The potential contribution of fucosyltransferases to the overexpression of sialyl-Lex antigen was investigated in the colon carcinoma cell line HT-29 and in human colon carcinoma tissue. In HT-29 cells as well as in normal or malignant colonic tissues Fuc-TIII, Fuc-TIV, Fuc-TVI but not Fuc-TV nor Fuc-TVII were detectable after RT-PCR. Sodium butyrate treatment of HT-29 cells increased (to about 200%) and DMSO treatment decreased (to about 20%) the expression of sialyl-Lex. This modulation of sialyl-Lex was concomitant with the analogous increase/decrease of mRNA of Fuc-TIII but not Fuc-TIV. Fuc-TVI was not detectable by Northern blotting in HT-29 cells. In six human colon carcinomas which exhibited strong overexpression of sialyl-Lex, the expression of Fuc-TIII-mRNA was the same or lower than in the corresponding normal colonic tissue. Thus Fuc-TIII expression may be affecting the expression of the sialyl-Lex moiety in HT-29 cells but not in human colon carcinoma tissue.  相似文献

Molecular evolution and radiation of dung beetles in Madagascar   总被引:1,自引:0,他引:1  
Madagascar is the world's fourth largest island and has a wide range of climates and ecosystems. Environmental diversity combined with long history of isolation (160 Myr) has generated a high level of endemism at different taxonomic levels, making Madagascar one of the hotspots of global biodiversity. Dung beetles, represented by the two tribes of Canthonini and Helictopleurini, exemplify a large insect taxon. Helictopleurini are completely endemic to Madagascar while Canthonini are endemic at generic level. Using data from mitochondrial and nuclear genes, phylogenetic relationships were investigated in a sample of 44 species. The phylogeny for Canthonini consists of several distinct clades, possibly reflecting multiple colonization of Madagascar. The phylogeny does not support the current taxonomy for all genera. The phylogeny for Helictopleurini lacks statistical support at supra‐specific level, and genetic divergence among the Helictopleurini species is comparable with that among species within genera in Canthonini. These results suggest that Helictopleurini has undergone rapid speciation and most likely more recently than Canthonini, consistent with the estimated radiation time based on mtDNA mutation rates in insects and with knowledge about the systematics and geographic distribution of dung beetles worldwide. Spatially structured populations in patchy habitats show much variation in migration rate, from patchy populations in which individuals move repeatedly among habitat patches to classic metapopulations with infrequent migration among discrete populations. To establish a common framework for population dynamics in patchy habitats, we describe an individual-based model (IBM) involving a diffusion approximation of correlated random walk of individual movements. As an example, we apply the model to the Glanville fritillary butterfly (Melitaea cinxia) inhabiting a highly fragmented landscape. We derive stochastic patch occupancy model (SPOM) approximations for the IBMs assuming pure demographic stochasticity, uncorrelated environmental stochasticity, or completely correlated environmental stochasticity in local dynamics. Using realistic parameter values for the Glanville fritillary, we show that the SPOMs mimic the behavior of the IBMs well. The SPOMs derived from IBMs have parameters that relate directly to the life history and behavior of individuals, which is an advantage for model interpretation and parameter estimation. The modeling approach that we describe here provides a unified framework for patchy populations with much movements among habitat patches and classic metapopulations with infrequent movements.  相似文献

The requirements for cholera toxin-catalyzed ADP ribosylation of the purified regulatory component of adenylate cyclase are described. In addition to the toxin, this reaction is dependent on or is facilitated by NAD, GTP, phospholipid, and a factor found associated with plasma membranes from several sources. Factor activity is heat-labile and protease-sensitive but is unaffected by treatment with N-ethylmaleimide. Gel filtration indicates that the factor behaves as a monodisperse species with a Stokes radius of 3.2 nm. The factor thus appears to be a protein that is distinct from any of the known components of adenylate cyclase. Factor activity was also detected in the cytoplasm of S49 cells. The cytoplasmic factor was smaller (Stokes radius = 2.0 nm) than the membrane-derived factor, and it was inactivated in the presence of sodium cholate. The initial rate of activation of the regulatory component of adenylate cyclase by toxin was found to be linearly related to the amount of factor present in the reaction. This has allowed the quantitation and partial purification (33-fold from detergent extracts) of the factor from turkey erythrocyte membranes.  相似文献

Group A streptococcus (GAS) induces its own entry into eukaryotic cells in vitro and in vivo. Fibronectin (Fn) bound to protein F1, a GAS surface protein, acts as a bridge connecting the bacterium to host cell integrins. This triggers clustering of integrins, which acquire a polar pattern of distribution similar to that of protein F1 on the GAS surface. A unique and transient adhesion complex is formed at the site of GAS entry, which does not contain alpha-actinin. Vinculin is recruited to the site of GAS entry but is not required for uptake. The invading GAS recruits focal adhesion kinase (FAK), which is required for uptake and is tyrosine phosphorylated. The Src kinases, Src, Yes and Fyn, enhance the efficiency of GAS uptake but are not absolutely required for GAS entry. In addition, Rac and Cdc42, but not Rho, are required for the entry process. We suggest a model in which integrin engagement by Fn-occupied protein F1 triggers two independent signalling pathways. One is initiated by FAK recruitment and tyrosine phosphorylation, whereas the other is initiated by the recruitment and activation of Rac. The two pathways subsequently converge to trigger actin rearrangement leading to bacterial uptake.  相似文献
