A series of sulfamide-based analogs related to L-796568 were prepared and evaluated for their biological activity at the human beta(3)-adrenergic receptor (AR). This modification allows for a significant reduction in molecular weight, while maintaining single-digit nanomolar potencies at the beta(3)-AR and high selectivities versus the beta(2)- or beta(3)-AR.  相似文献

The crystal structures of cyanide and azide-bound forms of the truncated hemoglobin from Synechocystis are presented at 1.8 angstroms resolution. A comparison with the structure of the endogenously liganded protein reveals a conformational shift unprecedented in hemoglobins, and provides the first picture of a hexacoordinate hemoglobin in both the bis-histidyl and the exogenously coordinated states. The structural changes between the different conformations are confined to two regions of the protein; the B helix, and the E helix, including the EF loop. A molecular "hinge" controlling movement of the E helix is observed in the EF loop, which is composed of three principal structural elements: Arg64, the heme-d-propionate, and a three-residue extension of the F helix. Additional features of the structural transition between the two protein conformations are discussed as they relate to the complex ligand-binding behavior observed in hexacoordinate hemoglobins, and the potential physiological function of this class of proteins.  相似文献

A methodology is presented to facilitate a declaration that an area is 'pest-free' following an eradication campaign against an insect pest. This involves probability models to assess null trapping results and also growth models to help verify, following a waiting period, that pests were not present when control was stopped. Two probability models are developed to calculate the probability of negative trapping results if in fact insects were present. If this probability is sufficiently low, then the hypothesis that insects are present is rejected. The models depend on knowledge of the efficiency and the area of attractiveness of the traps. To verify the results of the probability model, a waiting period is required to see if a rebound occurs. If an incipient but non-detectable population remains after control measures are discontinued, then a rebound should occur. Using a growth model, the rate of increase of an insect population is examined starting from one gravid female or one male and a female. An example is given for tsetse in which both means and confidence limits are calculated for a period of 24 reproductive periods after control is terminated. If no rebound is detected, then a declaration of eradication can be made.  相似文献

Trypanosomiasis control increasingly involves financial input from livestock owners and their active participation. If control is carried out on smaller scales than in the past, methods such as aerial and ground spraying and sterile insect techniques will have reduced application. There will be increased reliance on trypanocidal drugs, and bait methods of tsetse control--where flies are attracted to point sources and killed. If drug resistance develops, cheap and simple bait methods offer the only means of disease control that might be applied, and paid for, by stockowners themselves. The methods have been effective in some circumstances, but not in others, and it is important to understand the reasons for the successes and the failures. Analysis is presented of the results of two Tanzanian tsetse control campaigns involving the use of insecticide-treated cattle. Between 1991 and 1996, following the introduction of widespread dipping in the Kagera Region, trypanosomiasis declined from >19000 cases to <2400 and deaths from >4000 to 29. On four ranches in the region, tsetse have been almost eliminated and trypanosomiasis prophylaxis is no longer used. Similarly aggressive use of pyrethroids on Mkwaja Ranch in Tanga Region has not had such dramatic effects. Tsetse and trypanosomiasis are still common, despite high levels of prophylaxis and the deployment of approximately 200 odour-baited targets. The difference in the results is attributed to a combination of the much smaller area covered by treated animals at Mkwaja, a greater susceptibility to re-invasion and a more suitable habitat for the flies. A better understanding of the dynamics of the use of insecticide-treated cattle is needed before we can predict confidently the outcome of particular control operations.  相似文献

The effects that naturally occurring gases (oxygen, nitrogen, carbon monoxide) may cause in dormant giant foxtail (Setaria faberii) seed germination under favorable temperature and moisture conditions were investigated. The germination responses to gas mixtures supported the hypothesis that S. faberii germination behavior is regulated by the amount of oxygen taken into hydrated seed over time. Setaria faberii seed germination was markedly affected by O(2) concentration (in N(2)) above and below that of air (20% O(2)): the largest increase in germination (from 37 to 60%) occurred between 20-25% O(2); between 0-10% O(2), germination increased from 0-30%; and surprisingly germination at 10 and 20% O(2) was similar. These observations reveal an asymmetrical response to incremental changes in O(2) above and below that typically found in agricultural soils. Carbon monoxide had opposite effects on S. faberii germination in air depending on concentration, stimulation, and inhibition: germination increased from 37 to 56% with the addition of 1% CO, but decreased from 37 to 14% with 75% added CO. An explanation may be that there are two separate effects of CO, each occurring in different physiological systems of dormant seeds at the same time. At high concentrations (75%) in air CO inhibited seed germination, probably by inhibiting mitochondrial respiration. But low CO concentrations (0.1 or 1%) in air stimulated seed germination. It was not apparent which physiological system(s) CO and O(2) affected. It seems unlikely that CO-stimulated germination arises from effects on the respiratory apparatus, but may be a consequence of CO interactions with an as yet unknown physiological factor in the seed. We provide a model of Setaria spp. dormancy consistent with its seed morphology, the gas-germination data, and the hypothesized second physiological factor that may be involved in CO stimulated germination.  相似文献

