Yang et al. (14) have isolated from mammalian brain an octapeptide FLFQPQRF-NH2 (F-8-F-NH2) with certain antiopiate properties. Third ventricular injection of 2 micrograms of this peptide together with the aminopeptidase inhibitor bestatin precipitated an opiate-withdrawal syndrome in morphine-dependent but not in nondependent rats. Third ventricular injection in nondependent rats of 15 micrograms of the peptide together with bestatin induced a morphine-withdrawal-like behavioral syndrome. This syndrome was not produced by injection of bestatin or saline vehicle alone and was preventable by injection of 3.5 mg/kg morphine sulphate SC.  相似文献

Epidermal growth factor, a potent mitrogen for granulosa cells produced a three-fold stimulation of ornithine decarboxylase activity in porcine granulose cells in vitro. Fibroblast growth factor, another compound with mitogenic activity for granulose cells, did not stimulate ornithine decarboxylase. Maximally effective concentrations of a commercial preparation of bovine serum albumin equalled the maximal effect of epidermal growth factor on this enzyme activity. The dominant stimulator(s) in the albumin preparation eluted after bovine serum albumin in gel filtration. At maximally effective concentrations, luteinizing hormone produced substantially greater stimulation than either epidermal growth factor or the bovine albumin preparation. Combinations of saturating doses of any two of these stimulators produced additive effects on enzyme activity.  相似文献

Purified guinea pig basophils, or basophils either specifically degranulated with antigen or nonspecifically degranulated with lectin, were cultured with guinea pig line 1 hepatoma cells for 1 to 24 hr and studied ultrastructurally. As early as 1 hr of culture, degranulated or nongranulated basophils and tumor cells formed close contacts by mutually intertwined elongated cell processes and also in cultures containing degranulated basophils, extruded membrane-free basophil cytoplasmic granules became firmly attached to tumor cells. At later intervals, some tumor cells cultured with basophils exhibited cytostatic and cytopathic changes, including dense mitochondria, centralization of organelles, dilated perinuclear and rough endoplasmic cisternae, cell swelling and cytoplasmic lucency, disrupted cytoplasmic organelle and plasma membranes, nuclear pyknosis and fragmentation. Some tumor cell specialized surface attachments were either disrupted or damaged at points of basophil or basophil granule adhesion. Tumor damage was most extensive in cultures containing degranulated basophils, although only a minority of tumor cells (less than 10%) was affected. Tumor injury was seen much less frequently in the presence of nondegranulated basophils, and was absent in control cultures of tumor alone. The occasional viable tumor cells that phagocytosed basophil granules were apparently unharmed, suggesting that internalization of basophil granules by tumor cells is not cytotoxic.  相似文献

Species loss caused by anthropogenic disturbance threatens forest ecosystems globally. Until 50 years ago, the major sources of boreal forest disturbance in western Canada were a combination of forest wild fire events, pest insect outbreaks, and forest timber harvesting. However, in the 1960s, when the oil boom started in Alberta, oil and gas development along with oil sands mining quickly became another major forest disturbance agent. In this case study we report the effects of operational oil sands mine reclamation on terrestrial arthropod communities and compare them with nearby burned and mature forest sites as a way to provide a benchmark from which to understand the long-term trajectory of recovery for these groups. During the summer of 2016 over 6700 epigaeic beetles were collected using pitfall traps. A total of 43 species of ground beetles and 118 species of rove beetles were collected. Epigaeic beetle assemblages differed between the reclaimed, burned, and mature forest sites. Partitioning of beta diversity in the reclaimed, burned areas and mature forests indicated that species turnover formed the largest component of diversity. Species richness patterns were similar among sites; however, cluster analysis indicated that epigaeic beetle assemblages were only 20% similar between the reclaimed and natural sites. Although ground beetles of the reclaimed area showed positive spatial autocorrelation among treatments, both ground and rove beetles showed responses to the reclamation treatments. The reclaimed areas were dominated by small- to medium-sized open-habitat eurytopic species, whereas the fire and mature forest sites were dominated by larger forest species. The reclaimed area of this case study constitutes a novel, reconstructed ecosystem that is clearly not equivalent in species assemblage to burnt stands of similar age or to mature forest stands.  相似文献

