Fanconi anemia (FA) is a recessive chromosomal instability syndrome that is clinically characterized by multiple symptoms. Chromosome breakage hypersensitivity to alkylating agents is the gold standard test for FA diagnosis. In this study, we provide a detailed laboratory protocol for accurate assessment of FA diagnosis based on mitomycin C (MMC) test. Induced chromosomal breakage study was successful in 171 out of 205 aplastic anemia (AA) patients. According to the sensitivity of MMC at 50 ng/ml, 38 patients (22.22%) were diagnosed as affected and 132 patients (77.17%) as unaffected. Somatic mosaicism was suspected in an 11-year-old patient with a FA phenotype. Twenty-six siblings of FA patients were also evaluated and five of them (19.23%) were diagnosed as FA. From this study, a standard protocol for diagnosis of FA was developed. It is routinely used as a diagnostic test of FA in Tunisia.  相似文献

We report on the effectiveness of molecular studies regarding Fanconi anemia (FA) for a better selection of bone marrow graft donors and for post-transplant follow up. Ten unrelated FA patients and their families were analyzed by microsatellite markers. In 9 cases, the cytogenetic investigation of potential human leukocyte antigen (HLA)-identical related donors was normal, and the molecular analyses confirmed that they were also either normal or heterozygous carriers. For 1 patient, cytogenetic analysis of an HLA-identical sibling donor yielded ambiguous results with a relatively high number of chromosomal breakages using cross-linking agents. However, genotyping of this potential donor demonstrated his heterozygous state. Nine patients have received allogeneic bone marrow transplantation from HLA-matched related donors. Microsatellite analysis showed complete chimerism (CC) in all cases. The median follow up was 54 months (range 8-144 months). One patient out of 9 with CC rejected her graft without prior detection of a transitional mixed chimerism. Among these patients, 1 died 25 months after the transplantation of a chronic graft-versus-host-disease (GVHD). We conclude that, when the cytogenetic studies are not conclusive, molecular analyses are crucial to distinguish heterozygous carriers from asymptomatic FA Tunisian patients. Molecular analyses also allowed the evaluation of hematopoietic chimerism after allogeneic bone marrow transplantation and might be of value to identify patients with a high risk for graft rejection.  相似文献

Despite increasing evidence indicating the essential involvement of selenium (Se) on growth performance, antioxidant capacity, and meat quality of commercial broilers, the effects of different Se sources on local Chinese Subei chickens is unclear. A total of 360 50-day-old male chickens were individually weighed and randomly allocated to four treatment groups. Chickens in each of the four groups were fed diets supplemented with 0.3 mg Se/kg as sodium Se (SS), Se-enriched yeast (SY), selenomethionine (Met-Se), or nano red element Se (Nano-Se) for 40 days. At the end of the experiment, one bird of approximately average weight from each cage was selected and slaughtered, and blood and breast muscles samples were collected. The results showed that there was no significant difference in feed intake, body weight gain, or feed to gain ratio among treatments (P > 0.05). Dietary SY, Met-Se, and Nano-Se supplementation increased the activity of glutathione peroxidase in serum and breast muscles and decreased the concentration of malondialdehyde in serum and carbonyl in breast muscles compared with the SS group (P < 0.05). Moreover, SY, Met-Se, and Nano-Se supplementation increased pH45min, total protein solubility, and myofibrillar protein solubility, as well as decreased the shear force value compared with the SS group (P < 0.05). In addition, birds in the SY and Met-Se groups exhibited lower cooking loss compared with the SS group (P < 0.05). In conclusion, organic Se and Nano-Se supplementation resulted in an improvement of antioxidant capacity and meat quality in local Chinese Subei chickens relative to inorganic Se.  相似文献

This study investigated neurotoxicity of chronic fluorosis in the rat hippocampus. Newly weaning, male, Sprague-Dawley (SD) rats were administered 15, 30, and 60 mg/L sodium fluoride (NaF) solution (fluorine ion concentration 8.25, 16.50, and 33.00 mg/L, respectively), and tap water, for 18 months. The neurotoxicological mechanism was examined with a focus on intracellular calcium overload. Results showed that as the fluoride concentration increased, calcium ion concentration [Ca2+], the expression of calcium/calmodulin-dependent protein kinase II α (CaMKIIα), and the expression of catus proto-oncogene protein c-fos (c-fos) all tend to increase. Compared to the control group, Ca2+, CaMKIIα, and c-fos significantly increased (P < 0.05) in the moderate-fluoride and the high-fluoride groups. These results indicate that Ca2+/CaMKIIα/c-fos channel signal may be the molecular mechanism of central nervous system damage caused by chronic fluoride intoxication. Moreover, elevated Ca2+ concentration in the hippocampus may be the initiating factor of neuronal apoptosis induced by fluoride.  相似文献

