Insensitive munitions explosives are new formulations that are less prone to unintended detonation compared to traditional explosives. While these formulations have safety benefits, the individual constituents, such as 2,4-dinitroanisole (DNAN), have an unknown ecosystem fate with potentially toxic impacts to flora and fauna exposed to DNAN and/or its metabolites. Fungi may be useful in remediation and have been shown to degrade traditional nitroaromatic explosives, such as 2,4,6-trinitrotoluene and 2,4-dinitrotoluene, that are structurally similar to DNAN. In this study, a fungal Penicillium sp., isolated from willow trees and designated strain KH1, was shown to degrade DNAN in solution within 14 days. Stable-isotope labeled DNAN and an untargeted metabolomics approach were used to discover 13 novel transformation products. Penicillium sp. KH1 produced DNAN metabolites resulting from ortho- and para-nitroreduction, demethylation, acetylation, hydroxylation, malonylation, and sulfation. Incubations with intermediate metabolites such as 2-amino-4-nitroanisole and 4-amino-2-nitroanisole as the primary substrates confirmed putative metabolite isomerism and pathways. No ring-cleavage products were observed, consistent with other reports that mineralization of DNAN is an uncommon metabolic outcome. The production of metabolites with unknown persistence and toxicity suggests further study will be needed to implement remediation with Penicillium sp. KH1. The molecular mechanism of ethylenediaminetetraacetic acid (EDTA)-induced membrane destabilization has been studied using a combination of four biophysical techniques on artificial lipid membranes. Data from Langmuir film balance and epifluorescence microscopy revealed the fluidization and expansion effect of EDTA on phase behavior of monolayers of either 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) or mixtures of DPPC and metal-chelating lipids, such as N^a,N^a-Bis[carboxymethyl]-N^ε [(dioctadecylamino)succinyl]-L-lysine or 1,2-dioleoyl-sn-glycero-3-[N-(5-amino- 1 -carboxypentyl iminodiacetic acid) succinyl]. A plausible explanation could be drawn from the electrostatic interaction between negatively charged groups of EDTA and the positively charged choline head group of DPPC. Intercalation of EDTA into the lipid membrane induced membrane curvature as elucidated by atomic force microscopy. Growth in size and shape of the membrane protrusion was found to be time-dependent upon exposure to EDTA. Further loss of material from the lipid membrane surface was monitored in real time using a quartz crystal microbalance. This indicates membrane restabilization by exclusion of the protrusions from the surface. Loss of lipid components facilitates membrane instability, leading to membrane permeabilization and lysis. 相似文献

New discoveries have fuelled the ongoing discussion of panspermia, i.e. the transport of life from one planet to another within the solar system (interplanetary panspermia) or even between different planetary systems (interstellar panspermia). The main factor for the probability of interstellar panspermia is the average density of stellar systems containing habitable planets. The combination of recent results for the formation rate of Earth-like planets with our estimations of extrasolar habitable zones allows us to determine the number of habitable planets in the Milky Way over cosmological time scales. We find that there was a maximum number of habitable planets around the time of Earth's origin. If at all, interstellar panspermia was most probable at that time and may have kick-started life on our planet. 相似文献

Epifluorescence microscopy as well as atomic force microscopy was successfully applied to explore the orientation and lateral organization of a group of chimeric green fluorescent proteins (GFPs) on lipid membrane. Incorporation of the chimeric GFP carrying Cd-binding region (His6CdBP4GFP) to the fluid phase of DPPC monolayer resulted in a strong fluorescence intensity at the air-water interface. Meanwhile, non-specific adsorption of the GFP having hexahistidine (His6GFP) led to the perturbation of the protein structure in which very low fluorescence was observed. Specific binding of both of the chimeric GFPs to immobilized zinc ions underneath the metal-chelating lipid membrane was revealed. This specific binding could be reversibly controlled by addition of metal ions or metal chelator. Binding of the chimeric GFPs to the metal-chelating lipid membrane was proven to be the end-on orientation while the side-on adsorption was contrarily noted in the absence of metal ions. Increase of lateral mobility owing to the fluidization effect on the chelating lipid membrane subsequently facilitated crystal formation. All these findings have opened up a potential approach for a specific orientation of immobilization of protein at the membrane interface. Accumulation of hyperphosphorylated Tau protein as paired helical filaments in pyramidal neurons is a major hallmark of Alzheimer disease. Besides hyperphosphorylation, other modifications of the Tau protein, such as cross-linking, are likely to contribute to the characteristic features of paired helical filaments, including their insolubility and resistance against proteolytic degradation. In this study, we have investigated whether the four reactive carbonyl compounds acrolein, malondialdehyde, glyoxal, and methylglyoxal accelerate the formation of Tau oligomers, thioflavin T-positive aggregates, and fibrils using wild-type and seven pseudophosphorylated mutant Tau proteins. Acrolein and methylglyoxal were the most reactive compounds followed by glyoxal and malondialdehyde in terms of formation of Tau dimers and higher molecular weight oligomers. Furthermore, acrolein and methylglyoxal induced the formation of thioflavin T-fluorescent aggregates in a triple pseudophosphorylation-mimicking mutant to a slightly higher degree than wild-type Tau. Analysis of the Tau aggregates by electron microscopy study showed that formation of fibrils using wild-type Tau and several Tau mutants could be observed with acrolein and methylglyoxal but not with glyoxal and malondialdehyde. Our results suggest that reactive carbonyl compounds, particularly methylglyoxal and acrolein, could accelerate tangle formation in vivo and that this process could be slightly accelerated, at least in the case of methylglyoxal and acrolein, by hyperphosphorylation. Interference with the formation or the reaction of these reactive carbonyl compounds could be a promising way of inhibiting tangle formation and neuronal dysfunction in Alzheimer disease and other tauopathies. 相似文献

