Epifluorescence microscopy as well as atomic force microscopy was successfully applied to explore the orientation and lateral organization of a group of chimeric green fluorescent proteins (GFPs) on lipid membrane. Incorporation of the chimeric GFP carrying Cd-binding region (His6CdBP4GFP) to the fluid phase of DPPC monolayer resulted in a strong fluorescence intensity at the air-water interface. Meanwhile, non-specific adsorption of the GFP having hexahistidine (His6GFP) led to the perturbation of the protein structure in which very low fluorescence was observed. Specific binding of both of the chimeric GFPs to immobilized zinc ions underneath the metal-chelating lipid membrane was revealed. This specific binding could be reversibly controlled by addition of metal ions or metal chelator. Binding of the chimeric GFPs to the metal-chelating lipid membrane was proven to be the end-on orientation while the side-on adsorption was contrarily noted in the absence of metal ions. Increase of lateral mobility owing to the fluidization effect on the chelating lipid membrane subsequently facilitated crystal formation. All these findings have opened up a potential approach for a specific orientation of immobilization of protein at the membrane interface. This could have accounted for a better opportunity of sensor development.  相似文献

Cerebral endothelial cells accomplish the barrier functions between blood and brain interstitium. Structural features are the tight junctions between adjacent endothelial cells and the formation of marginal folds at the cell-cell contacts. The glucocorticoid hydrocortisone (HC) has been reported to enforce the blood-brain-barrier in vitro measurable by an increase of the transendothelial electrical resistance. This study shows the impact of HC on the mechanical and morphological properties of confluent cell layers of brain microvascular endothelial cells. HC induces an increase in height of these marginal folds and a reduction of the intercellular contact surface. These morphological changes are accompanied by changes in cell elasticity. Staining of fibrous actin indicates that HC induces a reorganization of the actin cortex. The quantitative determination of the local elastic properties of cells reveals for the first time an HC-induced increase of the representative Young's modulus according to cytoskeletal rearrangements. For this study, cells of two different species, porcine brain capillary endothelial cells and murine brain capillary endothelial cells, were used yielding similar results, which clearly demonstrates that the HC effect on the cell elasticity is species independent.  相似文献

The enzyme L-asparaginase is a crucial component in the treatment of acute lymphoblastic leukemia (ALL). As all asparaginases in clinical use are derived from microorganisms, immunological reactions are the most important adverse events associated with asparaginase treatment. Two different methods, phage display and the SPOTs method, were used for the determination of clinically relevant epitopes. Comparison of the results showed that essentially the same domains were identified by the two methods, and thus ascertainment of relevant epitopes can be assumed. Determination of the specificity of the epitopes will be performed with serum from patients with different modes of immunological reactions and from individuals without evidence of an immune response after asparaginase administration.  相似文献

We have previously reported that (i) progression of malignancy in patients bearing astrocytic tumors correlates with increased tumor levels of galectin-1; (ii) in vitro addition of purified galectin-1 to U87 human glioblastoma cells enhances tumor cell motility; and (iii) conversely, knocking down galectin-1 expression in this cell line by stable transfection with antisense galectin-1 mRNA impairs motility and delays mortality after their intracranial grafting to nude mice. We here used cDNA microarray analysis to compare the effect on gene expression of stable transfection with antisense galectin-1 vector to mock-transfected and wild-type cells. Among the 631 spots probing genes potentially involved in cancer that were valid for analysis on all the arrays the expression of 86 genes was increased at least 2-fold. Confirmation of increased protein levels was provided by immunocytochemistry for p21waf/cip1, cullin-2, p53, ADAM-15, and MAP-2. Major differences in the expression patterns of ADAM-15 and the actin stress fiber organization were also observed. U87 cells stably deficient for galectin-1 expression were significantly less motile than control. We conclude that the stable inhibition of galectin-1 expression alters the expression of a number of genes that either directly or indirectly influence adhesion, motility and invasion of human glioblastoma cells.  相似文献

The imprinting process is described by a model evolved from physiological data. For the case of sexual imprinting in zebra finches a second sensitive phase, occurring at sexual maturity, is considered. The first sensitive phase is not restricted exclusively to the imprinting of sexual objects. Properties of the model allow for the explanation of apparent reversibility of imprinting. The same physiological mechanisms involved in the organization of simple neuronal connections are assumed to operate in the highly complex networks associated with imprinting phenomena.  相似文献

