Decapod crustaceans exhibit considerable variation in fertilisation strategies, ranging from pervasive single paternity to the near-ubiquitous presence of multiple paternity, and such knowledge of mating systems and behaviour are required for the informed management of commercially-exploited marine fisheries. We used genetic markers to assess the paternity of individual broods in the European lobster, Homarus gammarus, a species for which paternity structure is unknown. Using 13 multiplexed microsatellite loci, three of which are newly described in this study, we genotyped 10 eggs from each of 34 females collected from an Atlantic peninsula in the south-western United Kingdom. Single reconstructed paternal genotypes explained all observed progeny genotypes in each of the 34 egg clutches, and each clutch was fertilised by a different male. Simulations indicated that the probability of detecting multiple paternity was in excess of 95% if secondary sires account for at least a quarter of the brood, and in excess of 99% where additional sire success was approximately equal. Our results show that multiple paternal fertilisations are either absent, unusual, or highly skewed in favour of a single male among H. gammarus in this area. Potential mechanisms upholding single paternal fertilisation are discussed, along with the prospective utility of parentage assignments in evaluations of hatchery stocking and other fishery conservation approaches in light of this finding.  相似文献

Background

Human cytomegalovirus (HCMV) uracil DNA glycosylase, UL114, is required for efficient viral DNA replication. Presumably, UL114 functions as a structural partner to other factors of the DNA-replication machinery and not as a DNA repair protein. UL114 binds UL44 (HCMV processivity factor) and UL54 (HCMV-DNA-polymerase). In the present study we have searched for cellular partners of UL114.

Methodology/Principal Findings

In a yeast two-hybrid screen SMARCB1, a factor of the SWI/SNF chromatin remodeling complex, was found to be an interacting partner of UL114. This interaction was confirmed in vitro by co-immunoprecipitation and pull-down. Immunofluorescence microscopy revealed that SMARCB1 along with BRG-1, BAF170 and BAF155, which are the core SWI/SNF components required for efficient chromatin remodeling, were present in virus replication foci 24–48 hours post infection (hpi). Furthermore a direct interaction was also demonstrated for SMARCB1 and UL44.

Conclusions/Significance

The core SWI/SNF factors required for efficient chromatin remodeling are present in the HCMV replication foci throughout infection. The proteins UL44 and UL114 interact with SMARCB1 and may participate in the recruitment of the SWI/SNF complex to the chromatinized virus DNA. Thus, the presence of the SWI/SNF chromatin remodeling complex in replication foci and its association with UL114 and with UL44 might imply its involvement in different DNA transactions.  相似文献

The relationship between physiology and temperature has a large influence on population-level responses to climate change. In natural settings, direct thermal effects on metabolism may be exaggerated or offset by behavioural responses influencing individual energy balance. Drawing on a newly developed proxy, we provide the first estimates of the thermal performance curve of field metabolism in a wild fish. We investigate the thermal sensitivity of field metabolic rate in two sympatric, genetically distinct ecotypes of Atlantic cod from the Skagerrak coast of southern Norway. The combined ecotype median of field metabolic rate increased with increasing temperature until around 16°C, coincident with the thermal optimum for growth for juvenile Atlantic cod. Individual cod experienced temperatures in excess of the thermal optimum for field metabolic rate, indicating some degree of thermal limitation of field metabolism in a complex natural environment with the potential for thermal refugia. The two cod ecotypes showed different thermal performance curves for field metabolic rate, revealing that genetic components to temperature sensitivity persist beyond acclimation effects. The cold-adapted fjord ecotype maintained higher field metabolic rates at cooler temperatures than the warm-adapted North Sea ecotype, which showed clear preference for warmer waters around the thermal optimum. Field metabolic rates of the two ecotypes were strongly influenced by year and location of sampling, implying more complex behavioural responses to environmental conditions. We emphasise that the energy uses reflecting physiological conditions in the field should be considered in the evaluation of the effect of climatic variables on fish population dynamics and demonstrate that otolith isotopes provide an analytical framework to answer this question.  相似文献