The availability of dense molecular markers has made possible the use of genomic selection (GS) for plant breeding. However, the evaluation of models for GS in real plant populations is very limited. This article evaluates the performance of parametric and semiparametric models for GS using wheat (Triticum aestivum L.) and maize (Zea mays) data in which different traits were measured in several environmental conditions. The findings, based on extensive cross-validations, indicate that models including marker information had higher predictive ability than pedigree-based models. In the wheat data set, and relative to a pedigree model, gains in predictive ability due to inclusion of markers ranged from 7.7 to 35.7%. Correlation between observed and predictive values in the maize data set achieved values up to 0.79. Estimates of marker effects were different across environmental conditions, indicating that genotype × environment interaction is an important component of genetic variability. These results indicate that GS in plant breeding can be an effective strategy for selecting among lines whose phenotypes have yet to be observed.PEDIGREE-BASED prediction of genetic values based on the additive infinitesimal model (Fisher 1918) has played a central role in genetic improvement of complex traits in plants and animals. Animal breeders have used this model for predicting breeding values either in a mixed model (best linear unbiased prediction, BLUP) (Henderson 1984) or in a Bayesian framework (Gianola and Fernando 1986). More recently, plant breeders have incorporated pedigree information into linear mixed models for predicting breeding values (Crossa et al. 2006, 2007; Oakey et al. 2006; Burgueño et al. 2007; Piepho et al. 2007).The availability of thousands of genome-wide molecular markers has made possible the use of genomic selection (GS) for prediction of genetic values (Meuwissen et al. 2001) in plants (e.g., Bernardo and Yu 2007; Piepho 2009; Jannink et al. 2010) and animals (Gonzalez-Recio et al. 2008; VanRaden et al. 2008; Hayes et al. 2009; de los Campos et al. 2009a). Implementing GS poses several statistical and computational challenges, such as how models can cope with the curse of dimensionality, colinearity between markers, or the complexity of quantitative traits. Parametric (e.g., Meuwissen et al. 2001) and semiparametric (e.g., Gianola et al. 2006; Gianola and van Kaam 2008) methods address these problems differently.In standard genetic models, phenotypic outcomes, , are viewed as the sum of a genetic value, , and a model residual, ; that is, . In parametric models for GS, is described as a regression on marker covariates (j = 1, … , p molecular markers) of the form , such that(or , in matrix notation), where is the regression of on the jth marker covariate .Estimation of via multiple regression by ordinary least squares (OLS) is not feasible when p > n. A commonly used alternative is to estimate marker effects jointly using penalized methods such as ridge regression (Hoerl and Kennard 1970) or the Least Absolute Shrinkage and Selection Operator (LASSO) (Tibshirani 1996) or their Bayesian counterpart. This approach yields greater accuracy of estimated genetic values and can be coupled with geostatistical techniques commonly used in plant breeding to model multienvironments trials (Piepho 2009).In ridge regression (or its Bayesian counterpart) the extent of shrinkage is homogeneous across markers, which may not be appropriate if some markers are located in regions that are not associated with genetic variance, while markers in other regions may be linked to QTL (Goddard and Hayes 2007). To overcome this limitation, many authors have proposed methods that use marker-specific shrinkage. In a Bayesian setting, this can be implemented using priors of marker effects that are mixtures of scaled-normal densities. Examples of this are methods Bayes A and Bayes B of Meuwissen et al. (2001) and the Bayesian LASSO of Park and Casella (2008).An alternative to parametric regressions is to use semiparametric methods such as reproducing kernel Hilbert spaces (RKHS) regression (Gianola and van Kaam 2008). The Bayesian RKHS regression regards genetic values as random variables coming from a Gaussian process centered at zero and with a (co)variance structure that is proportional to a kernel matrix K (de los Campos et al. 2009b); that is, , where , are vectors of marker genotypes for the ith and jth individuals, respectively, and is a positive definite function evaluated in marker genotypes. In a finite-dimensional setting this amounts to modeling the vector of genetic values, , as multivariate normal; that is, where is a variance parameter. The anti-angiogenic properties of thalidomide have led to the use of the agent as a remedy for multiple myeloma. Nevertheless, the anti-angiogenic moiety of thalidomide remains unidentified. In this study we examined the anti-angiogenic effects of thalidomide in an in vitro model using a three-dimensional collagen gel culture. Angiogenesis was significantly inhibited when the culture was treated with thalidomide plus cytochrome P-450 (CYP2B4), and the migrating cells and tubules were positive for active-caspase-3 in an accompanying immunohistochemical investigation. Transmission electron microscopic observation also confirmed that active-caspase-3-positive cells demonstrated apoptotic characteristics. This study is the first to morphologically demonstrate the effect of thalidomide in directly inducing the apoptosis of new tubules and migrating cells on a three-dimensional collagen gel culture of aorta. Taken together with earlier findings, our new results indicate that the thalidomide-induced inhibition of angiogenesis involves apoptosis in addition to the suppression of TNF- and inhibition of cell migration from aorta explants, i.e., the factors important for capillarogenesis. 相似文献
