The inhibition of the caspase-3 enzyme is reported to increase neuronal cell survival following cerebral ischemia. The peptide Z-DEVD-FMK is a specific caspase inhibitor, which significantly reduces vulnerability to the neuronal cell death. However, this molecule is unable to cross the blood-brain barrier (BBB) and to diffuse into the brain tissue. Thus, the development of an effective delivery system is needed to provide sufficient drug concentration into the brain to prevent cell death. Using the avidin (SA)-biotin (BIO) technology, we describe here the design of chitosan (CS) nanospheres conjugated with poly(ethylene glycol) (PEG) bearing the OX26 monoclonal antibody whose affinity for the transferrin receptor (TfR) may trigger receptor-mediated transport across the BBB. These functionalized CS-PEG-BIO-SA/OX26 nanoparticles (NPs) were characterized for their particle size, zeta potential, drug loading capacity, and release properties. Fluorescently labeled CS-PEG-BIO-SA/OX26 nanoparticles were administered systemically to mice in order to evaluate their efficacy for brain translocation. The results showed that an important amount of nanoparticles were located in the brain, outside of the intravascular compartment. These findings, which were also confirmed by electron microscopic examination of the brain tissue indicate that this novel targeted nanoparticulate drug delivery system was able to translocate into the brain tissue after iv administration. Age-related macular degeneration (AMD) is the leading cause of blindness in the elderly. We report an association between an Alu polymorphism in the angiotensin-converting enzyme (ACE) gene with the dry/atrophic form of AMD. Using the polymerase chain reaction (PCR) on genomic DNA isolated from patients with AMD (n=173), and an age-matched control population (n=189), we amplified a region polymorphic for an Alu element insertion in the ACE gene. The Alu(+/+) genotype occurred 4.5 times more frequently in the control population than the dry/atrophic AMD patient population, (p=0.004). The predominance of the Alu(+/+) genotype within the unaffected control group represents a protective insertion with respect to the human ocular disease, dry/atrophic AMD. This is the first demonstration of an Alu element insertion exerting protective effects against a known human disease. 相似文献

Anaerobic co-digestion of fruit and vegetable waste (FVW) and abattoir wastewater (AW) was investigated using anaerobic sequencing
batch reactors (ASBRs). The effects of hydraulic retention time (HRT) and temperature variations on digesters performances
were examined. At both 20 and 10 days biogas production for co-digestion was greater thanks to the improved balance of nutrients.
The high specific gas productions for the different digestion processes were 0.56, 0.61 and 0.85 l g−1 total volatile solids (TVS) removal for digesters treating AW, FVW and AW + FVW, respectively. At an HRT of 20 days, biogas
production rates from thermophilic digesters were higher on average than from mesophilic AW, FVW and AW + FVW digestion by
28.5, 44.5 and 25%, respectively. However, at 10 days of HRT results showed a decrease of biogas production rate for AW and

Biofilms are the most common mode of bacterial growth in nature. Their formation occurs on organic or inorganic solid surfaces in contact with a liquid, on gas-liquid and liquid-liquid boundaries as well. The aims of this study were, by combining cell enumeration, scanning electron microscopy and denaturing gel gradient electrophoresis (DGGE), to characterize the structural dynamics of dairy biofilm growth in the environments with a nutrient flow, and to evaluate the impact of adhesion of Lactococcus lactis on the biofilm community depending on the incubation time. Significantly higher values of biofilm volume and thickness were observed under dynamic conditions after 55 h. The populations of gram-positive bacteria and fungi exhibited a significantly higher biofilm organization after 2 days of cultivation than that of gram-negative bacteria. Also, results showed that Lc. lactis was able to adhere to silicone surface and the produced biofilm in which the number of adhered gram-positive and gram-negative bacteria decreased by nine orders of magnitude after 48 h of contact. This study constitutes a step ahead in developing the strategies to prevent microbial colonization by lactococcal protective biofilm. 相似文献

