Numerical assessment is made regarding Pauly and Schwan's theory which describes the dielectric behaviour of a suspension of “shell spheres” as a model of biological membrane-bounded particles. The results indicate that approximate expressions of the theory may give rise to serious errors when applied to particles smaller than about 1 Μm in diameter. With a view to performing analysis according to a general expression of the theory, some of the characteristic responses of dielectric parameters upon changes in phase parameters are examined with particular reference to some numerical ranges of biological interest. On this basis a simplified and systematic procedure is proposed for estimating the phase parameters of particles whose shell phase can be regarded as non-conductive. As the application of the procedure proposed, a set of dielectric data of a synaptosome suspension is analyzed, so that the following three phase parameters are successfully determined: membrane capacitance (or shell phase dielectric constant), internal phase conductivity and internal phase dielectric constant. Some limitations of the procedure are discussed for the cases of conducting shells and small particles.  相似文献

Dielectric measurements of Escherichia coli suspensions were carried out over a frequency range from 10 kHz to 100 MHz, and marked dielectric dispersions having characteristic frequency of approximately 1 MHz were observed. On the basis of the cell model that a spheroid is covered with two confocal shells, a dielectric theory was developed to determine accurately four electrical parameters for E. coli cells such as the conductivity of the cell wall, the dielectric constant of the cell membrane, and the dielectric constant and the conductivity of the protoplasm. The observed data were analyzed by means of the procedure based on the dielectric theory to yield a set of plausible electrical parameters for the cells. By taking account of the size distribution of the cells and a dielectric relaxation of the protoplasm, the observed dispersion curves were successfully reconstituted by the present theory.  相似文献

An electrochemical surface shear stress measurement was applied to a model of unilateral arterial stenosis. The unilateral stenosis model was made up of a removable stenosis plug, in an electrochemical shear stress measurement test section with 100 cathodes. Three dimensional wall shear stress distribution was measured under steady flow field. At a relatively low Reynolds number, Re = 270, there was a characteristic high and low wall shear distribution pattern downstream of the unilateral stenosis. There were also remarkable high shear stress areas on the opposite wall up- and downstream, and both side walls of the stenosis upstream. It was clearly shown that detailed three dimensional structure of the flow field must be studied in order to correlate it to pathological findings.  相似文献

A re-evaluation of the dielectric studies on isolated mitochondria (Pauly, H., Packer L., and Schwan, H.P. (1960) J. Biophys. Biochem. Cytol. 7, 589–601, and ibid. 7, 603–612) is presented. The suspensions of ‘mitoplasts’ prepared from rat liver mitochondria by a hyposmotic (10 mM KCI) treatment showed a dielectric dispersion with its characteristic frequency lying in the 1–100 MHz range. In the analysis of data special emphasis was put on the choice of the theoretical models to employ after serutiny of their applicability to the suspensions tested. As such we adopted the theory of Hanai et al. (Hanai, T., Asami, K., and Koizumi, N. (1979) Bull. Inst. Chem. Res., Kyoto Univ. 57, 297–305) that was advanced to include concentrated suspensions of shelled spheres. Curve fittings based on that theory resulted in a better agreement with experiment than the fittings based on a conventional theory for dilute suspensions. Major findings from our analyses on the swollen mitoplasts are that: (i) the limiting membrane of the mitoplasts has a specific electrical capacity of 1 μF/cm2, (ii) the ratio of permittivity (or dielectric constant) for the mitoplast interior and permittivity for the external medium is 0.6–0.7, and (iii) the conductivity ratio between the interior phase and the medium is approx. 0.6. Reasons for discrepancy between the results of Pauly et al. and ours are discussed.  相似文献

When the competitive inhibitor benzeneboronic acid (BBA) forms a complex with alpha-chymotrypsin [EC 3.4.21.1] protons are released in the acidic pH region. The proton release can be measured by a difference potentiometric technique. The proton release is also observed in chymotrypsinogen A but not in TRCK-, DIP-, and anhydrochymotrypsins. Based on these observations, a simple procedure to estimate the equilibrium constants of the trigonal-tetrahedral interconversion of BBA is proposed. Thermodynamic parameters of the ionization of His 57 and of each step involved in BBA binding can be estimated from the temperature dependence of the proton release. Those of His 57 are essentially the same as those of imidazole in water. Regarding the interconversion of BBA on the enzyme, the value of delta S is similar to delta S not equal to of the deacylation step of nonspecific substrates, and delta H is remarkably reduced from that for the ionization of BBA in water. The enthalpic gain of enzymic process is suggested to be due to the change of the proton acceptor, which is water in the case of the ionization of BBA in water, to imidazole on the enzyme.  相似文献

