We use specific restriction fragments as defined primers for DNA synthesis on single-stranded circular phage fd DNA. These structures are relatively poor templates for a highly purified DNA polymerase α from Xenopus laevis eggs. However, DNA synthesis is stimulated about 5-fold by addition of ATP to the reaction mixture. We show that the deoxynucleotide polymers, synthesized in the presence of ATP, are significantly longer than those produced in the absence of ATP. We also show that this effect is due to a more tenacious binding of DNA polymerase α to DNA and conclude that ATP increases the processivity of the enzyme.  相似文献

Pollen of Lilium longiflorum Thunb. was germinated for 12 h in growth medium containing 1·10-4 M chlorotetracycline (CTC), or growing tubes were treated with 1·10-4 M CTC for up to 2 h. These treatments have drastic effects: In the CTC-containing medium, out-growing tubes form only short tubes. Irregular wall thickenings are visible. Thirty minutes CTC-treatment cause growing tubes to bend and grow back toward the grain. Electron micrographs of CTC-treated tubes show that CTC affects the organelle distribution: The polar zonation of organelles is disturbed. Vesicle-and endoplasmic reticulum-accumulations are found in the wrong places, together with extensive wall thickenings and a very irregular plasma membrane. The structural details of most cell organelles look normal after CTC treatment, but the mitochondria possess unusual cristae, and microtubules are absent. The disoriented growth is interpreted as an effect of the ability of CTC to chelate intracellular calcium ions, to bind them to membranes, and thus to disturb the dynamics of the delicate Ca2+-equilibria thought to regulate oriented exocytosis.Abbreviations CTC chlorotetracycline - ER endoplasmic reticulum  相似文献

The effects of the broad-range cationophore X-537A on pollen tubes of Lilium longiflorum were investigated, using both light and electron microscopy. Pollen tube growth is completely inhibited within 30 min after the application of 5·10-5 M ionophore X-537A; cytoplasmic streaming is stopped only after 60 min of ionophore treatment. Ultrastructurally, X-537A effects are a vacuolation of Golgi cisternae and a general vacuolation. The wall is thickened at the very tip. Coated vesicles and coated regions are enriched close to and at the plasma membrane. The minicell-producing Escherichia coli strain P 678-54 was transformed with a series of defined PTY chimeric plasmids consisting of yeast 2-μm DNA and E. coli plasmid pCR1. In minicells the integrated 2-μm DNA from yeast directed specifically the synthesis of six polypeptides with apparent molecular weights of 15 000, 17 500, 20 000, 22 000, 37 000, and 48 000. The specificity of five other polypeptides, which cover a molecular weight range of 19 000 to 28 000, has not yet been established with certainty. Neither the orientation of the integrated DNA, nor the inversion which distinguishes the two structural forms of 2-μm DNA affected the polypeptides synthesized. However, integration at a given EcoRI site appeared to be correlated with the absence of one particular polypeptide band; this suggests that at least one of these sites is located in an expressed region of the DNA.  相似文献

Classically, HLA-DR expressed on antigen presenting cells (APC) initiates lymphocyte activation via presentation of peptides to TCR bearing CD4+ T-Cells. Here we demonstrate that HLA-DR alpha 2 domain (sHLA-DRalpha2) also induces negative signals by engaging TIRC7 on lymphocytes. This interaction inhibits proliferation and induces apoptosis in CD4+ and CD8+ T-cells via activation of the intrinsic pathway. Proliferation inhibition is associated with SHP-1 recruitment by TIRC7, decreased phosphorylation of STAT4, TCR-zeta chain & ZAP70, and inhibition of IFN-gamma and FasL expression. HLA-DRalpha2 and TIRC7 co-localize at the APC-T cell interaction site. Triggering HLA-DR - TIRC7 pathway demonstrates that sHLA-DRalpha2 treatment inhibits proinflammatory-inflammatory cytokine expression in APC & T cells after lipopolysaccaride (LPS) stimulation in vitro and induces apoptosis in vivo. These results suggest a novel antiproliferative role for HLA-DR mediated via TIRC7, revise the notion of an exclusive stimulatory interaction of HLA-DR with CD4+ T cells and highlights a novel physiologically relevant regulatory pathway.  相似文献

Summary 1. The present paper reviews some investigations on the problem of temperature adaptation of the eel. The experiments were made in order to find out why the metabolic rate of muscle in vitro does not reflect the capacity adaptation of the intact eel.2. Oxygen tension in the muscle tissue and in the venous blood has been measured by inserting micro oxygen electrodes. Oxygen tension in the muscle of the tail is very low; tension in the large caudal vein is more than ten times higher.3. Oxygen tension in the muscle is not altered by changing the adaptation temperature. The cold-acclimated eel shows a lower oxygen tension in the venous blood than the warm-adapted fish.4. Oxygen tension in the caudal vein depends largely on the breathing rate; this can be seen when the experimental temperature is changed and differently adapted individuals are tested. Therefore we suggest that the metabolic rate is certainly influenced by adaptive changes in the nervous system.

Kurzfassung Beim Aal weisen der Grundstoffwechsel, gemessen am Sauerstoffverbrauch des Ganztieres, und die Atmung des Muskelgewebes in vitro eine partielle Kompensation auf. Die Höhe des Standardsauerstoffverbrauchs ist aber unabhängig von der Stoffwechselaktivität eines größeren Teiles der Skelettmuskulatur. Es wird über Messungen des Sauerstoffpartialdrucks in der Muskulatur und im venösen Blut unterschiedlich adaptierter Aale berichtet. Die niedrige Sauerstoffspannung im Gewebe und die starke Abhängigkeit des Sauerstoffpartialdrucks im venösen Blut von der Atmungsintensität sprechen dafür, daß ein Mechanismus, der den Standardsauerstoffverbrauch des adaptierenden Ganztieres steuert, in einer Regulation des Kreislaufs und der Atmung zu sehen ist.
