The ectoparasitic mite Pyemotes tritici (Lagrèze-Fossat & Montané) (Acari: Pyemotidae) caused paralysis and reduced longevity in eucalyptus longhorned borer, Phoracantha semipunctata F., under laboratory rearing conditions. Application of dusting sulfur to logs that contained pupating borers greatly reduced densities of mites on emerging adult beetles and increased beetle survivorship. Uniform application to all logs in a glasshouse effectively eradicated the mite infestation. A bioassay showed that sulfur may physically impede the dispersal of immature mites by adhering to the cuticle, but sulfur vapor did not act as a toxin.  相似文献

We tested the hypothesis that contact phermones mediate mate recognition for four species of longhorned beetles, Neoclytus mucronatus mucronatus (F.), Megacyllene caryae (Gahan), Megacyllene robiniae (Forster), and Plectrodera scalator (F.). All tested males of all four species attempted to mate with females only after contacting them with their antennae. From 66.7 to 80% of tested males attempted to mate with hexane-extracted dead females treated with 0.1–1.0 female equivalents of conspecific female extracts, confirming that nonpolar compounds on the cuticle of females are essential for mate recognition in all four species. These findings are further evidence of the critical role of contact pheromones in mating systems of longhorned beetles.  相似文献

The Eucalyptus longhorned borer, Phoracantha semipunctata (F.), is native to Australia, but it has been introduced without its natural enemies into many parts of the world in which its Eucalyptus spp. host has been planted. The beetle has developed large populations in these novel habitats and has been responsible for the mortality of large numbers of trees. Although there is a considerable catalogue of the parasitoids of the beetle in Australia, limited ecological information on the assemblage of parasitoids attacking P. semipunctata is available. We removed bark from 40 felled trees, recorded gallery width and bark thickness over parasitized larvae, and removed all parasitoids. Adult size, sex, and species were recorded when the parasitoid pupae eclosed. Syngaster lepidus Brullè, Jarra phoracantha Austin, Quicke, and Marsh, J. maculipennis Austin, Quicke, and Marsh, and J. painei Austin and Dangerfield were most commonly collected. The solitary parasitoid S. lepidus preferred smaller larvae than did the gregarious Jarra spp. The two species with shorter ovipositors, J. maculipennis and J. painei, parasitized larvae under thinner bark than did the other two species with longer ovipositors. There was a significant positive correlation between host larval size and number of parasitoid pupae of the gregarious species. Also, there was a significant positive correlation between host larval size and parasitoid adult size. The ecological relationships between this assemblage of parasitoids and their beetle host may be useful in establishing an effective biological control program.  相似文献

Sotos syndrome is a childhood overgrowth syndrome characterized by a distinctive facial appearance, height and head circumference >97th percentile, advanced bone age, and developmental delay. Weaver syndrome is characterized by the same criteria but has its own distinctive facial gestalt. Recently, a 2.2-Mb chromosome 5q35 microdeletion, encompassing NSD1, was reported as the major cause of Sotos syndrome, with intragenic NSD1 mutations identified in a minority of cases. We evaluated 75 patients with childhood overgrowth, for intragenic mutations and large deletions of NSD1. The series was phenotypically scored into four groups, prior to the molecular analyses: the phenotype in group 1 (n=37) was typical of Sotos syndrome; the phenotype in group 2 (n=13) was Sotos-like but with some atypical features; patients in group 3 (n=7) had Weaver syndrome, and patients in group 4 (n=18) had an overgrowth condition that was neither Sotos nor Weaver syndrome. We detected three deletions and 32 mutations (13 frameshift, 8 nonsense, 2 splice-site, and 9 missense) that are likely to impair NSD1 functions. The truncating mutations were spread throughout NSD1, but there was evidence of clustering of missense mutations in highly conserved functional domains between exons 13 and 23. There was a strong correlation between presence of an NSD1 alteration and clinical phenotype, in that 28 of 37 (76%) patients in group 1 had NSD1 mutations or deletions, whereas none of the patients in group 4 had abnormalities of NSD1. Three patients with Weaver syndrome had NSD1 mutations, all between amino acids 2142 and 2184. We conclude that intragenic mutations of NSD1 are the major cause of Sotos syndrome and account for some Weaver syndrome cases but rarely occur in other childhood overgrowth phenotypes.  相似文献

Focal adhesion kinase (FAK) is a key signaling molecule regulating cellular responses to integrin-mediated adhesion. Integrin engagement promotes FAK phosphorylation at multiple sites to achieve full FAK activation. Phosphorylation of FAK Tyr-397 creates a binding site for Src-family kinases, and phosphorylation of FAK Tyr-576/Tyr-577 in the kinase domain activation loop enhances catalytic activity. Using novel phosphospecific antibody reagents, we show that FAK activation loop phosphorylation is significantly elevated in cells expressing activated Src and is an early event following cell adhesion to fibronectin. In both cases, this regulation is largely dependent on Tyr-397. We also show that the FAK activation loop tyrosines are required for maximal Tyr-397 phosphorylation. Finally, immunostaining analyses revealed that tyrosine-phosphorylated forms of FAK are present in both newly forming and mature focal adhesions. Our findings support a model for reciprocal activation of FAK and Src-family kinases and suggest that FAK/Src signaling may occur during both focal adhesion assembly and turnover.  相似文献

