A histochemical study of adenosine triphosphatase and other nucleotide phosphatases in young root tips

Summary The distribution of ATP-ase and other nucleotide phosphatases has been studied in young root tips of maize, barley and broad bean using frozen and paraffin sections stained by standard lead sulphide precipitation procedures. High ATP-ase activity was found at the root and cell surface which is in agreement with previous biochemical studies using excised roots and cell wall preparations. Staining was also found in the nuclei and at particulate sites in the cytoplasm. Differences were observed between the present work and the staining pattern obtained for -glycerophosphatase, and between ATP-ase staining in the three roots studied. These results are discussed in relation to the possible physiological activity of the enzymes and to the differences found between earlier histochemical studies of ATP-ase activity.Abbreviations F.L. frozen longitudinal - F.T. frozen transverse     

POLYCYTHEMIA VERA—Peripheral Vascular Manifestations

The presenting manifestations of polycythemia vera are often complications involving the vascular system. These include myocardial infarction, cerebro-vascular accidents and ischemic changes in the extremities.The concept of increased atherogenesis in cases of polycythemia vera has been questioned. A possible mechanism by which small, otherwise subclinical atheromatous plaques produce ischemic symptoms in patients with polycythemia vera is discussed. The blood in polycythemic patients has been shown to have an increased viscosity resulting in a prolonged circulation time. If a small atheromatous plaque is present in association with increased blood viscosity, this combination may well produce ischemic symptoms. This explains why treatment of polycythemia vera, with restoration of blood to normal viscosity, often reverses the patient''s ischemic symptoms.Two cases of polycythemia vera here reported, in which the presenting manifestations were gangrenous extremities, emphasize the need for prompt diagnosis and treatment of polycythemia vera. In the first case, early recognition and treatment of polycythemia vera successfully reversed the ischemic changes in the extremities, while failure of early recognition and treatment in the second case resulted in two major amputations.     

ADVANCED NEOPLASTIC DISEASE—Treatment with 5-Fluorouracil and Irradiation

One hundred and twelve patients with far advanced, inoperable neoplastic disease were treated by a method utilizing the simultaneous administration of the pyrimidine antagonist, 5-fluorouracil, and ionizing irradiation to an estimated tumor dose of 2,000 roentgen units. Seventy-seven of them had periods of objective regression of tumor of three months or more.The data presented suggest that either there may be an additive effect when the two modes of therapy are used simultaneously or one mode of therapy may potentiate the antitumor effect of the other.     

Clostridium perfringens in Meat and Meat Products

A total of 262 specimens of meat and meat dishes were examined for the presence of Clostridium perfringens. Of this total, 161 were raw, unprocessed beef, veal, lamb, pork, or chicken; 101 were processed meats and meat dishes. C. perfringens was isolated from 113 (43.1%) of these specimens. The highest percentage of contamination (82%) was found in veal cuts, and the lowest (4.7%) in sliced sandwich meats and spreads. Only 2 of the 113 isolates were shown to produce heat-resistant spores, which indicates a very low incidence (0.8%) of contamination. These findings indicate that outbreaks of C. perfringens food-borne disease in the Cincinnati area are caused principally by the contamination of the food with vegetative cells or spores of the organism after cooking. Studies of the effects of various holding temperatures on the growth of C. perfringens indicated that, in the range of 5 to 15 C, no multiplication would occur, but that viable cells would still be present at the end of a 5-day holding period. Extremely rapid growth occurred at temperatures around 45 C, and complete inhibition of growth was accomplished between 49 and 52 C.     

Cerebrovascular Occlusive Disease—Experience with Panarteriography in 300 Consecutive Cases

Three hundred patients with cerebrovascular occlusive disease have had cerebral angiographic examination at the Veterans Administration Hospital, San Francisco, in the last five years. The present technique consists of preliminary visualization of the aortic arch and the major extracranial branches, followed by selective study of the subclavian and carotid arteries as necessary for evaluation of the intracranial circulation.Nine major complications occurred (an over-all incidence of 3 per cent). Two patients died after angiography and seven had major neurologic deficits persisting for more than 24 hours. Three of these patients had permanent damage, but four recovered completely.One-third of the patients had extracranial disease and one-third had intracranial disease. No significant lesion was found in the remainder. In the 212 patients with lesions, multiple lesions were common, the average number being three. Six patients had brain tumors and five had aneurysms.The mechanism of the stroke could be ascertained readily in most of the patients, but the extent of the disease and the resulting symptoms varied considerably. Several patients with occlusion of most of the cerebral vessels had minimal symptoms, while others had catastrophic symptoms but only minimal findings at arteriography.