In Tunisia, Tipuana tipu (Benth .) Kuntze is an exotic tree, which was introduced many years ago and planted as ornamental street, garden, and park tree. The present work reported, for the first time, the chemical composition and evaluates the allelopathic effect of the hydrodistilled essential oils of the different parts of this tree, viz., roots, stems, leaves, flowers, and pods gathered in the area of Sousse, a coastal region, in the East of Tunisia. In total, 86 compounds representing 89.9 – 94.9% of the whole oil composition, were identified in these oils by GC‐FID and GC/MS analyses. The root essential oil was clearly distinguished for its high content in sesquiterpene hydrocarbons (β‐caryophyllene, 1 (44); 24.1% and germacrene D, 2 (53); 20.0%), while those obtained from pods, leaves, stems, and flowers were dominated by non‐terpene hydrocarbons. The most important ones were n‐tetradecane (41, 16.3%, pod oil), 1,7‐dimethylnaphthalene (43, 15.6%, leaf oil), and n‐octadecane (77, 13.1%, stem oil). The leaf oil was rich in the apocarotene (E)‐β‐ionone ( 4 (54); 33.8%), and the oil obtained from flowers was characterized by hexahydrofarnesylacetone ( 5 (81); 19.9%) and methyl hexadecanoate (83, 10.2%). Principal component and hierarchical cluster analyses separated the five essential oils into three groups and two subgroups, each characterized by the major oil constituents. Contact tests showed that the germination of lettuce seeds was totally inhibited by the root essential oil tested at 1 mg/ml. The inhibitory effect on the shoot and root elongation varied from ?1.6% to ?32.4%, and from ?2.5% to ?64.4%, respectively.  相似文献

The effects of light intensity and temperature on Arthrospira platensis growth and production of extracellular polymeric substances (EPS) in batch culture were evaluated using a three-level, full-factorial design and response surface methodology. Three levels were tested for each parameter (temperature: 30, 35, 40°C; light intensity: 50, 115, 180 μmol photons m−2 s−1). Both growth and EPS production are influenced mainly by the temperature factor but the interaction term temperature*light intensity also had a significant effect. In addition, conditions optimising EPS production are different from those optimising growth. The highest growth rate (0.414 ± 0.003 day−1) was found at the lowest temperature (30°C) and highest light intensity (180 μmol photons m−2 s−1) tested, no optima were detectable within the given test range. Obviously, optima for growth must be at a temperature lower than 30°C and a light intensity higher than 180 μmol photons m−2 s−1. For EPS production, light intensity had a positive linear effect (optimum obviously higher than 180 μmol photons m−2 s−1), but for the temperature parameter a maximum effect was detectable at 35°C.  相似文献

Distributed systems provide geographically distributed resources for large-scale applications while managing large volumes of data. In this context, replication of data in several sites of the system is an effective solution for achieving interesting performances. A number of data replication strategies have been proposed in the literature. Data popularity is one of the most important parameters taken into consideration by these strategies. It analyzes the historic of the data access pattern, and provides predictions for future data requests. However, measuring data popularity is a challenging task because there are several factors that contribute to the evaluation of data popularity. In this paper, a new adaptive measurement for data popularity in distributed systems is proposed. The proposed measurement covers all factors taken into consideration by previous work of the literature. It also takes into consideration new factors to deal with the dynamic nature of the system so it can adapt to any access pattern. We show that the exploitation of our measurement improves the performances of replication strategies, while offering the possibility to use the data popularity parameter in new contexts in replication management.  相似文献

Background and aimsIn situations of oxidative stress, l-glutamine (Gln) exhibits protective effects which may be potentiated by its combination with antioxidants. The aim of this study was to compare the effects of a Gln-antioxidants formula vs Gln alone in intestinal epithelial cell lines Caco-2 and HCT-8 submitted to hydrogen peroxide-induced oxidative injury.MethodsCells were cultured during 24 h with 0, 1, 2, 4, 8 or 16 mM Gln or isomolar Gln combined to cysteine, vitamine C, vitamine E, β-carotene, Zn and Se (Gln-AOX). After 24 h, membrane integrity was assessed by means of LDH leakage, the level of oxidative stress by analysis of 8-isoprostane concentration and cell viability by colorimetric-MTT assay.ResultsLDH activity decreased (P < 0,05) with a dose-dependent manner in both cell types with Gln-AOX whereas Gln decreased LDH release only in the Caco-2 line at 1 mM. For both cell lines, release of 8-isoprostane was not blunted by any treatment and increased paradoxically with 16 mM Gln (P < 0.05). Cell viability was higher (P < 0.05) using Gln-AOX vs Gln at 4, 8 and 16 mM in both cell lines.ConclusionsThese results suggest that Gln-AOX is more efficient than Gln alone to preserve cell membrane integrity and viability in intestinal cell lines submitted to oxidative injury.  相似文献

Anguillicola crassus, parasite nematode of the European eel Anguilla anguilla, was recorded for the first time in Tunisia (1999) in the Ichkeul lagoon. Its distribution has since spread toward Bizerte and Ghar El Melh lagoons. The monthly epidemiological survey reveals that A. crassus exists throughout the year in the Ichkeul lagoon. In this lagoon, its prevalence is low in winter (12% in December), with a marked increases in the spring reaching a maximum in March (35%), before it starts to decrease in summer with a minimum in July (4.35%), which in turn is followed by a pronounced new rise in autumn (30% in November). However, mean intensity values do not show such a marked variation. The majority of the values are between 1 and 1.5 parasites per host. In the Bizerte and Ghar El Melh lagoons, the presence of this nematode is limited only to one to three months. Investigations in the Tunis lagoon did not reveal until now the presence of A. crassus. It has been observed that the length of the eel influences the prevalence values: A. crassus becomes less common if the length of the eel increases. Comparatively with the global epidemiological values of A. crassus signalled subsequently (1999) in the Ichkeul lagoon, we note that the present values record a clean increase.  相似文献
