Multiple introductions can play a prominent role in explaining the success of biological invasions. One often cited mechanism is that multiple introductions of invasive species prevent genetic bottlenecks by parallel introductions of several distinct genotypes that, in turn, provide heritable variation necessary for local adaptation. Here, we show that the invasion of Aegilops triuncialis into California, USA, involved multiple introductions that may have facilitated invasion into serpentine habitats. Using microsatellite markers, we compared the polymorphism and genetic structure of populations of Ae. triuncialis invading serpentine soils in California to that of accessions from its native range. In a glasshouse study, we also compared phenotypic variation in phenological and fitness traits between invasive and native populations grown on loam soil and under serpentine edaphic conditions. Molecular analysis of invasive populations revealed that Californian populations cluster into three independent introductions (i.e. invasive lineages). Our glasshouse common garden experiment found that all Californian populations exhibited higher fitness under serpentine conditions. However, the three invasive lineages appear to represent independent pathways of adaptation to serpentine soil. Our results suggest that the rapid invasion of serpentine habitats in California may have been facilitated by the existence of colonizing Eurasian genotypes pre‐adapted to serpentine soils.  相似文献

Natural products play an important role in the development of anticancer drugs. To date, predominantly metabolites from plants and bacteria served as lead structures for anticancer agents. Fungal metabolites and derivatives thereof are much less investigated for their potential in cancer therapy. There are, however, some promising candidates derived from fungi in clinical phases I and II studies. This review gives an overview on the role of natural products in cancer therapy and summarises some of the latest results of our group in this area.  相似文献

Buffering compounds like TRIS are frequently used in chemical, biochemical and biomedical applications to control pH in solution. One of the prerequisites of a buffer compound, in addition to sufficient buffering capacity and pH stability over time, is its non-reactivity with other constituents of the solution. This is especially important in the field of analytical chemistry where analytes are to be determined quantitatively. Investigating the enzymatic hydrolysis of G-type nerve agents sarin, soman and cyclosarin in buffered solution we have identified stable buffer adducts of TRIS, TES and other buffer compounds with the nerve agents. We identified the molecular structure of these adducts as phosphonic diesters using 1D 1H-31P HSQC NMR and LC-ESI-MS/MS techniques. Reaction rates with TRIS and TES are fast enough to compete with spontaneous hydrolysis in aqueous solution and to yield substantial amounts (up to 20–40%) of buffer adduct over the course of several hours. A reaction mechanism is proposed in which the amino function of the buffer serves as an intramolecular proton acceptor rendering the buffer hydroxyl groups nucleophilic enough for attack on the phosphorus atom of the agents. Results show that similar buffer adducts are formed with a range of hydroxyl and amino function containing buffers including TES, BES, TRIS, BIS-TRIS, BIS-TRIS propane, Tricine, Bicine, HEPES and triethanol amine. It is recommended to use alternative buffers like MOPS, MES and CHES when working with G-type nerve agents especially at higher concentrations and over prolonged times.  相似文献

Three years after a river restoration scheme in the Syr Valley (Luxembourg) we investigated habitat development and habitat use of Galloway cattle deployed in a low-intensity grazing system on a permanent floodplain pasture. Habitats were delimited with a mobile GPS/GIS mapping system and their spatial development was assessed over three consecutive years. During these three years, the patches of the six habitats decreased to 40% of mean initial size, and a rapid net area expansion of wetland habitats (large sedge swamps: +100%, marsh and tall forb grasslands: +43%) was observed. The behavioural patterns and grazing preferences of the cattle were observed directly during the vegetation period in June, August and November. These observations were complemented by a transect analysis of cattle impact indicators in June and November. The cattle grazed the different habitats very selectively, as they preferred the mesophilic, and ruderal grasslands 1.6, and 5.6 times more than expected respectively. During the growing season, the grazing niche breadth declined (3.92 in June to 2.68 in November), and less preferred forage habitats like large sedge swamps were grazed primarily in the autumn. We used bite and step rates to investigate grazing intensity by habitat type. During summer, grazing intensity correlated with forage quality in the different habitat patches, whereas in autumn it was obviously influenced by the effort required to access the desired forage plants in a given habitat. The impact indicators revealed a matter transfer from riparian areas to the valley edge. Here, we give a first insight into habitat development and habitat use of Galloway cattle in a recently restored floodplain area and derive recommendations for the adaptive management of future projects.  相似文献

Ischemic stroke is the second leading cause of death worldwide. Only one moderately effective therapy exists, albeit with contraindications that exclude 90% of the patients. This medical need contrasts with a high failure rate of more than 1,000 pre-clinical drug candidates for stroke therapies. Thus, there is a need for translatable mechanisms of neuroprotection and more rigid thresholds of relevance in pre-clinical stroke models. One such candidate mechanism is oxidative stress. However, antioxidant approaches have failed in clinical trials, and the significant sources of oxidative stress in stroke are unknown. We here identify NADPH oxidase type 4 (NOX4) as a major source of oxidative stress and an effective therapeutic target in acute stroke. Upon ischemia, NOX4 was induced in human and mouse brain. Mice deficient in NOX4 (Nox4 −/−) of either sex, but not those deficient for NOX1 or NOX2, were largely protected from oxidative stress, blood-brain-barrier leakage, and neuronal apoptosis, after both transient and permanent cerebral ischemia. This effect was independent of age, as elderly mice were equally protected. Restoration of oxidative stress reversed the stroke-protective phenotype in Nox4 −/− mice. Application of the only validated low-molecular-weight pharmacological NADPH oxidase inhibitor, VAS2870, several hours after ischemia was as protective as deleting NOX4. The extent of neuroprotection was exceptional, resulting in significantly improved long-term neurological functions and reduced mortality. NOX4 therefore represents a major source of oxidative stress and novel class of drug target for stroke therapy.  相似文献

Transport of glucose 1-phosphate (G1P) and highly purified triose phosphate into storage tissue amyloplasts was studied. Isolated amyloplasts from potato ( Solanum tuberosum L., dihaploid stock, HH 258) were transport-functional and metabolically active in starch synthesis. Fourty percent of the amyloplasts were intact and there was only a small degree (0–1.6%) of contamination by other cellular compartments. G1P showed a clear uptake pattern paralleled by starch synthesis. Uptake of triose phosphates was virtually nil. Uptake of GIP was pH dependent with a sharp maximum at pH 5.7 and showed Michaelis-Menten kinetics with an apparent Km of 0.5 m M . Temperature influenced the rate of uptake, the highest rate being at 25°C. Fructose l-phosphate, ADP-glucose, glucose, and inorganic phosphate inhibited the uptake of G1P. Uptake was also inhibited by DIDS (1–25 μ M ) and by Phloretin (45–750 μW). It is therefore concluded that the transport of GIP across the inner amyloplast membrane is mediated by a hexose phosphate translocator selective for phosphate and glucose moieties of the molecule. Considering the low pH maximum for G1P uptake it is possible that the uptake of G1P, and eventually starch synthesis, is regulated by an acidification of the intermembrane space by proton pumps of the inner amyloplast membrane.  相似文献