Within the last two decades, ecological stoichiometry (ES) and nutritional geometry (NG, also known as geometric framework for nutrition) have delivered novel insights into core questions of nutritional ecology. These two nutritionally explicit frameworks differ in the ‘nutrient currency’ used and the focus of their past research; behavioural feeding strategies in NG, mainly investigating terrestrial organisms, and trophic ecology in ES, mainly in aquatic settings. However, both NG and ES have developed in explaining patterns across various scales of biological organization. Integrating specific tools of these frameworks could advance the field of nutritional ecology by unifying theoretical and empirical approaches from the organismal to ecosystem level processes. Toward this integration, we identified 1) nutrient/element budgets as a shared concept of both frameworks that encompass nutrient intake, retention, and release, 2) response surface plots of NG as powerful tools to illustrate processes at the organismal level and 3) the concept of consumer‐driven nutrient recycling (CNR) of ES as a useful tool bridging organism and ecosystem scales. We applied response surface plots to element budget data from an ES study to show how this approach can deliver new insights at the organismal level, e.g. by showing the interplay between egestion and excretion depending simultaneously on the consumed amount of carbon and phosphorus based on variation across individuals. By integrating concepts of ES and NG to model microbial uptake and mineralization of nitrogenous wastes reported in a NG study, we also demonstrate that considering biochemically explicit mineralization rates of organic wastes can improve predictions of CNR by reducing over‐ or underestimation of mineralization depending on the quality of the consumer's diet. Our presented tools and approaches can help to bridge the organismal and ecosystem level, advancing the predictive power of studies in nutritional ecology at multiple ecological scales.  相似文献

Two membrane fractions prepared from the Ehrlich ascites-tumor cell show non-identical stimulatory responses to certain amino acids in their Mg2+-dependent activity to cleave ATP, despite the presence of ouabain and the absence of Na+ or K+. The first of these, previously described, shows little (Na+ + K+)-ATPase activity, and is characteristically stimulated by the presence of certain diamino acids with low pK2, and at pH values suggesting that the cationic forms of these amino acids are effective. The evidence indicates that these effects are not obtained through occupation of the kinetically discernible receptor site serving characteristically for the uphill transport of these amino acids into the Ehrlich cell. The second membrane preparation was purified with the goal of concentrating the (Na+ + K+)-ATPase activity. It also is stimulated by the model diamino acid, 4-amino-1-methylpiperidine-4-carboxylic acid, and several ordinary amino acids. The diamino acids were most effective at pH values where the neutral zwitterionic forms might be responsible. Among the optically active amino acids tested, the effects of ornithine and leucine were substantially stronger for the l than for d isomers. The list of stimulatory amino acids again corresponds poorly to any single transport system, although the possibility was not excluded that stimulation might occur for both preparations by occupation of a membrane site which ordinarily is kinetically silent in the transport sequence. The high sensitivity to deoxycholate and to dicyclohexylcarbodiimide of the hydrolytic activity produced by the presence of l-ornithine and 4-amino-1-methyl-piperidine-4-carboxylic acid suggests that the stimulatory effect is not merely a general intensification of the background Mg+-dependent hydrolytic activity.  相似文献

Background

To achieve a child mortality reduction according to millennium development goal 4, it is necessary to considerably reduce neonatal mortality. We report stillbirth and early neonatal mortality risks as well as determinants of perinatal mortality in Eastern Uganda.

Methods

A community-based prospective cohort study was conducted between 2006 and 2008. A total of 835 pregnant women were followed up for pregnancy outcome and survival of their children until 7 days after delivery. Mother''s residence, age, parity, bed net use and whether delivery took place at home were included in multivariable regression analyses to identify risk factors for perinatal death.

