超极化活化环核苷酸门控(hyperpolarization-activated cyclic-nucleotide-gated,HCN)通道参与调制心脏跳动的节律和速率。与HCN1和HCN2有所不同,慢通道HCN4可能不存在电压依赖的滞后现象。本研究采用单细胞膜片钳方法,在稳定转染hHCN4的HEK293细胞上进行电生理记录,观察hHCN4通道是否存在滞后现象,以及cAMP对其的调制作用;同时采用实时定量RT-PCR方法检测窦房结和心房组织中HCNs的表达。电压钳实验结果显示hHCN4电流(Ih)激活随着保持电位超极化的变化而向去极化方向移动。三角电位变化钳(triangular ramp)和动作电位钳的结果也显示了hHCN4的滞后现象。cAMP增加Ih电流幅度,且使电流激活向去极化方向移动,从而改变内源性hHCN4滞后行为。RT-PCR结果显示,人窦房结组织主要表达HCN4,占75%,HCN1占21%,HCN2占3%,HCN3占0.7%。以上结果提示,人窦房结组织主要表达HCN4亚型,hHCN4的Ih存在电压依赖性的滞后现象,且受cAMP调制。由此推断,hHCN4通道的滞后现象可能在窦房结起搏活动中起到了关键作用。  相似文献

The ranges of 10 tropical Pacific phytoplankton species are shown in the maps. Five types of these ranges are discussed. Except for broadly tropical species some species with much narrower ranges are characterized. The bases of ranges and the expatriation areas are given. The bases of ranges of all the species studied fall into similar temperature and salinity limits. Most of the species inhabit a wide range of nutrients concentration, but Climacodium frauenfeldianum and Hemiaulus hauckii are only found at low phosphate and silicate concentrations. These peculiarities determine the geography of ranges of these two species. Among the species investigated only Ceratium gravidum belongs to the shade flora.  相似文献

The effects of retinoic acid (RA) on lung cancer cells were investigated. Both all-trans (t-RA) and 13-cis RA (c-RA) decreased specific 125I-VIP binding to NCI-H1299 cells in a time- and concentration-dependent manner. After 20 hr, 30 μM t-RA decreased specific 125I-VIP binding by 60%. By Scatchard analysis, the density of VIP binding sites but not the affinity was reduced by 42%. NCI-H1299 VPAC1 receptor mRNA was reduced by 48%. VIP caused a 3-fold elevation in the NCI-H1299 cAMP, and the increase in cAMP caused by VIP was reduced by 38% if the NCI-H1299 cells were treated with t-RA. Using the MTT assay, 3 μM t-RA and 3 μM c-RA inhibited NCI-H1299 proliferation by 60 and 23% respectively. Also, transforming growth factor (TGF)-β2 increased after treatment of NCI-H1299 cells with t-RA whereas TGF-β1 mRNA was unaffected and TGF-β3 mRNA was decreased. These results suggest that RA may inhibit lung cancer growth by down-regulating VPAC1 receptor and TGF-β3 mRNA but up-regulating TGF-β2 mRNA.  相似文献

Apicomplexan Cryptosporidium parasites infect a wide range of vertebrate hosts. While some species are limited to a single host group, such as Cryptosporidium baileyi, which infects chickens, other species of this genus, such as C. parvum, infect a wide range of mammalian species from mice to humans. During an investigation of Cryptosporidium infection in cattle on a farm in northern Poland, we identified an infection caused by C. felis, in addition to known infections with C. muris and C. parvum. This new infection was identified based on the size of the oocysts (mean size, 4.3 ± 0.4 μm; range, 3.5 to 5.0 μm), as well as by analysis of the molecular sequence of the variable region of the small-subunit rRNA. This finding demonstrates the complex host specificity and circulation in the environment of Cryptosporidium species.  相似文献

The tritium-labelled cloned 5S DNA from Lupinus luteus was used for localization of 5S RNA genes in Vicia faba subsp. minor metaphase chromosomes. In situ hybridization sites were found to be localized in chromosomes I and VI. In chromosome I the probe hybridized to the region adjoining NOR whereas in chromosome VI silver grains were found in the median part of the long arm. After prolonged exposure the autoradiographic grains expanded in the proximal part of that chromosome arm.  相似文献

Decrease in interstitial pH of the tumor stroma and over-expression of low density lipoprotein (LDL) receptors by several types of neoplastic cells have been suggested to be important determinants of selective retention of photosensitizers by proliferative tissues. The interactions of chlorin e6 (Ce6), a photosensitizer bearing three carboxylic groups, with plasma proteins and DOPC unilamellar vesicles are investigated by fluorescence spectroscopy. The binding constant to liposomes, with reference to the DOPC concentration, is 6 × 103 M− 1 at pH 7.4. Binding of Ce6 to LDL involves about ten high affinity sites close to the apoprotein and some solubilization in the lipid compartment. The overall association constant is 5.7 × 107 M− 1 at pH 7.4. Human serum albumin (HSA) is the major carrier (association constant 1.8 × 108 M− 1 at pH 7.4). Whereas the affinity of Ce6 for LDL and liposomes increases at lower pH, it decreases for albumin. Between pH 7.4 and 6.5, the relative affinities of Ce6 for LDL versus HSA, and for membranes versus HSA, are multiplied by 4.6 and 3.5, respectively. These effects are likely driven by the ionization equilibria of the photosensitizer carboxylic chains. Then, the cellular uptake of chlorin e6 may be facilitated by its pH-mediated redistribution within the tumor stroma.  相似文献

