The presence of significant amount of heavy metals in rivers and canals due to mixing of untreated industrial effluents is a common phenomenon, especially in developing countries. The agricultural crops are influenced by the presence of various pollutants in the sewage, being applied for irrigation purpose. The effluents containing copper affect the growth and development of crop species, thereby, ought to be mitigated by foliar spray of osmoprotectants, e.g. proline. A pot culture experiment was conducted at the University of Agriculture, Faisalabad-Pakistan during the crop season 2015–2016. The treatments consisted of (a) three wheat varieties (Punjab-96, MH-97, FSD-83), (b) two levels of copper (0, 400?µM) applied through rooting medium, and (c) two levels of proline (0, 80?mM) applied through foliar application. The treatments were arranged in a completely randomized design with four replications. The results showed that application of 400?µM copper caused a reduction in biomass accumulation, chlorophyll (‘a’ and ‘b’) contents, and eventually yield (100-grain weight). There were also significant decreases in gas exchange parameters (stomatal conductance, internal CO2 concentration), photosynthetic rate, water-use-efficiency, and transpiration rate in response to copper stress. Metal toxicity caused the maximum reduction in productivity of PSII, electron transport rate, and photochemical quenching, while higher values of non-photochemical quenching were recorded in the wheat varieties. The activities of antioxidant enzymes (superoxide dismutase, catalase, peroxidase), as well as quantities of proline, protein and calcium contents were accelerated in response to copper stress. The uptake of calcium, magnesium, and potassium constituents by plants was reduced, while assimilation of calcium was increased in plants under copper stress. However, the occurrence of negative effects on these parameters due to copper stress was mitigated by foliar spray of proline at the rate of 80?mM solution. The exogenous application of proline at the rate of 80?mM resulted in the reduction of generation of reactive oxygen species and enhanced accumulation of proline and protein contents in wheat varieties under copper stress environment.  相似文献

Although radiotherapy and most cancer drugs target the proliferation of cancer cells, it is metastasis, the complex process by which cancer cells spread from the primary tumor to other tissues and organs of the body where they form new tumors, that leads to over 90% of all cancer deaths. Thus, there is an urgent need for anti-metastasis strategies alongside chemotherapy and radiotherapy. An important step in the metastatic cascade is migration. It is the first step in metastasis via local invasion. Here we address the question whether ionizing radiation and/or chemotherapy might inadvertently promote metastasis and/or invasiveness by enhancing cell migration. We used a standard laboratory irradiator, Faxitron CellRad, to irradiate both non-cancer (HCN2 neurons) and cancer cells (T98G glioblastoma) with 2 Gy, 10 Gy and 20 Gy of X-rays. Paclitaxel (5 μM) was used for chemotherapy. We then measured the attachment and migration of the cells using an electric cell substrate impedance sensing device. Both the irradiated HCN2 cells and T98G cells showed significantly (p < 0.01) enhanced migration compared to non-irradiated cells, within the first 20–40 h following irradiation with 20 Gy. Our results suggest that cell migration should be a therapeutic target in anti-metastasis/anti-invasion strategies for improved radiotherapy and chemotherapy outcomes.  相似文献

The human minisatellite arrays, 33.6 and 33.15, consist of tandem reiterations of a 37-nucleotide (nt) and a 16-nt repeat unit sequence, respectively, both of which contain a majority of purine bases on one strand. Knot-like tertiary structures, which mapped to the cloned arrays, were observed by electron microscopy (EM) in homoduplex molecules produced by denaturation and reannealing in vitro. They result from a primary hybridization between misaligned repeat units of the array, forming a slipped-strand structure with staggered single-stranded DNA loops, followed by a secondary hybridization between repeat units in the two loops. Depending on the relative alignment of the loops when they hybridize, a particular form of intrahelical pseudoknot is produced. Theta-shaped, figure-of-eight, and bow-shaped structures were the most common conformational isomers observed in homoduplexes flattened into two dimensions during EM preparation. At the site of a bow-shaped structure, a conformation-dependent bend of approximately 60 degrees between the flanking DNA segments is induced; the other conformations generally do not deflect the line of the main DNA axis. Paired loops, similar to the bow-shaped structure, were apically situated in some supercoiled plasmids containing the 33.6 array. Both plasmids formed intermolecular associations, consisting of two (or more) homoduplex molecules held together at or immediately adjacent to a nexus which mapped to the minisatellite sequences. These associations might arise either by interhelical hybridization between arrays or by knot-like structures interfering with branch migration of chi-form Holliday junctions.  相似文献
