Bayesian networks are knowledge representation tools that model the (in)dependency relationships among variables for probabilistic reasoning. Classification with Bayesian networks aims to compute the class with the highest probability given a case. This special kind is referred to as Bayesian network classifiers. Since learning the Bayesian network structure from a dataset can be viewed as an optimization problem, heuristic search algorithms may be applied to build high-quality networks in medium- or large-scale problems, as exhaustive search is often feasible only for small problems. In this paper, we present our new algorithm, ABC-Miner, and propose several extensions to it. ABC-Miner uses ant colony optimization for learning the structure of Bayesian network classifiers. We report extended computational results comparing the performance of our algorithm with eight other classification algorithms, namely six variations of well-known Bayesian network classifiers, cAnt-Miner for discovering classification rules and a support vector machine algorithm. 相似文献
DNA replication is tightly controlled in eukaryotic cells to ensure that an exact copy of the genetic material is inherited by both daughter cells. Oscillating waves of cyclin-dependent kinase (CDK) and anaphase-promoting complex/cyclosome (APC/C) activities provide a binary switch that permits the replication of each chromosome exactly once per cell cycle. Work from several organisms has revealed a conserved strategy whereby inactive replication complexes are assembled onto DNA during periods of low CDK and high APC activity but are competent to execute genome duplication only when these activities are reversed. Periods of high CDK and low APC/C serve an essential function by blocking reassembly of replication complexes, thereby preventing rereplication. Higher eukaryotes have evolved additional CDK-independent mechanisms for preventing rereplication.The Eukarya include a wide spectrum of organisms, with genome sizes ranging from ∼107 bp in yeasts to ∼1012 bp in protozoa. Rapid duplication of large genomes is achieved by distribution of the genetic material across several chromosomes. Each of these chromosomes initiates replication from sites called replication origins, which must fire no more than once per cell cycle to ensure a single error-free copy of the genome. Generating replication forks from an origin more than once leads to rereplication, an event that creates multiple copies of a single genomic region within a single cell. This leads to gene amplification and promotes genome instability (Green et al. 2010), a phenomenon observed in many human cancers (Lengauer et al. 1998). The process of genome duplication is therefore under stringent control to ensure that few, if any, defects are transmitted from one generation to the next. 相似文献
Large swaths of the nutrient‐poor surface ocean are dominated numerically by cyanobacteria (Prochlorococcus), cyanobacterial viruses (cyanophage), and alphaproteobacteria (SAR11). How these groups thrive in the diverse physicochemical environments of different oceanic regions remains poorly understood. Comparative metagenomics can reveal adaptive responses linked to ecosystem‐specific selective pressures. The Red Sea is well‐suited for studying adaptation of pelagic‐microbes, with salinities, temperatures, and light levels at the extreme end for the surface ocean, and low nutrient concentrations, yet no metagenomic studies have been done there. The Red Sea (high salinity, high light, low N and P) compares favorably with the Mediterranean Sea (high salinity, low P), Sargasso Sea (low P), and North Pacific Subtropical Gyre (high light, low N). We quantified the relative abundance of genetic functions among Prochlorococcus, cyanophage, and SAR11 from these four regions. Gene frequencies indicate selection for phosphorus acquisition (Mediterranean/Sargasso), DNA repair and high‐light responses (Red Sea/Pacific Prochlorococcus), and osmolyte C1 oxidation (Red Sea/Mediterranean SAR11). The unexpected connection between salinity‐dependent osmolyte production and SAR11 C1 metabolism represents a potentially major coevolutionary adaptation and biogeochemical flux. Among Prochlorococcus and cyanophage, genes enriched in specific environments had ecotype distributions similar to nonenriched genes, suggesting that inter‐ecotype gene transfer is not a major source of environment‐specific adaptation. Clustering of metagenomes using gene frequencies shows similarities in populations (Red Sea with Pacific, Mediterranean with Sargasso) that belie their geographic distances. Taken together, the genetic functions enriched in specific environments indicate competitive strategies for maintaining carrying capacity in the face of physical stressors and low nutrient availability. 相似文献
