The fungitoxic effects of different plant extracts on Fusarium udum, which causes wilt disease of Cajanus cajan in vitro and in vivo, were examined. The complete arrest of the radial growth of the pathogen occurred at a 10% concentration of leaf extract from Adenocallyma alliaceum. A leaf extract of Citrus medica, a root extract of Asparagus adscendens, rhizome extracts of Curcuma longa and Zingiber officinale, and a bulb extract of Allium sativum inhibited up to 100% growth at higher concentrations. A. alliaceum controlled the disease up to 100% by amending its 4% powder in unsterilized soil and 2% in sterilized soil. The population of F. udum was found to be markedly reduced following treatments with plant powders.  相似文献

d-Hydantoinase from the lentil, Lens esculenta, seed is quite unstable, and has been immobilized on Diethyl amino ethyl (DEAE) cellulose by an adsorption and cross-linking method. The immboilized d-hydantoinase exhibited 80% enzyme activity and contained 86% protein. The immobilization of the enzyme preparation does not change its optimum pH, temperature or affinity constant, but increases its shelf-life, thermostability and stability in various organic solvents. This immobilized d-hydantoinase can be used effectively for the production of d-amino acids from the corresponding hydantoins and may therefore be of use in the chemical and pharmaceutical industries. There is a need for new analytical approaches to better characterize the nature of the concentration-dependent, reversible self-association (RSA) of monoclonal antibodies (mAbs) directly, and with high resolution, when these proteins are formulated as highly concentrated solutions. In the work reported here, hydrogen exchange mass spectrometry (HX-MS) was used to define the concentration-dependent RSA interface, and to characterize the effects of association on the backbone dynamics of an IgG1 mAb (mAb-C). Dynamic light scattering, chemical cross-linking, and solution viscosity measurements were used to determine conditions that caused the RSA of mAb-C. A novel HX-MS experimental approach was then applied to directly monitor differences in local flexibility of mAb-C due to RSA at different protein concentrations in deuterated buffers. First, a stable formulation containing lyoprotectants that permitted freeze-drying of mAb-C at both 5 and 60 mg/mL was identified. Upon reconstitution with RSA-promoting deuterated solutions, the low vs. high protein concentration samples displayed different levels of solution viscosity (i.e., approx. 1 to 75 mPa.s). The reconstituted mAb-C samples were then analyzed by HX-MS. Two specific sequences covering complementarity-determining regions CDR2H and CDR2L (in the variable heavy and light chains, respectively) showed significant protection against deuterium uptake (i.e., decreased hydrogen exchange). These results define the major protein-protein interfaces associated with the concentration-dependent RSA of mAb-C. Surprisingly, certain peptide segments in the VH domain, the constant domain (CH2), and the hinge region (CH1-CH2 interface) concomitantly showed significant increases in local flexibility at high vs. low protein concentrations. These results indicate the presence of longer-range, distant dynamic coupling effects within mAb-C occurring upon RSA.  相似文献

This investigation aims to evaluate the antitumor and antioxidant potential of Chrysaora quinquecirrha (sea nettle) nematocyst venom on Ehrlich ascites carcinoma (EAC) tumor model. Tumor was induced in mice by intraperitoneal injection of EAC cells. The antitumor effect of sea nettle nematocyst venom (SNV) peptide was evaluated by assessing in vitro cytotoxicity, survival time, hematological, and antioxidant parameters. Intraperitoneal injection of SNV peptide increased the survival time of the EAC-bearing mice. The SNV peptide brought back the altered levels of the hematological and antioxidant parameters in a dose dependent manner in EAC-bearing mice. The results were comparable to that of the result obtained from the animals treated with the standard drug 5-fluorouracil (20 mg/kg bw). Thus, present study revealed that SNV peptide possessed significant antitumor and antioxidant activity.  相似文献

Population dynamics of five different species of grasshopper were analyzed for the first time in Darjeeling (Lebong and Happy Valley) of the eastern Himalayan region of India. The study is based on the relationship between monthly samples collected using sweep nets for three years (March, 2005 to February, 2008) in relation to meteorological parameters (monthly average rainfall, monthly mean maximum and minimum temperatures). The population for each of the five species of grasshopper is plotted against the Aridity Ratio (A.R.). For all species, the population increases at lower A.R. values and then decreases exponentially at higher A.R. values. The exponentially decreasing part of the population is modeled using a simple formula. The monthly population of A. crenulata nymphs and adults in Lebong has also been modeled by iterative equations using A.R. and results compared satisfactorily with the sample data. These works show the possibility of forecasting grasshopper populations using a simple model and thereby easing the regulation process.  相似文献