Subcellular organelle fractionation of nitrogen-fixing nodules of soybean (Glycine max (L.) Merr.) indicates that a number of enzymes involved in the assimilation of ammonia into amino acids and purines are located in the proplastids. These include asparagine synthetase (EC 6.3.1.1), phosphoribosyl amidotransferase (EC 2.4.2.14), phosphoglycerate dehydrogenase (EC 1.1.1.95), serine hydroxymethylase (EC 2.1.2.1), and methylene-tetrahydrofolate dehydrogenase (EC 1.5.1.5). Of the two isoenzymes of asparate aminotransferase (EC 2.6.1.1) in the nodule, only one was located in the proplastid fraction. Both glutamate synthase (EC 1.4.1.14) and triosephosphate isomerase (EC 5.3.1.1) were associated at least in part with the proplastids. Glutamine synthetase (EC 6.3.1.2) and xanthine dehydrogenase (EC 1.2.1.37) were found in significant quantities only in the soluble fraction. Phosphoribosylpyrophosphate synthetase (EC 2.7.6.1) was found mostly in the soluble fraction, although small amounts of it were detected in other organelle fractions. Summary The genetical control of six characters, which were taken as jointly reflecting the overall shape of the plant, was analysed using four true-breeding lines of Nicotiana rustica. F1 F2 and first backcross generations were raised from all of the possible pairwise combinations between the lines. The particular relationships between the lines provided a basis for the analysis which was an extension of the normal model fitting procedures described by Mather and Jinks (1971).The first step in the analysis was to test whether the allelic differences present between the inbred lines p1 and P5 had been maintained in the two lines B2 and B35, derived from an earlier cross between the former. If the allelic differences between p1 and P5 were present between B2 and B35, it was possible to proceed straight-forwardly by fitting a model consisting of m, two symmetrical [d]'s and the relevant non-additive parameters. If B2 and B35 were homozygous for the same alleles at loci by which p1 and P5 differed, in other words if significant asymmetry in the gene distributions was present, the model had to be extended to cover the effects of such genes.All six characters investigated were shown to be subject to genetical variation. From the composition of the genetical models that were necessary to account for the observations from each of the characters, it was inferred that they should be amenable to at least partially independent adjustment by selection.  相似文献

Transplantation of hematopoietic stem cells from the peripheral blood   总被引:7,自引:2,他引:7  
Hematopoietic stem cells can be collected from the peripheral blood. These hematopoietic stem cells (HSC), or better progenitor cells, are mostly expressed as the percentage of cells than react with CD34 antibodies or that form colonies in semi-solid medium (CFU-GM). Under steady-state conditions the number of HSC is much lower in peripheral blood than in bone marrow. Mobilization with chemotherapy and/or growth factors may lead to a concentration of HSC in the peripheral blood that equals or exceeds the concentration in bone marrow. Transplantation of HSC from the peripheral blood results in faster hematologic recovery than HSC from bone marrow. This decreases the risk of infection and the need for blood-product support. For autologous stem-cell transplantation (SCT), the use of peripheral blood cells has completely replaced the use of bone marrow. For allogeneic SCT, on the other hand, the situation is more complex. Since peripheral blood contains more T-lymphocytes than bone marow, the use of HSC from the peripheral blood increases the risk of graft-versus-host disease after allogeneic SCT. For patients with goodrisk leukemia, bone marrow is still preferred, but for patients with high-risk disease, peripheral blood SCT has become the therapy of choice.  相似文献

Objective

How do the holidays – and the possible New Year’s resolutions that follow – influence a household’s purchase patterns of healthier foods versus less healthy foods? This has important implications for both holiday food shopping and post-holiday shopping.

Methods

207 households were recruited to participate in a randomized-controlled trial conducted at two regional-grocery chain locations in upstate New York. Item-level transaction records were tracked over a seven-month period (July 2010 to March 2011). The cooperating grocer’s proprietary nutrient-rating system was used to designate “healthy,” and “less healthy” items. Calorie data were extracted from online nutritional databases. Expenditures and calories purchased for the holiday period (Thanksgiving-New Year’s), and the post-holiday period (New Year’s-March), were compared to baseline (July-Thanksgiving) amounts.

Results

During the holiday season, household food expenditures increased 15% compared to baseline ($105.74 to $121.83; p<0.001), with 75% of additional expenditures accounted for by less-healthy items. Consistent with what one would expect from New Year’s resolutions, sales of healthy foods increased 29.4% ($13.24/week) after the holiday season compared to baseline, and 18.9% ($9.26/week) compared to the holiday period. Unfortunately, sales of less-healthy foods remained at holiday levels ($72.85/week holiday period vs. $72.52/week post-holiday). Calories purchased each week increased 9.3% (450 calories per serving/week) after the New Year compared to the holiday period, and increased 20.2% (890 calories per serving/week) compared to baseline.

Conclusions

Despite resolutions to eat more healthfully after New Year’s, consumers may adjust to a new “status quo” of increased less-healthy food purchasing during the holidays, and dubiously fulfill their New Year’s resolutions by spending more on healthy foods. Encouraging consumers to substitute healthy items for less-healthy items may be one way for practitioners and public health officials to help consumers fulfill New Year’s resolutions, and reverse holiday weight gain.  相似文献