Viruses are obligatory parasites that depend on host cellular factors for their replication as well as for their local and systemic movement to establish infection. Although myosin motors are thought to contribute to plant virus infection, their exact roles in the specific infection steps have not been addressed. Here we investigated the replication, cell-to-cell and systemic spread of Tobacco mosaic virus (TMV) using dominant negative inhibition of myosin activity. We found that interference with the functions of three class VIII myosins and two class XI myosins significantly reduced the local and long-distance transport of the virus. We further determined that the inactivation of myosins XI-2 and XI-K affected the structure and dynamic behavior of the ER leading to aggregation of the viral movement protein (MP) and to a delay in the MP accumulation in plasmodesmata (PD). The inactivation of myosin XI-2 but not of myosin XI-K affected the localization pattern of the 126k replicase subunit and the level of TMV accumulation. The inhibition of myosins VIII-1, VIII-2 and VIII-B abolished MP localization to PD and caused its retention at the plasma membrane. These results suggest that class XI myosins contribute to the viral propagation and intracellular trafficking, whereas myosins VIII are specifically required for the MP targeting to and virus movement through the PD. Thus, TMV appears to recruit distinct myosins for different steps in the cell-to-cell spread of the infection. 相似文献
Buildings consume a large amount of energy for space cooling during the summer season, creating an overall sustainability concern. The upfront cost associated with sustainability repels the decision-makers to often end up adopting solutions that have huge operations and maintenance costs. Therefore, the purpose of this study is to assess the lifecycle cost (LCC) implications of optimum configurations of active and passive strategies for reducing the cooling load in buildings.
Methods
Several green building active and passive strategies and technologies were assimilated and their thermal performance in a hot semi-arid climate of Lahore in Pakistan using DesignBuilder V6.1 was simulated to obtain the most optimum cooling load configuration. Furthermore, LCC is estimated, and overall efficiency is evaluated to identify the most effective space cooling configuration.
Results and discussion
The results suggest that a configuration of EPS for external wall insulation, vertical louvers for external shading, 6 mm blue HRG (low-E soft coated)?+?12 mm air space?+?6 mm clear glass for windows, polystyrene as roof insulation, cross ventilation through windows, and LED lighting system has the best performance. This is the first-of-its-kind study in the hot semi-arid climate of South Asia with the city of Lahore in Pakistan as the test case and can be generalized for places with similar conditions. The findings will help the decision-makers in selecting the most load-efficient and cost-effective green building technologies to help improve overall sustainability.
Conclusion
The implementation of the proposed strategies not only aids in providing user-friendly and effective decision-making but also promotes the adoption of sustainability in buildings by leveraging the existing green building technologies to enhance the environmental and economic aspects. This is a promising approach to facilitate the spread of green building construction in developing countries. It is recommended to utilize the strategies grouped in Scenario 8 to achieve a reduced cooling load and LCC of a residential building throughout its lifecycle.
Hyperphosphorylation and deposition of tau into neurofibrillary tangles is a hallmark of Alzheimer disease (AD). Alternative splicing of tau exon 10 generates tau isoforms containing three or four microtubule binding repeats (3R-tau and 4R-tau), which are equally expressed in adult human brain. Dysregulation of exon 10 causes neurofibrillary degeneration. Here, we report that cyclic AMP-dependent protein kinase, PKA, phosphorylates splicing factor SRSF1, modulates its binding to tau pre-mRNA, and promotes tau exon 10 inclusion in cultured cells and in vivo in rat brain. PKA-Cα, but not PKA-Cβ, interacts with SRSF1 and elevates SRSF1-mediated tau exon 10 inclusion. In AD brain, the decreased level of PKA-Cα correlates with the increased level of 3R-tau. These findings suggest that a down-regulation of PKA dysregulates the alternative splicing of tau exon 10 and contributes to neurofibrillary degeneration in AD by causing an imbalance in 3R-tau and 4R-tau expression. 相似文献
Protein phosphatase-2A (PP2A) activity, which is compromised in Alzheimer disease brain, is regulated by two endogenous inhibitors, one of them being I(2)(PP2A), a 277 amino acid long protein also known as SET. Here we report that both the amino terminal fragment (I(2NTF); aa 1-175) and the carboxy terminal fragment (I(2CTF); aa 176-277) of I(2)(PP2A) inhibit PP2A by binding to its catalytic subunit PP2Ac and cause hyperphosphorylation of tau. The C-terminal acidic region in I(2CTF) and Val 92 in I(2NTF) are essential for their association with PP2Ac and inhibition of the phosphatase activity. 相似文献