Poly(ADP-ribose) polymerase (PARP) is conserved in eukaryotes. To analyze the function of PARP, we isolated and characterized the gene for PARP in Drosophila melanogaster. The PARP gene consisted of six translatable exons and spanned more than 50 kb. The DNA binding domain is encoded by exons 1-4. Although the consensus cleavage site of CED-3 like protease during apoptosis is conserved from human to Xenopus laevis PARPs, it is neither conserved in the corresponding region of Drosophila nor Sarcophaga peregrina. There are two cDNAs species in Drosophila. One cDNA could encode the full length PARP protein (PARP I), while the other is a truncated cDNA which could encode a partial-length PARP protein (PARP II), which lacks the automodification domain and is possibly produced by alternative splicing. The expression of these two forms of PARP in E. coli demonstrated that while PARP II has the catalytic NAD-binding domain and DNA-binding domain it is enzymatically inactive. On the other hand PARP I is active. A deletion mutant of PARP gene could grow to the end of embryogenesis but did not grow to the adult fly. These results suggest that the PARP gene plays an important function during the development of Drosophila.  相似文献

BALB/c3T3 cell homogenates have guanylate cyclase in both 105000 g paniculate and soluble fraction. The activity in particulate fraction was much higher than that in the soluble fraction. Both enzyme activities were 2- to 3-fold greater in the resting state (G0 phase) than in the logarithmically growing state. In addition, cGMP-phosphodiesterase activity was 2-fold greater in resting than in growing cells. When G0-arrested cells entered into the G1 phase by serum addition, cGMP levels rapidly increased, whereas guanylate cyclase activities did not change within 30 min after serum addition. Four hours after serum addition, these activities had, however, decreased to one third and remained at that low level throughout the G1 phase. The relationship between cell growth and guanylate cyclase activity is discussed.  相似文献

A theory of complex dielectric constant (ε1) for the suspension of “multi-stratified” spherical particles is presented. Based on Maxwell's theory of interfacial polarization, we derive a general expression which correlates ε1 with the electrical and geometrical parameters of each stratum. It can be shown that such a “multi-stratified” system in general should give rise to multiple dielectric dispersions, the number of which corresponds to the number of interfaces lying between the successive shell phases. The conditions for a full number of different “unit” dispersions to occur are also discussed. As an example, a special case of the “double-shell” model consisting of a spherical core and three layers of concentric phases is solved numerically by using a set of parameter values pertinent to a lymphoma cell. In light of the characteristic behavior of ε1 thus revealed, we propose a scheme of procedure that applies to the determination of electrical parameters associated with the specific “double-shell” model.  相似文献

Na+-dependent Ca2+ efflux was demonstrated in cells isolated from the rat renal cortex, suggestive of the presence of a Na+/Ca2+ exchange carrier in the cells. Parathyroid hormone, when incubated with the cells in vitro, increased Na+-dependent Ca2+ efflux about 60%. The effect of the hormone was specific for biologically active parathyroid hormone analogs and could be mimicked by cyclic nucleotides and forskolin. The effects of parathyroid hormone concentration on Ca2+ efflux and cyclic AMP formation were similar. These findings would be consistent with the view that the cyclic nucleotide might act as the intracellular messenger to increase Na+/Ca2+ exchange activity. Cells isolated from parathyroidectomized rats had decreased Na+-dependent Ca2+ efflux. When these cells were treated in vitro with parathyroid hormone, Na+-dependent Ca2+ efflux was enhanced to the same rate as found with cells from sham-operated animals. Parathyroid hormone-sensitive Na+/Ca2+ exchange activity was markedly blunted in cells from senescent (24 months) rats. Basal Na+-dependent Ca2+ efflux and Na+-independent Ca2+ efflux were not altered in the aged animal. Parathyroid-stimulated adenylate cyclase was also decreased in aging. In contrast, forskolin-stimulated Na+-dependent Ca2+ efflux and adenylate cyclase did not change with senescence. These findings would be compatible with a mechanism of desensitization that occurred at the level of the receptor or hormone-receptor coupling to adenylate cyclase. These results may be of physiological significance in understanding calcium homeostasis and the imbalances in mineral metabolism associated with old age.  相似文献