Results

The stillbirth risk was 19 per 1,000 pregnancies and the early neonatal death risk 22 per 1,000 live births. Overall, the perinatal mortality risk was 41 [95%CI: 27, 54] per 1,000 pregnancies. Of the deaths, 47% followed complicated deliveries and 24% preterm births. Perinatal mortality was 63/1,000 pregnancies among teenage mothers, 76/1,000 pregnancies among nulliparous women and 61/1,000 pregnancies among women delivering at home who, after controlling for potential confounders, had a 3.7 (95%CI: 1.8, 7.4) times higher perinatal mortality than women who gave birth in a health facility. This association was considerably stronger among nulliparous women [RR 8.0 (95%CI: 2.9, 21.6)] than among women with a previous live birth [RR 1.8 (95%CI: 0.7, 4.5)]. All perinatal deaths occurred among women who did not sleep under a mosquito net. Women living in urban slums had a higher risk of losing their babies than those in rural areas [RR: 2.7 (95%CI: 1.4, 5.3)].

Conclusion

Our findings strengthen arguments for ensuring that pregnant women have access to and use adequate delivery facilities and bed nets.  相似文献

In vivo 15N and 14N nuclear magnetic resonance spectroscopy was used to investigate the assimilation of nitrate and ammonium in seedlings of Norway spruce (Picea abies [L.] Karst.). The main objective was to study accumulation of free NH+4 and examine to what extent the nitrogen source affects the composition of the free amino acid pools in roots, stems and needles. NH+4 concentrations in plants growing in the presence of 0.5–50 mM ammonium were quantified using 14N NMR. The NH+4 values in tissues ranged from 6 to 46 μmol (g fresh weight)?1. with highest concentrations in roots and needles. The tissue NH+4 peaked at 5.0 mM NH+4 in the medium. and failed to increase when NH+4 in the medium was increased to 50 mM, indicating metabolic control of the concentration of this cation in tissues. The 14N NMR spectra were used to estimate pH of the NH+4 storage pools. Based on the pH sensitivity of the quintet of 14NH+4 resonance, we suggest that the pH of the ammonium storage compartments in the roots and stems should be 3.7–3.8, and in needles 3.4–3.5, representing extremely low pH values of the tissue. 15N from nitrate or ammonium was first incorporated into the amide group of glutamine and then into α-amino groups, confirming that the glutamine synthetase/ glutamate synthase cycle is the major route of nitrogen assimilation into amino acids and thus plays a role in lowering the levels of NH+4 in the cytoplasm. NH+4 can also be assimilated in roots in plants growing in darkness. The main 15N-labelled amino acids were glutamine. arginine and alanine. Almost no 15N signals from needles were observed. Double labelling (δN + w, wN) of arginine is consistent with the operation of the ornithine cycle, and enrichment indicates that this cycle is a major sink of newly assimilated nitrogen. Nitrogen assimilation in roots in the presence of added methionine sulphoximine and glutamate indicated the catabolic action of glutamate dehydrogenase. The 15N NMR spectra of plants grown on 15N-urea showed a marked increase in the labelling of ammonium and glutamine. indicating high urease activity. Amino acids were also quantified using high pressure liquid chromatography. Arginine was found to be an important transport form of nitrogen in the stem.  相似文献

Wrasse (Labridae) fisheries have increased markedly in Norway since 2010. Wrasse are being used as cleaner fish in salmonid aquaculture to control sea-lice infestations. However, fundamental knowledge on the demography and abundance of the targeted wrasse populations in Norwegian waters is lacking, and the consequences of harvesting at the current intensity have not been assessed. Here, we compared catch per unit effort (CPUE), size, age and sex ratio of goldsinny wrasse (Ctenolabrus rupestris) and corkwing wrasse (Symphodus melops) between marine protected areas (MPAs) and control areas open for fishing at four localities on the Skagerrak coast in Southern Norway. The CPUE of goldsinny larger than the minimum size limit was 33–65% higher within MPAs, while for corkwing three of four MPAs had higher CPUE with the relative difference between MPAs and control areas ranging from ?16% to 92%. Moreover, corkwing, but not goldsinny, was significantly older and larger within MPAs than in control areas. Sex ratios did not differ between MPAs and control areas for either species. Our study suggests that despite its short history, the wrasse fisheries have considerable impacts on the target populations and, further, that small MPAs hold promise as a management tool for maintaining natural population sizes and size structure. Goldsinny, being a smaller-sized species, also seems to benefit from the traditional minimum size limit management tool, which applies outside MPAs.  相似文献