The discovery of phototropins, photoreceptors for chloroplast responses in Arabidopsis thaliana, brought about renewed interest in these blue light-controlled movements. Recent progress in research on their mechanisms in higher plants is briefly summarized. Phototropins mediate phototropism, chloroplast relocations and stomatal movements. Their functions are partially overlapping, with phot1 active predominantly in weak light and phot2 active in strong light. The accumulation response of chloroplasts appears to be mediated by phot1 and phot2 whereas the avoidance response is controlled by phot2. The role of Ca2+ as a potential intracellular messenger has been discussed in view of the recently demonstrated blue light-induced transient increases in the cytosolic Ca2+ mediated differently by phot1 and phot2. Differential inhibition of accumulation and avoidance responses by wortmannin, the inhibitor of phosphoinositide-3 kinases, in Lemna trisulca points to an important role of these enzymes in the signal transduction. A new, multi-domain component controlling chloroplast positioning and movement, CHUP1, encodes an actin-binding protein in Arabidopsis.  相似文献

The combining site of the Erythrina cristagalli lectin was studied by quantitative precipitin and precipitin inhibition assays. The lectin precipitated best with two fractions of a precursor human ovarian cyst blood group substance with I and i activities. A1, A2, B, H, Lea, and Leb blood group substances precipitated poorly to moderately and substances of the same blood group activity precipitated to varying extents. These differences are attributable to heterogeneity resulting from incomplete biosynthesis of carbohydrate chains. Specific precipitates with the poorly reactive blood group substances were found to be more soluble than those reacting strongly. Precipitation was minimally affected by EDTA or divalent cations. Among the monosaccharides and glycosides tested for inhibition of precipitation, p-nitrophenyl βdGal was most active and was 10 times more active than methyl βdGal, indicating involvement of hydrophobic contacts in site specificity. Methyl αdGalNAc, p-nitrophenyl αdGalNAc, methyl αdGal, N-acetyl-d-galactosamine, p-nitrophenyl αdGal, methyl βdGal, and p-nitrophenyl βdGalNAc were progressively less active than p-nitrophenyl βdGal. The best disaccharide inhibitor dGalβ1 → 4dGlcNAc was 7.5 times more potent than p-nitrophenyl βdGal. A tetraantennary and triantennary oligosaccharide containing four and three dGalβ1 → 4dGlcNAcβ1 → branches, respectively, were, because of cooperative binding effects, 1.6 and 2.5 times more active than the bi- and monoantennary oligosaccharides, respectively. dGalβ1 → 4dGlcNAcβ1 → 6dGal and dGalβ1 → 4dGlcNAcβ1 → 2dMan had the same activity, being 1.5 times more active than dGalβ1 → 4dGlcNAc, which was 2.6 and 8.5 times more active than dGalβ1 → 3dGlcNAc and dGalβ1 → 3dGlc, respectively. Substitutions by N-acetyl-d-galactos-amine or l-fucose on the d-galactose of inhibitory compounds blocked activity. These results suggest that a hydrophobic interaction with the subterminal sugar is important in the binding and that the specificity of the lectin combining site involves a terminal dGalβ1 → 4dGlcNAc and the β linkage of a third sugar.  相似文献

Cereals are known to be recalcitrant to the induction of morphogenesis in vitro and the majority of the methods used are callus-mediated and species/genotype-dependent. In the present investigation, a method of morphogenesis induction from immature scutella of selected barley cultivars was used, and particular attention was paid to histology in the initial stages of the regeneration process in order to confirm whether it occurs directly or indirectly (via callus formation). The length of the period from inoculating scutella on the medium to obtaining plantlets depended on the cultivar and the individual scutellum of the barley and varied between 2.5–4 months. The regeneration efficiency and viability of barley scutella was revealed to be highly genotype dependent. The average number of regenerated plants per regenerating scutellum was highest in the case of cv Granal (3.7). A histological analysis of the cultured explants showed both non-morphogenic and morphogenic callus formation. Two types of indirect morphogenetic response were observed: organogenesis (shoot bud formation) and somatic embryogenesis. This is the first report concerning an analysis of in vitro regeneration from immature scutella of barley cultivars (Stratus, Ryton, Granal and Binal).  相似文献

The aim of the study was to identify and determine the percent content of polyphenols in extracts from leaves and hawthorn bark, to examine the effect of the extracts on the properties of the biological membrane as well as to determine their antioxidant activity toward membrane lipids. In particular, a biophysical investigation was conducted on the effect of hawthorn extracts on the osmotic resistance and morphology of erythrocyte cells and on the packing of the heads of membrane lipids. Analysis of the polyphenol content of extracts used the HPLC method. Analysis of the polyphenol composition has shown a dominant share of procyanidins and epicatechin in both extracts. The research showed that the polyphenolic compounds contained in hawthorn extracts are incorporated mainly into the hydrophilic part of the erythrocyte membrane, inducing echinocyte shapes. They also diminish the packing order of the lipid polar heads of the membrane, as evidenced by the lowered generalized polarization values of Laurdan. The substances used induced increased osmotic pressure of erythrocytes, making them less sensitive to changes in osmotic pressure. The presence of the extract compounds in the outer hydrophilic part of the erythrocyte membrane, evidenced by examination of the shapes and packing in the hydrophilic part of membrane, indicates that the substances constitute a kind of barrier that protects the erythrocyte membrane against free radicals, while the membrane-bound extracts do not disturb the membrane structure and, thus, do not cause any side effects.  相似文献
