【背景】出芽短梗霉菌株PA-2是一株分离自青海省海东市平安区患病杨树叶片上的真菌,前期研究表明该菌株具有除草和抑菌能力,说明其在生物农药方面具有潜在的应用前景。【目的】了解菌株PA-2的基因组序列信息,挖掘其生防相关功能基因簇,为进一步研究解析该菌株生防机理及生防功能改造提供遗传背景信息。【方法】利用IlluminaHiSeq高通量测序平台对生防菌株PA-2进行全基因组测序,用生物信息学的方法对测序数据进行基因组组装、基因预测及功能注释、碳水化合物活性酶预测、次级代谢产物合成基因簇预测,利用刚果红染色等方法对水解酶活性进行衡量。【结果】菌株PA-2基因组序列全长28 932 793 bp,平均GC含量为50%,共编码10 839个基因,预测到该菌株具有4个已知的次级代谢产物合成基因簇,编码Melanin、Burnettramic Acid A、ACR-Toxin I、Abscisic Acid,该菌株能水解纤维素和果胶。【结论】有助于在基因组层面上解析菌株PA-2生防机制的内在原因,为深入了解出芽短梗霉菌次级代谢物合成途径提供参考,对菌株PA-2的下一步相关研究具有重要意义。  相似文献

为了探究甘蓝型油菜中钙依赖蛋白激酶(calcium dependent protein kinase, CPK)在植物对逆境响应中的作用和机制,同时为油菜品质的升级改良发掘新的基因资源,开展了对于BnaCPK6研究的分子生物学试验。首先,通过在本氏烟草中瞬时表达BnaCPK6与GFP融合蛋白来检测它的亚细胞定位情况;其次,利用双分子荧光互补(Bimolecular fluorescence complementation)试验来检测BnaCPK6与ABA信号通路中转录因子BanABF1/3/4、BnaABI5、BnaAREB3的相互作用情况。结果显示BnaCPK6具有典型的钙依赖蛋白激酶特征,N端具有潜在的棕榈酰化和豆蔻酰化位点,并且与AtCPK6在进化上有着很高的同源性。亚细胞定位的结果发现BnaCPK6主要分布于细胞膜和细胞核中。同时,双分子荧光互补试验还发现BnaCPK6与调控ABA信号转导的关键转录因子BnaABF3/4、BnaABI5以及BnaAREB3之间存在相互作用。本研究为进一步研究BnaCPK6在ABA信号通路中的作用提供了依据。  相似文献

唐家河自然保护区小哺乳动物空间生态位初步研究   总被引:3,自引:0,他引:3  
1989年4~11月,采用去除取样法,对四川省唐家河自然保护区内小哺乳动物的空间生态位宽度和重叠度进行了研究。共安置ll000铗日,捕获小哺乳动物14种846只。根据香农一威纳多样性指数公式以及Cowell和Futuyma公式分别计算了各小哺乳动物种群的空间生态位宽度和空间生态位重叠指数。在14种小哺乳动物中,社鼠的空间生态位宽度指数最大(1.00),大足鼠、川I西长尾朐、川朐和灰麝朐的最小,均为0。14种小哺乳动物空间生态位重叠指数最大为1,最小为0,其中大足鼠与川西长尾朐及灰麝朐、针毛鼠与川朐的空间生态位彼此完全重叠;藏鼠兔与针毛鼠、大足鼠、洮洲绒鼠、川西长尾朐、川朐以及灰麝朐的空间生态位完全分离。空间生态位完全重叠的种类,是通过选择不同的食物以及不同的微生境来避免过度竞争。回归分析显示,小哺乳动物种群的空间生态位宽度指数与它们利用空间资源的平均值呈显著的正相关(r=0.8285,P〈0.05)。空间生态位宽度指数的季节变化表明,社鼠种群分布没有明显的季节差异,中华姬鼠和安氏白腹鼠每年可能有一个数量高峰期,高山姬鼠一年则可能有两个数量高峰。  相似文献

Purple apiculus is one of the important agronomic traits of rice. Single-segment substitution line (SSSL) W23-07-6-02-14 in the genetic background of an elite rice variety Huajingxian74 (HJX74) with the substituted interval of RM225-RM217-RM253 on the chromosome 6 was found to have purple apiculus (Pa). To map the gene governing Pa, W23-07-6-02-14 was crossed with the recipient HJX74 to develop an F2 secondary segregation population. The ratio of purple apiculus to green apiculus showed a good fit to 3:1 ratio, indicating that Pa was controlled by a major dominant gene. The gene locus for Pa was tentatively designated as Pa-6. Using 430 individuals from the F2 segregation population, the Pa-6 locus was mapped between two SSR markers RM19556 and RM19561 with genetic distances of 0.2 and 0.3 cM, respectively. For fine mapping of the Pa-6 gene, a large F2:3 segregation population of 3890 individuals was developed from F2 heterzygous plants in the RM19556-RM19561 region. Recombinant analyses further mapped the Pa-6 gene locus to an interval of 41.7-kb bounded L02 and RM19561. Sequence analysis of this 41.7-kb region revealed that it contains eleven open reading frames (ORFs), of which, ORF5 is classified as the one that is associated with the C (chromogen for anthocyanin) gene, it was presumed to be the candidate gene for Pa. This result provided a foundation of map-based cloning and function analysis of the Pa-6 gene.  相似文献