Pulmonary amine extraction (E) was measured by triple-indicator dilution techniques from bolus injections of trace amounts of 5-hydroxy[14C]tryptamine ([14C]HT)m [3H]norepinephrine ([3H]NE), indocyanine green dye before and after glass-bead embolization in 23 anesthetized dogs. Control E(5-[14C]-HT) was 89.7 +/- 1.7%; 10 min after embolization (which approximately doubled pulmonary artery pressure and pulmonary vascular resistance), E(5-[14C]HT) was significantly reduced to 65.9 +/- 3.0% (kappa +/- SE; n = 10) (P less than 0.01). Control E([3H]NE) (40.1 +/- 4.5%) was unaffected by embolization. Imipramine (8 mg/kg) depressed control E(5-[14C]HT) to 38.7 +/- 1.5% and control E([3H]NE)d to 35.0 +/- 3.9% (P less than 0.05; n = 4). In these animals, pulmonary hemodynamic changes secondary to embolization were comparable to those in non-drug-treated dogs, but E(5-[14C]HT) and E([3H]NE) were not further depressed. Progressive pulmonary lobar artery ligation (n = 5) did not affect amine extraction until perfusion was limited to one lobe. The selectivity of the effect of embolization on E(5-[14C]HT), the lack of an effect on imipramine-insensitive E(5-[14C]HT) extraction, and the much smaller changes after progressive lobar ligation indicate that, although derecruitment of vascular surface area secondary to mechanical obstruction may contribute to postembolization depression of E(5-[14C]HT), additional mechanisms such as local saturation of 5-HT uptake or selective damage to endothelial cell transport of 5-HT may underlie these observations.  相似文献

The amino terminal sequence of the spinach (Spinacia oleracea L. cv Bloomsdale Long Standing) leaf cytoplasmic phosphorylase was determined and shown to have little similarity to the known sequence of the potato tuber phosphorylase. The antigenic reaction of spinach chloroplast phosphorylase and rabbit muscle phosphorylase a to antiserum prepared against spinach leaf cytoplasmic phosphorylase was tested. Neither phosphorylase gave a positive reaction when tested by immunodiffusion or neutralization of enzyme activity. The two spinach phosphorylases were assayed throughout the growth of the plant. Activity of cytoplasmic phosphorylase increased 4- to 8-fold at 30 to 35 days from sowing. Enzyme protein levels, as measured by antibody neutralization, increased by a similar amount. There was no corresponding increase in chloroplast phosphorylase activity. The chloroplast phosphorylase varied in parallel with the chloroplast enzyme ADPglucose pyrophosphorylase. Starch levels were high during the earlier stages of growth and then fell to a constant low level just before the increase in cytoplasmic phosphorylase. The results are discussed with respect to the relationship and functions of the two phosphorylases.  相似文献

The 1980-81 epidemic of influenza A/Bangkok 79 was responsible for increased absenteeism (1.7 times the rate for the corresponding period of the subsequent nonepidemic year) among selected hospital staff in Winnipeg''s Health Sciences Centre. Retrospective study of employment records for 25 of the centre''s largest departments showed excess sick-leave costs of about $24 500 during the 2-week period of peak absenteeism that included the epidemic. Although the centre was sampling prospectively for the virus the first positive results became available too late for chemoprophylactic measures to have been effective. The greater increase in absenteeism among nursing staff caring for patients with chronic respiratory disease and nurses working on general medical or pediatric acute infection/isolation wards suggested that these groups be targeted for influenza vaccination in hospitals.  相似文献

We examined the role of Ca(2+) in the control of basal and hormone-stimulated ornithine decarboxylase activity in isolated pig granulosa cells maintained under chemically defined conditions in vitro. Omission of Ca(2+) from the incubation medium (measured Ca(2+) concentration 5mum) decreased basal enzymic activity, and significantly (P<0.01) impaired the response to maximally stimulating doses of either lutropin or follitropin. No significant alteration occurred in the concentration of either gonadotropin required to elicit half-maximal effects. The addition of EGTA (1.27-2.0mm) to chelate residual extracellular Ca(2+) further decreased hormone-induced rises in ornithine decarboxylase activity. Despite the presence of 1.27mm concentrations of extracellular Ca(2+), the administration of presumptive Ca(2+) antagonists, believed to impair trans-membrane Ca(2+) influx [verapamil (10-100mum), nifedipine (1-100mum) or CoCl(2) (1mm)] suppressed hormone-stimulated ornithine decarboxylase activity. The inhibitory effects of verapamil or of Ca(2+) omission from the medium were not overcome by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (0.25mm), or by cholera toxin, or by an exogenously supplied cyclic AMP analogue, 8-bromo cyclic AMP. Conversely, micromolar concentrations of a putative bivalent-cation ionophore, A23187, increased significantly the stimulation of ornithine decarboxylase activity by saturating concentrations of lutropin or 8-bromo cyclic AMP. Thus the present observations implicate Ca(2+) ions in the modulation of hormone action and cellular function in normal ovarian cells.  相似文献
