GFP has proven to be useful in several insect orders. However, detection of multiple transgenes and reporters for gene expression requires the development of new visible markers that can be unambiguously detected when co-expressed with GFP The DsRed fluorescentprotein has spectral characteristics that are most distinct from GFP and GFP variants, and we have explored the use of DsRed as a selectable marker for piggyBac transformation in Drosophila melanogaster and its use as a reporter when co-expressed with GFP. Transformants marked with polyubiquitin-regulated DsRed1 were detected throughout development at a relatively high frequency, and they exhibited brighter fluorescence than transformants marked with EGFP. The use of a Texas Red filter set eliminated detection of EGFP fluorescence and autofluorescence, and DsRed expressedfrom a reporter construct could be unambiguously detected when co-expressed with EGFP DsRed should prove to be a highly efficient marker system for the selection of transformant insects and as a reporter in gene expression studies.  相似文献

An intranuclear microsporidium is described from hemoblastic cells of the chinook salmon, Oncorhynchus tshawytscha. The infection is associated with an acute anemia in the fish. Up to 47% of the hemoblast nuclei were infected in anemic fish. The organisms, found only in spleen and kidney tissues, were 1-2 microns in diameter and consisted of vegetative and early sporulation forms. This microsporidium differs from known species which parasitize fish in its tissue location; however, the absence of mature spores and other life cycle stages precludes determination of its precise taxonomic identity.  相似文献

Somatic embryos of Ipomoea batatas Lam. (sweet potato cv. White Star) were produced in an airlift bioreactor. This work describes the optimization of the embryogenic system on semisolid medium, followed by transfer of the system to liquid cultures and ultimately to the airlift bioreactor. The physiological age of embryogenic callus influenced the number and overall morphology of the embryo population in both semisolid and liquid medium. Maximum mature embryo production (35 embryos 10 mg-1 inoculum) was obtained from six-week-old callus at 30°C. Somatic embryogenesis occurred in liquid cultures containing 20 mM NH4NO3 and 30 mM KCl. Globular embryos formed and continued development in suspension producing viable, mature cotyledonary embryos by day 14. Embryo formation and development was limited in the bioreactor. Although shear stress was responsible for some embryogenic damage, the effect of purging the system with fresh air needed to be investigated. To isolate aeration effects from shear stress effects, atmospheric determinations were performed on shaker flask cultures. Initially the gas composition within the Erlenmeyer headspace was that of room air. Ethylene increased to a maximum of 6.4 ppm (day 16), maximum CO2, 21.2%, was also evident on day 16, and oxygen was depleted to a minimum of 8.1% by day 14. Purging the cultures with fresh air reduced the number of embryos formed; however, they were significantly longer than those formed in closed flasks. The gas response model of Ipomoea batatas will enable atmosphere replenishment in the bioreactor mimicking that of the shaker flask environment. Once the damaging effects of shear stress have been overcome, the regulation of bioreactor gasses should allow somatic embryo formation in the bioreactor comparable to that in shaker flasks.  相似文献

The anthropological study of women should rest upon a sound understanding of women's reproductive heritage. Current research in reproductive physiology can contribute to this understanding, particularly with reference to the lactation period. In preindustrial societies as a rule, lactation is prolonged and intensive, while menstruation is correspondingly uncommon. This is related primarily to cultural factors that interact with the suckling frequency, but other factors may be involved. Industrialization and the propagation of western attitudes have been associated with a worldwide decline in lactation; an example from Taiwan is presented. The concept of the menstrual cycle as a liminal state is introduced, and anthropologists are urged to rethink their understanding of physiologic and symbolic womanhood .