Different substances such as dimethyl sulfoxide, tetramethylene sulfoxide, 2-pyrollidone, and the naturally occurring compatible solute betaine enhance PCR amplification of GC-rich DNA templates with high melting temperatures. In particular, cyclic compatible solutes outperform traditional PCR enhancers. We therefore investigated the effects that cyclic naturally occurring ectoine-type compatible solutes and their synthetic derivatives have on melting temperature of double-stranded DNA (dsDNA) and on PCR amplification of different templates. L-ectoine, betaine, and derivatives of L-ectoine decreased, whereas beta-hydroxyectoine increased, the melting temperature of dsDNA. The ability to decrease the melting temperature was greatest for homoectoine, a new synthetic derivative of l-ectoine. Furthermore, compatible solutes, especially homoectoine, enhanced PCR amplification of GC-rich DNA (72.6% GC content; effective range: 0.1-0.5M).  相似文献

The acceptor reaction of dextransucrase from Leuconostoc mesenteroides NRRL-B512F with glucose as acceptor is of technical interest for isomaltooligosaccharide (IMOs) synthesis. Different experimental conditions were investigated for free and immobilized enzyme. The data for oligosaccharide formation up to a degree of polymerization 4 were correlated with a model developed earlier, and optimal reaction conditions for immobilized dextransucrase design and application were identified for later continuous application. Whole boar semen or seminal plasma has been demonstrated to advance the time of ovulation in gilts. As a means of clarifying this influence, the contribution of uterine lymphatics and their white cell populations has been examined. After duct visualisation with Evan's blue, lymph was sampled from a mesometrial vessel in eight pre-ovulatory gilts whose uterine lumen was infused simultaneously with whole semen in one ligated horn and saline in the contralateral ligated horn. Lymph was collected from cannulated vessels for periods of up to four hours under general anaesthesia. Thereafter, mesometrial lymph nodes, utero-tubal junction and uterine wall tissues were sampled. The proportion of nucleated cells in the sampled lymph increased towards the end of the collection period, but erythrocytes were found in all instances preventing a meaningful differentiation and identification of leukocytes. Prominent uterine lymph nodes were present in the mesometrium on both sides of the reproductive tract in 7 of 10 gilts. Differences in cellular contents were demonstrated between the side of the tract infused with semen and that infused with saline control. Two of 4 gilts had lower values for CD4 (Cluster Differentiation) and 3 of 6 gilts higher values for MHC II (Major Histocompatibility Complex) markers on the side challenged with semen. In contrast, values remained constant for CD8 but ranged widely for CD18. Immunohistochemical analysis of uterine tissue samples for MHC II+ cells revealed significant differences (P < 0.05) between the control and semen-treated ligated portions of the horns, as well as between the tissue sample of uterine wall and that from the utero-tubal junction, but there were no significant differences for CD4+ cells. It therefore remains plausible that semen-induced cytokines in the uterine lymph undergo counter-current transfer to the ipsilateral ovary and accelerate the final maturation of pre-ovulatory Graafian follicles.  相似文献

Reduction of tympanal hearing organs is repeatedly found amongst insects and is associated with weakened selection for hearing. There is also an associated wing reduction, since flight is no longer required to evade bats. Wing reduction may also affect sound production. Here, the auditory system in four silent grasshopper species belonging to the Podismini is investigated. In this group, tympanal ears occur but sound signalling does not. The tympanal organs range from fully developed to remarkably reduced tympana. To evaluate the effects of tympanal regression on neuronal organisation and auditory sensitivity, the size of wings and tympana, sensory thresholds and sensory central projections are compared. Reduced tympanal size correlates with a higher auditory threshold. The threshold curves of all four species are tuned to low frequencies with a maximal sensitivity at 3–5 kHz. Central projections of the tympanal nerve show characteristics known from fully tympanate acridid species, so neural elements for tympanal hearing have been strongly conserved across these species. The results also confirm the correlation between reduction in auditory sensitivity and wing reduction. It is concluded that the auditory sensitivity of all four species may be maintained by stabilising selective forces, such as predation.  相似文献

