Bone morphogenetic proteins (BMPs) are cytokines from the TGF-β superfamily, with important roles during embryonic development and in the induction of bone and cartilage tissue differentiation in the adult body. In this contribution, we report the expression of recombinant human BMP-4, BMP-9, BMP-10, BMP-11 (or growth differentiation factor-11, GDF-11) and BMP-14 (GDF-5), using Escherichia coli pET-25b vector. BMPs were overexpressed, purified by affinity his-tag chromatography and shown to induce the expression of early markers of bone differentiation (e.g. smad-1, smad-5, runx2/cbfa1, dlx5, osterix, osteopontin, bone sialoprotein and alkaline phosphatase) in C2C12 cells and in human adipose stem cells. The described approach is a promising method for producing large amounts of different recombinant BMPs that show potential for novel biomedical applications.  相似文献

The purpose of this study was to describe, interpret and compare the EMG activation patterns of ankle muscles – tibialis anterior (TA), peroneus longus (PL) and gastrocnemius lateralis (GL) – in volleyball players with and without ankle functional instability (FI) during landing after the blocking movement. Twenty-one players with FI (IG) and 19 controls (CG) were studied. The cycle of movement analyzed was the time period between 200 ms before and 200 ms after the time of impact determined by ground reaction forces. The variables were analyzed for two different phases: pre-landing (200 ms before impact) and post-landing (200 ms after impact). The RMS values and the timing of onset activity were calculated for the three studied muscles, in both periods and for both groups. The co-activation index for TA and PL, TA and GL were also calculated. Individuals with FI presented a lower RMS value pre-landing for PL (CG = 43.0 ± 22.0; IG = 26.2 ± 8.4, p < 0.05) and higher RMS value post-landing (CG = 47.5 ± 13.3; IG = 55.8 ± 21.6, p < 0.10). Besides that, in control group PL and GL activated first and simultaneously, and TA presented a later activation, while in subjects with FI all the three muscles activated simultaneously. There were no significant differences between groups for co-activation index. Thus, the rate of contraction between agonist and antagonist muscles is similar for subjects with and without FI but the activation individually was different. Volleyball players with functional instability of the ankle showed altered patterns of the muscles that play an important role in the stabilization of the foot–ankle complex during the performance of the blocking movement, to the detriment of the ligament complex, and this fact could explain the usual complaints in these subjects.  相似文献

Denture stomatitis and its risk indicators in south Brazilian older adults Objective: The purpose of this study was to evaluate if socio‐demographic, behaviour and health variables are risk indicators for denture‐related stomatitis in community‐dwelling older adults from Carlos Barbosa, Brazil. Materials and methods: In this cross‐sectional study, 872 subjects aged 60 years or more were evaluated. Data collection included dental examinations and questionnaires to assess socio‐demographic, behaviour, general and oral health data. All questionnaires and examinations were carried out during 2004. Associations between the independent variables and the outcomes ‘prevalence of denture stomatitis’ or ‘prevalence of granular stomatitis’ were adjusted by means of multivariate logistic regressions. Results: Participants with denture stomatitis were significantly more likely to be females, to display increased amounts of plaque, and significantly less likely to visit the dentist for treatment. Participants with granular stomatitis were significantly more likely to be females and participants with granular stomatitis were significantly more likely to be females, aged 70 years or more and current smokers. In addition, participants with granular stomatitis displayed increased amounts of plaque. Conclusion: The data presented here confirms the central role of denture biofilm accumulation in denture stomatitis. It also indicates that smoking may increase susceptibility to infection in its most severe form.  相似文献

Parkinson's disease (PD) is a progressive movement disorder resulting from the death of dopaminergic neurons in the substantia nigra. Neurotoxin-based models of PD using 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) recapitulate the neurological features of the disease, triggering a cascade of deleterious events through the activation of the c-Jun N-terminal kinase (JNK). The molecular mechanisms underlying the regulation of JNK activity under cellular stress conditions involve the activation of several upstream kinases along with the fine-tuning of different endogenous JNK repressors. Glutathione S-transferase pi (GSTP), a phase II detoxifying enzyme, has been shown to inhibit JNK-activated signaling by protein-protein interactions, preventing c-Jun phosphorylation and the subsequent trigger of the cell death cascade. Here, we use C57BL/6 wild-type and GSTP knockout mice treated with MPTP to evaluate the regulation of JNK signaling by GSTP in both the substantia nigra and the striatum. The results presented herein show that GSTP knockout mice are more susceptible to the neurotoxic effects of MPTP than their wild-type counterparts. Indeed, the administration of MPTP induces a progressive demise of nigral dopaminergic neurons together with the degeneration of striatal fibers at an earlier time-point in the GSTP knockout mice when compared to the wild-type mice. Also, MPTP treatment leads to increased p-JNK levels and JNK catalytic activity in both wild-type and GSTP knockout mice midbrain and striatum. Moreover, our results demonstrate that in vivo GSTP acts as an endogenous regulator of the MPTP-induced cellular stress response by controlling JNK activity through protein-protein interactions.  相似文献

The biodiversity crisis has had particularly harsh impacts on marine environments. However, there is still considerable uncertainty about how many species have been seriously impacted and the effectiveness of protection measures (e.g., marine protected areas or MPAs) due to high levels of cryptic species in many taxa. Here, we employ an integrative taxonomy approach to mullet species in the genus Mugil. In addition to its high economic value, this genus is notable for having diversified ~29 million years ago without marked morphological and ecological divergence. We obtained 129 specimens of Mugil from the Coral Coast MPA, the largest of its kind in the Tropical Southwestern Atlantic marine province. Although morphometric and meristic traits revealed six taxonomically recognized species, only five mitochondrial lineages were observed. All individuals morphologically identified as M. incilis belonged to the mitochondrial lineage of Mugil curema, which is consistent with misidentification of morphologically similar species and an overestimation of species diversity. Remarkably, Mugil species in our sample that diverged up to ~23 million years ago are also the most morphologically similar (M. curema and M. rubrioculus), suggesting extreme morphological conservatism, possibly driven by similarities in habitat use and life‐history traits. This study demonstrates the potential utility of integrative taxonomy (including DNA barcoding) for contributing to the conservation and sustainable use of natural resources.  相似文献
