Movement of water overlying the sediments has not been taken into consideration in most of the experiments conducted to estimate the dissolved material flux from the sediment. Even in recent experiments that incorporated the stirring motion, interpretation of the data is difficult, because the mixing rate used may be different from actual mixing rate in the field. We propose a method to estimate the in situ mixing rate that should be used to set the flow rate in a flow-through core incubation system. The flow rate is calculated from the vertical mixing rate of the water that is deduced from the heat diffusivity. Release rates of NO3+NO2–N obtained from our flow-through incubation system were higher by 1–3 orders of magnitude than those from the conventional diffusion calculation method that estimates the flux from the gradient of nutrient concentration across the sediment-water interface. Increase in NO3+NO2–N flux is considered to be due to intensification of the nitrification process as a result of an increase in dissolved oxygen (DO) supply with the motion of water. DO supply is also considered to be an important factor controlling macrofaunal abundance and consequently their excretory contributions to the fluxes of dissolved organic nitrogen as well as NH4–N. From this point of view, we strongly recommend the application of heat and mass balance theory to estimate nitrogen flux using a flow-through experimental system.  相似文献

Lactose is a principal carbohydrate in nearly all species of mammalian milk. In order to examine the acceptor substrate specificity of lactose synthase in vivo, D-xylose as an acceptor substrate was injected into the jugular vein of a Holstein cow during lactation, then a milk sample obtained by milking. beta1, beta1-galactopyranosyl xylopyranoside, a nonreducing disaccharide, was separated from the bovine milk sample after elimination of reducing sugars, identified by fast-atom bombardment (FAB)-MS and 1H-NMR analysis. A mixture of beta1,beta1- and beta1, 4-galactopyranosyl xylopyranoside fractions was also obtained by thin layer chromatography from the milk sample and elucidated by electrospray ionization (ESI)-MS and 1H NMR analysis. Comparison of the integrated intensity of the products shows that the beta1,beta1 and beta1,4 isomers are present in a ratio of 1.0 : 1.4, suggesting that D-xylose, transported from capillary blood across the plasma membrane of the mammary gland, was recognized by lactose synthase in its normal and reverse orientation owing to high symmetry of its structure. While the beta1,4-isomer is known as a fragment of the linkage region between the protein and the polysaccharide chain of proteoglycans, the beta1,beta1-isomer has not been identified in vivo. Here, we demonstrate that galactosylation of D-xylose transported from the capillary blood can occur by lactose synthase catalysis in the mammary gland while the usual galactosylation of D-glucose proceeds. In addition, these results suggest that the possibility of endogenous occurrence of the beta,beta-trehalose type disaccharide in the mammary gland of lactating mammals may not be ruled out.  相似文献

This study evaluated the effect of inter-tree variation in the bud phenology of Picea glehnii on susceptibility to the shoot-boring sawfly. Pleroneura piceae , and found that individual susceptibility fluctuates from year to year. The mechanism for the fluctuation between 1994 and 1997 is discussed.
Inter-tree difference in the time of bud swelling is probably genetically based, since most of the trees that began to swell early in 1995 also swelled early in 1997, and those that began to swell late also did so in both years. Damage severity of each tree was evaluated by damage ratio: proportion of the number of damaged current shoots on the previous year's leader shoot. The rank of the bud swelling phenology of a tree was positively correlated to the rank of the damage ratio. This means that genetically based differences in phenology could explain why some trees are subjected to higher levels of herbivory than others.
There was year-to-year variation in the damage severity for each tree. Nevertheless, no significant differences were found in the rank of the damage ratio between years. However the standard deviation of the damage ratios of each tree was highest for trees of intermediate rank. The skew of the frequency distribution of damage ratio was negatively correlated to the cumulated daily mean temperature in spring, which means that the spruce is more susceptible to the sawfly in warm springs than in cool springs.
The mean growth rate of the lightly damaged trees increased constantly, while that of the heavily damaged trees seemed to reach a limit and then became lower than that of the lightly damaged trees.  相似文献

Upstream-migration of juvenileRhinogobius sp. OR was studied along the Ado River, a tributary of Lake Biwa. Juvenile gobies, mainly 18–21 mm in standard length, migrated from the lake in a belt of about 3 m wide in lower reaches and about 10–20 cm wide in middler reaches. Their upstream-migration was restricted to places shallower than 20 cm with bottom current velocities less than 20 cm/sec. Individual migration speed varied between 4.2–8.9 m/min at places with bottom current velocity of 8.8–20.0 cm/sec, slowing down with increasing current velocity. Most active migration occurred between 14:00 and 16:00, number of upstream-migrating juveniles decreasing with distance from the river mouth. Riffles in the mid-and upper reaches, having few slow current areas, probably present barriers to upstream-migration of the juvenile gobies, although both adults and subadults of the species are distributed in more upper reaches.  相似文献

Orexin is a neuropeptide that plays a highly important role in mechanisms that regulate sleep/wake states. Lack of the orexin gene or orexin-producing neurons (orexin neurons) results in narcolepsy in several mammalian species, suggesting that orexin is an important factor for the maintenance of wakefulness. Constitutive, ectopic expression of orexin in transgenic mice resulted in severe fragmentation of non–rapid eye movement sleep, along with abnormal muscle tone regulation during REM sleep, suggesting that activity of orexin neurons should be appropriately decreased during sleep to maintain consolidated sleep states. This review will discuss the mechanisms by which the orexin system is regulated during sleep.

Standard hydrogen bonds are of great importance for protein structure and function. Ionic hydrogen bonds often are significantly stronger than standard hydrogen bonds and exhibit unique properties, but their role in proteins is not well understood. We report that hydrogen/deuterium exchange causes a redshift in the visible absorbance spectrum of photoactive yellow protein (PYP). We expand the range of interpretable isotope effects by assigning this spectral isotope effect (SIE) to a functionally important hydrogen bond at the active site of PYP. The inverted sign and extent of this SIE is explained by the ionic nature and strength of this hydrogen bond. These results show the relevance of ionic hydrogen bonding for protein active sites, and reveal that the inverted SIE is a novel, to our knowledge, tool to probe ionic hydrogen bonds. Our results support a classification of hydrogen bonds that distinguishes the properties of ionic hydrogen bonds from those of both standard and low barrier hydrogen bonds, and show how this classification helps resolve a recent debate regarding active site hydrogen bonding in PYP.  相似文献

An improved methodology for providing a more stable and targetable drug carrier has been developed. This method involves the synthesis of a newly designed immunoliposome by coating the outermost surface of large oligolamellar vesicles of egg phosphatidylcholine with the polysaccharide pullulan, modified to carry both cholesterol, as the hydrophobic anchor, and the monoclonal antibody fragment (anti-sialosyl Lewis X, IgMs) as the sensory device. Compared with the binding of pullulan-coated liposomes, that of this immunoliposome to specific cells in vitro was significantly increased by factors of 447 to PC-9 and 295 to KATO-III, but only by a factor of 148 to the less specific cell, 3LL. This strong and specific binding of the immunoliposome to the cell surface of PC-9 was also confirmed by a fluorescence-microscopic investigation using the immunoliposome, which bore the hydrophobic fluorescent probe, terbium trisacetylacetonate, in the liposomal membrane.  相似文献