Myrtus communis is a typical plant of the Mediterranean area, which is mainly used as animal and human food and, in folk medicine, for treating some disorders. In the present study, we evaluated in vitro antibacterial and antifungal properties of the essential oils of Myrtus communis (McEO), as well as its phytochemical composition. The GC/MS analysis of the essential oil revealed 17 compounds. Myrtenyl acetate (20.75%), 1,8‐cineol (16.55%), α‐pinene (15.59%), linalool (13.30%), limonene (8.94%), linalyl acetate (3.67%), geranyl acetate (2.99%), and α‐terpineol (2.88%) were the major components. The antimicrobial activity of the essential oil was also investigated on several microorganisms. The inhibition zones and minimal inhibitory concentration (MIC) values of bacterial strains were in the range of 16–28 mm and 0.078–2.5 mg/ml, respectively. The inhibitory activity of the McEO against Gram‐positive bacteria was significantly higher than against Gram‐negative. It also exhibited remarkable activity against several fungal strains. The investigation of the mode of action of the McEO by the time‐kill curve against Listeria monocytogenes (food isolate) showed a drastic bactericidal effect after 5 min using a concentration of 312 μg/ml. These results evidence that the McEO possesses antimicrobial properties, and it is, therefore, a potential source for active ingredients for food and pharmaceutical industries. 相似文献

Toto bodies are eosinophilic structures that resemble the cells of the superficial cell layer of the oral epithelium. Toto bodies commonly are associated with inflammatory gingival and other mucosal lesions including pyogenic granuloma, irritational fibroma, epulis fissuratum, peripheral giant cell granuloma and inflammatory hyperplastic gingivitis. We evaluated staining characteristics of Toto bodies to establish their origin and to identify their significance in lesions. We investigated pyogenic granuloma, fibroma and leukoplakia with epithelium that exhibited Toto bodies after hematoxylin and eosin (staining. Sections were stained with Alcian blue, periodic acid-Schiff and Ayoub-Shklar stains to evaluate staining intensity and distribution. More Toto bodies were found in pyogenic granuloma than in fibroma and leukoplakia. PAS and Alcian blue staining exhibited mild intensity and did not establish the origin of Toto bodies. High staining intensity and diffuse distribution of stain was observed using Ayoub-Shklar staining, which indicated that Toto bodies originate from keratin. 相似文献

The confirmed case fatality rate for the coronavirus disease 2019 (COVID-19) in Ghana has dropped from a peak of 2% in March to be consistently below 1% since May 2020. Globally, case fatality rates have been linked to the strains/clades of circulating severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) within a specific country. Here we present 46 whole genomes of SARS-CoV-2 circulating in Ghana, from two separate sequencing batches: 15 isolates from the early epidemic (March 12–April 1 2020) and 31 from later time-points ( 25–27 May 2020). Sequencing was carried out on an Illumina MiSeq system following an amplicon-based enrichment for SARS-CoV-2 cDNA. After genome assembly and quality control processes, phylogenetic analysis showed that the first batch of 15 genomes clustered into five clades: 19A, 19B, 20A, 20B, and 20C, whereas the second batch of 31 genomes clustered to only three clades 19B, 20A, and 20B. The imported cases (6/46) mapped to circulating viruses in their countries of origin, namely, India, Hungary, Norway, the United Kingdom, and the United States of America. All genomes mapped to the original Wuhan strain with high similarity (99.5–99.8%). All imported strains mapped to the European superclade A, whereas 5/9 locally infected individuals harbored the B4 clade, from the East Asian superclade B. Ghana appears to have 19B and 20B as the two largest circulating clades based on our sequence analyses. In line with global reports, the D614G linked viruses seem to be predominating. Comparison of Ghanaian SARS-CoV-2 genomes with global genomes indicates that Ghanaian strains have not diverged significantly from circulating strains commonly imported into Africa. The low level of diversity in our genomes may indicate lower levels of transmission, even for D614G viruses, which is consistent with the relatively low levels of infection reported in Ghana. 相似文献