CBL-CIPK是高等植物中广泛存在的一类解析Ca~(2+)信号的蛋白。该研究在前期工作基础上,对甘蓝型油菜(Brassica napus L.)的BnaCIPK15基因进行了亚细胞定位、双分子荧光互补(BiFC)、酵母双杂交和qRT-PCR检测等一系列分析,以探究BnaCIPK15蛋白在ABA激素响应中的作用。结果显示:(1)亚细胞定位发现,BnaCIPK15蛋白定位于细胞质和细胞核中; BiFC分析发现,BnaCIPK15蛋白与BnaCBL1/3/4/9蛋白之间的互作较强,与BnaCBL10仅有微弱互作。(2)qRT-PCR检测发现,BnaCIPK15基因受ABA和冷胁迫的诱导极显著上调表达,而对百草枯(Paraquat)、活性氧(H_2O_2)和热胁迫的诱导较弱,表明BnaCIPK15基因很可能参与ABA和冷胁迫的调控过程。(3)酵母滴定实验结果显示,BnaCIPK15蛋白与脱落酸(ABA)信号通路中的BnaHAB1蛋白(属于蛋白磷酸酶PP2C家族)存在明显的互作,而与BnaABFs/AREB3/ABI5转录因子无明显互作;BiFC验证显示,BnaCIPK15与BnaHAB1蛋白之间存在互作信号,而BnaCIPK15与BnaHAB2组合没有观察到信号,证明BnaCIPK15与BnaHAB1磷酸酶具有特异互作特征,推测BnaCIPK15可能参与调控ABA信号转导。研究认为,甘蓝型油菜中可能存在基于BnaCIPK15-BnaHAB1的互作模块,并参与ABA的信号转导和网络调控。  相似文献

The current Na+ storage performance of carbon‐based materials is still hindered by the sluggish Na+ ion transfer kinetics and low capacity. Graphene and its derivatives have been widely investigated as electrode materials in energy storage and conversion systems. However, as anode materials for sodium‐ion batteries (SIBs), the severe π–π restacking of graphene sheets usually results in compact structure with a small interlayer distance and a long ion transfer distance, thus leading to low capacity and poor rate capability. Herein, partially reduced holey graphene oxide is prepared by simple H2O2 treatment and subsequent low temperature reduction of graphene oxide, leading to large interlayer distance (0.434 nm), fast ion transport, and larger Na+ storage space. The partially remaining oxygenous groups can also contribute to the capacity by redox reaction. As anode material for SIBs, the optimized electrode delivers high reversible capacity, high rate capability (365 and 131 mAh g?1 at 0.1 and 10 A g?1, respectively), and good cycling performance (163 mAh g?1 after 3000 cycles at a current density of 2 A g?1), which is among the best reported performances for carbon‐based SIB anodes.  相似文献

Emergy analysis is able to account for ecosystems’ contribution to industrial activity. Accordingly, it is an ecologically conscious tool useful for assessing the environmental impact and sustainability of industrial systems. The emergy-based approach requires proper system boundary definitions and uses several standard indices. In this article some perspectives on the deficiencies of three standard emergy indicators - environmental loading ratio (ELR), emergy yield ratio (EYR) and emergy index of sustainability (EIS) - when applied to industrial systems involving waste management are put forward and suggestions for overcoming them given. In addition, in order to account for the impact of waste emissions on the environment, a simple impact amplification factor is proposed for inclusion in the improved emergy indicators. To demonstrate their usefulness and highlight their superiority over standard indices, the improved emergy indicators are used to evaluate the interaction between a commercial polyethylene production process incorporating waste management and its surrounding environment.  相似文献

唐家河自然保护区小型兽类生物量的初步研究   总被引:1,自引:1,他引:0  
1989年4~11月在四川省唐家河自然保护区,用去除取样法对该保护区内小型兽类群落的物种密度和生物量进行研究。共安置11000多铗日,捕获小型兽类846只,隶属21种。采用回归方法计算出唐家河自然保护区4个不同小型兽类群落夏季物种的密度,分别为18·85、23·01、15·99和25·38只·hm-2;平均密度为19·49只·hm-2。夏季生物量分别为1053·9、1497·13、1051·99和1815·15g·hm-2,平均生物量为1273·04g·hm-2。在唐家河自然保护区小型兽类群落中,主要优势种社鼠的生物量在群落C1、C2、C3分别达到91%、90%、79%,另一优势种藏鼠兔的生物量则占群落4总生物量的79%。它们决定了唐家河自然保护区小型兽类群落的主要特征。  相似文献
