The three-dimensional conformation of Met-enkephalin, corresponding to the lowest minimum of the empirical potential energy function ECEPP/2 (empirical conformational energy program for peptides), has been determined using a new algorithm, viz. the Electrostatically Driven Monte Carlo Method. This methodology assumes that a polypeptide or protein molecule is driven toward the native structure by the combined action of electrostatic interactions and stochastic conformational changes associated with thermal movements. These features are included in the algorithm that produces a Monte Carlo search in the conformational hyperspace of the polypeptide, using electrostatic predictions and a random sampling technique to locate low-energy conformations. In addition, we have incorporated an alternative mechanism that allows the structure to escape from some conformational regions representing metastable local energy minima and even from regions of the conformational space with great stability. In 33 test calculations on Met-enkephalin, starting from arbitrary or completely random conformations, the structure corresponding to the global energy minimum was found inall the cases analyzed, with a relatively small search of the conformational space. The applicability of the concept of negentropy to the study of evolution is tested by tracing the phylogeny of the information content of the communication systems used for recruitment in the Formicidae. Data from the literature is reviewed and ordered around theoretical evolutionary routes based on increasing negentropy of the chemical communication systems used. One pathway presupposes the development of chemical orientation prior to chemical attraction, i.e. signalling the presence of food; whereas the others begin with the chemical attraction of nestmates without orientation with chemical cues to the food. The results suggest that the Myrmicinae, Ponerinae, Dolichoderinae, Pseudomyrmycinae and possibly Dorylinae evolved chemical recruitment via chemical attraction, whereas the Formicinae evolved chemical recruitment via chemical orientation. These alternative patterns permit the establishment of phyletic trends based on glandular evolution for chemical recruitment. The results are compared with phylogenetic studies based on morphological and chemical characters, and it emerges that the recruitment behaviour proves to be a particularly conservative characteristic, in that it is evident in the same form in a wider range of species. The conclusion is reached that the negentropy content of living systems is a powerful tool in phylogenetic studies, even of small taxonomic groups. 相似文献

Ethanol dependence was induced in rats by maintaining them for 3 weeks on a liquid diet containing ethanol. When ethanol was abruptly replaced with sucrose in the diet, animals showed withdrawal symptoms. Eight hours later, the accumulation in brain and heart of 3H-norepinephrine synthesized from 3H-tyrosine, and of 3H-norepinephrine metabolites was greater than in animals not undergoing withdrawal. An injection of ethanol (3 g/kg) 1 or 5 hours after the initiation of withdrawal resulted in less accumulation of newly synthesized 3H-norepinephrine and of 3H-norepinephrine metabolites in both brain and heart. If the rate of ethanol withdrawal was slow, i.e., the ethanol in the diet was replaced gradually with sucrose over a 3-day period, less accumulation of 3H-norepinephrine and 3H-norepinephrine metabolites occured in heart and brain than as a result of abrupt withdrawal. Also, no behavioral symptoms of withdrawal were observed. These results indicate that (a) gross withdrawal symptoms and the accompanying activation of noradrenergic neurons can be blocked during withdrawal by an acute dose of ethanol, and (b) ethanol withdrawal can be modified by altering the rate of withdrawal, a finding that may prove useful in clinical situations. We conclude that the withdrawal symptoms and the activation of noradrenergic neurons during withdrawal are caused by the sudden lack of ethanol in the system. 相似文献

Fertile chicken eggs were incubated in an altitude chamber in a near 100% O2 atmosphere at 225 torr. Both N2 and CO2 were kept under 0.5%. Temperature was a standard 37.5° but a high relative humidity of 90% was required to prevent dehydration. In ten trials involving 382 eggs, hatchability averaged 21% of controls and weight of chicks was 11% less than controls, but embryo mortality was distributed similarly. Males and females of the common bed bug, Cimex lectularius L. (Heteroptera: Cimicidae), have been shown to produce and respond to an aggregation pheromone. We tested whether juvenile C. lectularius also produce and respond to aggregation pheromone, and whether the pheromone is perceived by contact chemoreception. In dual‐choice laboratory experiments, juveniles, but not males or females, preferred juvenile‐exposed paper discs to control discs. Unlike juveniles, males and females preferred male‐exposed paper discs to control discs. Neither juveniles, males, nor females preferred female‐exposed discs to control discs. When test stimuli were inaccessible, C. lectularius failed to show any preference. Male‐ and juvenile‐specific contact pheromones may have contrasting functions of marking shelters as safe refugia for development and growth (juveniles) or mate encounter (adults), but result in the same phenomenon, the aggregation of conspecifics. 相似文献

Fibronectin (FN) matrix fibrils have long been thought to be formed by disulfide-bonded FN multimers, although there is no direct evidence that they are covalently linked with each other. To understand the biochemical properties of these fibrils, we extracted a crude FN matrix from FN-YPet transfected 3T3 cell culture using 0.2% deoxycholate and DNase. The insoluble extracted matrix preserved fibrillar structures and a major portion of the extracted proteins migrated as FN monomers on an SDS gel under reducing conditions. Under non-reducing conditions, some FN molecules appeared to be trapped at the top of the stacking gel. We tested this by mixing fluorescently labeled FN dimers with the extracted matrix just before loading on an SDS gel, and found that most of them were trapped with the extracted proteins at the top of the stacking gel. These results suggested that some components of the extracted matrix plugged the stacking gel and FN dimers were trapped with them. Rotary shadowing electron microscopy showed that the extracted matrix had some fibers that resembled fibrillin microfibrils. Peptide mass fingerprinting confirmed the presence of fibrillin in the extracted matrix. Fibrillin is known to form disulfide-bonded multimers and it is likely to be one of the components that plug the stacking gel and trap FN molecules in this system. The phenomenon by which FN molecules appear to migrate as multimers on SDS gels is thus an artifact rising from the presence of other large components in the extract. We conclude that FN matrix fibrils are made of FN dimers that are further cross-linked by non-covalent protein–protein bonds. 相似文献
