The carotid body (CB) is the primary hypoxic chemosensory organ. Its hypoxic response appears to be genetically controlled. We have hypothesized that: 1) genes related to CB function are expressed less in the A/J mice (low responder to hypoxia) compared with DBA/2J mice (high responder to hypoxia); and 2) gene expression levels of morphogenic and trophic factors of the CB are significantly lower in the A/J mice than DBA/2J mice. This study utilizes microarray analysis to test these hypotheses. Three sets of CBs were harvested from both strains. RNA was isolated and used for global gene expression profiling (Affymetrix Mouse 430 v2.0 array). Afromontane forests, like those in the Aberdare National Park (ANP) in Kenya, sustain unique avifaunal assemblages. There is a growing need for biodiversity inventories for Afromontane forests, especially through the utilisation of unskilled observers. Acoustic surveys are a potential aid to this, but more comparisons of this technique with that of traditional point counts are needed. We conducted a systematic survey of the ANP avifauna, assessing whether acoustic and traditional surveys resulted in different species richness scores, and whether this varied with habitat and species characteristics. We also investigated the role of habitat and elevation in driving variation in species richness. The ANP provides habitat types including scrub, moorland, montane, hagenia and bamboo forests. Overall, the surveys yielded 101 identified species. The acoustic method resulted in higher species richness scores compared to the traditional method across all habitats, and the relative performance of the two methods did not vary with habitat type or visibility. The methods detected different species, suggesting that they should be used together to maximise the range of species recorded. We found that habitat type was the primary driver of variation in species richness, with scrub and montane forest having higher species richness scores than other habitats.  相似文献

Therapeutically exploiting vascular and metabolic endpoints becomes critical to translational cancer studies because altered vascularity and deregulated metabolism are two important cancer hallmarks. The metabolic and vascular phenotypes of three sibling breast tumor lines with different metastatic potential are investigated in vivo with a newly developed quantitative spectroscopy system. All tumor lines have different metabolic and vascular characteristics compared to normal tissues, and there are strong positive correlations between metabolic (glucose uptake and mitochondrial membrane potential) and vascular (oxygen saturations and hemoglobin concentrations) parameters for metastatic (4T1) tumors but not for micrometastatic (4T07) and nonmetastatic (67NR) tumors. A longitudinal study shows that both vascular and metabolic endpoints of 4T1 tumors increased up to a specific tumor size threshold beyond which these parameters decreased. The synchronous changes between metabolic and vascular parameters, along with the strong positive correlations between these endpoints suggest that 4T1 tumors rely on strong oxidative phosphorylation in addition to glycolysis. This study illustrates the great potential of our optical technique to provide valuable dynamic information about the interplay between the metabolic and vascular status of tumors, with important implications for translational cancer investigations.   相似文献

There are two principal types of nickel (Ni) deposits: sulfide and laterite ores. Interest in low-grade Ni-laterite ores has increased in recent years as high-grade Ni-sulfide deposits are being quickly depleted. However, processing of Ni laterites has proven technically difficult and costly, and the development of alternative low-cost biotechnologies for Ni solubilization has been encouraged. In this context, by the first time, a sample of Brazilian Ni-laterite ore was analyzed mineralogically and subjected to bioleaching tests using a heterotrophic Bacillus subtilis strain. SEM-analysis indicated that the primary Ni carrier mineral is goethite. Chemical analysis of different grain size fractions indicated a homogeneous distribution of Ni. XRF-analysis showed that the ore consists mainly in lizardite (32.6% MgO) and contains1.0% NiO (0.85% Ni). Bioleaching batch experiments demonstrated that about 8.1% Ni (0.7 mg Ni/g ore) were solubilized by the B. subtilis after 7 days. Application of microwave heating as a Ni-laterite pretreatment was also tested. This pretreatment increased the bioextraction of Ni from 8% to 26% (2.3 mg Ni g−1 ore).  相似文献

The invasive frugivore Drosophila suzukii (Matsumura) (Diptera: Drosophilidae) utilizes a wide range of host plants and damages important fruit crops, including blueberries, cherries, blackberries, raspberries, and strawberries. Field infestations of D. suzukii often exceed one larva per berry, suggesting that intraspecific competition may frequently occur. Because dietary resources are also likely to vary across the host range of D. suzukii, we designed a laboratory assay to measure larval performance across diets of varying quality: a standard artificial diet, a fruit‐based medium, a low‐protein, and a low‐carbohydrate diet. We manipulated egg density across these diets to provide increasing levels of competition and measured larval performance by observing survival to pupation and adulthood, and development times for both life stages. Although increasing density generally negatively impacted D. suzukii performance across diets, the magnitude of these impacts varied by diet type. Drosophila suzukii performance was generally similar in fruit and standard diets, although larval development was more rapid in fruit diets at lower densities. Even at low densities (5 or 10 eggs per arena), survival was reduced and development time increased in low‐protein diets relative to standard and fruit diets. At the two highest larval densities (20 or 40 eggs per arena), survivorship was reduced in low‐carbohydrate diets as compared to standard and fruit diets. There is evidence that larvae compensated in both low‐quality diets by extending development time, which could have consequences for population dynamics. Population models for use in D. suzukii management may need to account for both host nutritional quality and relative competition to accurately predict turnover and geographic expansion.  相似文献