The availability of dense molecular markers has made possible the use of genomic selection (GS) for plant breeding. However, the evaluation of models for GS in real plant populations is very limited. This article evaluates the performance of parametric and semiparametric models for GS using wheat (Triticum aestivum L.) and maize (Zea mays) data in which different traits were measured in several environmental conditions. The findings, based on extensive cross-validations, indicate that models including marker information had higher predictive ability than pedigree-based models. In the wheat data set, and relative to a pedigree model, gains in predictive ability due to inclusion of markers ranged from 7.7 to 35.7%. Correlation between observed and predictive values in the maize data set achieved values up to 0.79. Estimates of marker effects were different across environmental conditions, indicating that genotype × environment interaction is an important component of genetic variability. These results indicate that GS in plant breeding can be an effective strategy for selecting among lines whose phenotypes have yet to be observed.PEDIGREE-BASED prediction of genetic values based on the additive infinitesimal model (Fisher 1918) has played a central role in genetic improvement of complex traits in plants and animals. Animal breeders have used this model for predicting breeding values either in a mixed model (best linear unbiased prediction, BLUP) (Henderson 1984) or in a Bayesian framework (Gianola and Fernando 1986). More recently, plant breeders have incorporated pedigree information into linear mixed models for predicting breeding values (Crossa et al. 2006, 2007; Oakey et al. 2006; Burgueño et al. 2007; Piepho et al. 2007).The availability of thousands of genome-wide molecular markers has made possible the use of genomic selection (GS) for prediction of genetic values (Meuwissen et al. 2001) in plants (e.g., Bernardo and Yu 2007; Piepho 2009; Jannink et al. 2010) and animals (Gonzalez-Recio et al. 2008; VanRaden et al. 2008; Hayes et al. 2009; de los Campos et al. 2009a). Implementing GS poses several statistical and computational challenges, such as how models can cope with the curse of dimensionality, colinearity between markers, or the complexity of quantitative traits. Parametric (e.g., Meuwissen et al. 2001) and semiparametric (e.g., Gianola et al. 2006; Gianola and van Kaam 2008) methods address these problems differently.In standard genetic models, phenotypic outcomes, , are viewed as the sum of a genetic value, , and a model residual, ; that is, . In parametric models for GS, is described as a regression on marker covariates (j = 1,  …  , p molecular markers) of the form , such that(or , in matrix notation), where is the regression of on the jth marker covariate .Estimation of via multiple regression by ordinary least squares (OLS) is not feasible when p > n. A commonly used alternative is to estimate marker effects jointly using penalized methods such as ridge regression (Hoerl and Kennard 1970) or the Least Absolute Shrinkage and Selection Operator (LASSO) (Tibshirani 1996) or their Bayesian counterpart. This approach yields greater accuracy of estimated genetic values and can be coupled with geostatistical techniques commonly used in plant breeding to model multienvironments trials (Piepho 2009).In ridge regression (or its Bayesian counterpart) the extent of shrinkage is homogeneous across markers, which may not be appropriate if some markers are located in regions that are not associated with genetic variance, while markers in other regions may be linked to QTL (Goddard and Hayes 2007). To overcome this limitation, many authors have proposed methods that use marker-specific shrinkage. In a Bayesian setting, this can be implemented using priors of marker effects that are mixtures of scaled-normal densities. Examples of this are methods Bayes A and Bayes B of Meuwissen et al. (2001) and the Bayesian LASSO of Park and Casella (2008).An alternative to parametric regressions is to use semiparametric methods such as reproducing kernel Hilbert spaces (RKHS) regression (Gianola and van Kaam 2008). The Bayesian RKHS regression regards genetic values as random variables coming from a Gaussian process centered at zero and with a (co)variance structure that is proportional to a kernel matrix K (de los Campos et al. 2009b); that is, , where , are vectors of marker genotypes for the ith and jth individuals, respectively, and is a positive definite function evaluated in marker genotypes. In a finite-dimensional setting this amounts to modeling the vector of genetic values, , as multivariate normal; that is, where is a variance parameter. One of the most attractive features of RKHS regression is that the methodology can be used with almost any information set (e.g., covariates, strings, images, graphs). A second advantage is that with RKHS the model is represented in terms of n unknowns, which gives RKHS a great computational advantage relative to some parametric methods, especially when pn.This study presents an evaluation of several methods for GS, using two extensive data sets. One contains phenotypic records of a series of wheat trials and recently generated genomic data. The other data set pertains to international maize trials in which different traits were measured in maize lines evaluated under severe drought and well-watered conditions.  相似文献