Plant nonsymbiotic hemoglobins are hexacoordinate heme proteins found in all plants. Although expression is linked with hypoxic environmental conditions (Taylor, E. R., Nie, X. Z., Alexander, W. M., and Hill, R. D. (1994) Plant Mol. Biol. 24, 853-862), no discrete physiological function has yet been attributed to this family of proteins. The crystal structure of a nonsymbiotic hemoglobin from rice has recently been determined. The crystalline protein is homodimeric and hexacoordinate with two histidine side chains coordinating the heme iron atom. Despite the fact that the amino acids responsible for the subunit interface are relatively conserved among the nonsymbiotic hemoglobins, previous work suggests that this group of proteins might display variability in quaternary structure (Duff, S. M. G., Wittenberg, J. B., and Hill, R. D. (1997) J. Biol. Chem. 272, 16746-16752; Arredondo-Peter, R., Hargrove, M. S., Sarath, G., Moran, J. F., Lohrman, J., Olson, J. S., and Klucas, R. V. (1997) Plant Physiol. 115, 1259-1266). Analytical ultracentrifugation and size exclusion high pressure liquid chromatography were used to investigate the quaternary structure of rice nonsymbiotic hemoglobin at various states of ligation and oxidation. Additionally, site-directed mutagenesis was used to test the role of several interface amino acids in dimer formation and ligand binding. Results were analyzed in light of possible physiological functions and indicate that the plant nonsymbiotic hemoglobins are not oxygen transport proteins but more closely resemble known oxygen sensors.  相似文献

A detection procedure was developed in which a newly devised lysine-iron medium was used as a one-step selective and enrichment medium for detection of salmonellae by the fluorescent-antibody technique. Incubation was conducted in two steps: initially at 30 C for 5 hr to resuscitate sublethally stressed cells, followed by incubation at 39 C for 17 hr. Twenty-seven strains of salmonellae from groups A-I were utilized in the development of this procedure which was sensitive enough to detect one Salmonella bacterium in 100 g of nonfat dry milk.  相似文献

A multiple stressor risk assessment was conducted at Yuma Proving Ground, Arizona, as a demonstration of the Military Ecological Risk Assessment Framework (MERAF). The focus was a testing program at Cibola Range that involved an Apache Longbow helicopter firing Hellfire missiles at moving targets, that is, M60-A1 tanks. This article describes the ecological risk assessment, using the MERAF, for the tracked vehicle movement component of the testing program. The principal stressor associated with tracked vehicle movement was soil disturbance, and a resulting, secondary stressor was hydrological change. Water loss to desert wash vegetation was hypothesized to result from increased infiltration and/or evaporation associated with vehicle disturbances to surrounding desert pavement, potentially affecting mule deer as well as vegetation. The simulated exposure of wash vegetation to water loss was quantified using estimates of disturbed land area from a digital orthogonal quarter quadrangle aerial photo and field observations, a 30-m digital elevation model, the flow accumulation feature of ESRI ArcInfo GIS, and a two-step runoff process dependent on soil characteristics and the extent of disturbance. In all simulated scenarios, the absolute amount of water lost increased with distance from the disturbance downslope in the washes; however, the percentage of water lost was greatest in land areas immediately downslope of a disturbance. Potential effects on growth and survival of desert wash trees were quantified by comparing water availability from the hydrologic model to water volume thresholds required for wash trees to survive and persist, derived from a local study. For both the incremental risk of the test program and for the combination of test and pretest disturbances, this demonstration of MERAF found no significant risk to either wash vegetation growth and survival or mule deer abundance and reproduction.  相似文献

Background

The behaviour of insect vectors has an important bearing on the epidemiology of the diseases they transmit, and on the opportunities for vector control. Two sorts of electrocuting device have been particularly useful for studying the behaviour of tsetse flies (Glossina spp), the vectors of the trypanosomes that cause sleeping sickness in humans and nagana in livestock. Such devices consist of grids on netting (E-net) to catch tsetse in flight, or on cloth (E-cloth) to catch alighting flies. Catches are most meaningful when the devices catch as many as possible of the flies potentially available to them, and when the proportion caught is known. There have been conflicting indications for the catching efficiency, depending on whether the assessments were made by the naked eye or assisted by video recordings.

Methodology/Principal Findings

Using grids of 0.5m2 in Zimbabwe, we developed catch methods of studying the efficiency of E-nets and E-cloth for tsetse, using improved transformers to supply the grids with electrical pulses of ~40kV. At energies per pulse of 35–215mJ, the efficiency was enhanced by reducing the pulse interval from 3200 to 1ms. Efficiency was low at 35mJ per pulse, but there seemed no benefit of increasing the energy beyond 70mJ. Catches at E-nets declined when the fine netting normally used became either coarser or much finer, and increased when the grid frame was moved from 2.5cm to 27.5cm from the grid. Data for muscoids and tabanids were roughly comparable to those for tsetse.

Conclusion/Significance

The catch method of studying efficiency is useful for supplementing and extending video methods. Specifications are suggested for E-nets and E-cloth that are ~95% efficient and suitable for estimating the absolute numbers of available flies. Grids that are less efficient, but more economical, are recommended for studies of relative numbers available to various baits.  相似文献

A novel saccharide host containing four boronic acid recognition units on a single DNA duplex terminus was constructed. This construct allowed boronic acid sugar recognition in the context of double-stranded DNA to be established while highlighting the benefits of multivalency. Following the solid-phase synthesis of a bis-boronic acid tag, two end-functionalized oligonucleotides with complementary sequences were functionalized through amide ligation. By annealing the boronic acid-DNA conjugates, a tetra-boronic acid DNA duplex was assembled. The saccharide binding ability of this tetra-boronic acid host was revealed through cellulose paper chromatography in the absence and presence of various saccharides. While no appreciable saccharide binding was seen in the case of a bis-boronic DNA conjugate, the increased migration of the tetra-boronic acid host relative to the control sequences in the presence of selected monosaccharides underscored the importance of multivalent effects. We thus identified a requirement for multiple recognition sites in these conjugate systems and expect the results to facilitate future efforts toward applying synthetic recognition systems to the realm of macromolecules.  相似文献
