The purpose of this work was to evaluate two methods (floral dip and vacuum infiltration) of in planta transformation of Arabidopsis thaliana. The key issue of this work is the identification of the developmental stages of A. thaliana flower buds subjected to agroinfection, optimal for the successful transformation. Histological tests performed after agroinfection
made it possible to establish the patterns of a GUSPlus reporter gene expression in the examined plants and thus precisely define the range of flower developmental stages most appropriate
for efficient transformation. Two plasmids, CAMBIA 1305.1 and CAMBIA 2301, were used. Verification of the transgenic nature
of plants was carried out by detection of CaMV::gusA and CaMV::GUSPlus transgenes and their expression in transgenic plants by appropriate molecular and histochemical methods. For the flower dip
transformation, three concentrations of Silwet L-77 surfactant and two inoculation times were tested. The most efficient treatment
appeared to be 2-min-long flower bud inoculation and 400 μl/l surfactant (pCAMBIA 1305.1 − 1.73%; pCAMBIA 2301 − 2.01%). In
the case of vacuum infiltration method, the highest efficiency of the transformation occurred when the inoculation time was
4 min (pCAMBIA 2301 − 1.55%; pCAMBIA 1305.1 − 1.37%).

The series of quinoline derivatives were prepared. The synthetic approach, analytical, and spectroscopic data of all synthesized compounds are presented. All the prepared derivatives were analyzed using the reversed-phase high performance liquid chromatography (RP-HPLC) method for the lipophilicity measurement. In the present study, the correlation between RP-HPLC retention parameter log K (the logarithm of capacity factor K) and various calculated log P data is shown. The relationships between the lipophilicity and the chemical structure of the studied compounds are discussed as well. The prepared compounds were tested for their in vitro antifungal activity. 2-[(3-Hydroxyphenylimino)methyl]quinolin-8-ol (8), 2-[(4-hydroxyphenylimino)methyl]quinolin-8-ol (9) and 2-[(2,5-dichloro-4-nitrophenylamino)methoxymethyl]quinolin-8-ol (10) showed in vitro antifungal activity comparable to or higher than that of the standard fluconazole. Structure-activity relationships among the chemical structure, the physical properties, and the biological activities of the evaluated compounds are discussed in the article. 相似文献

Nitrate is a key node in the network of the assimilatory and respiratory nitrogen pathways. As one of the ‘fixed’ forms of nitrogen, nitrate plays an essential role in both nature and industry. For bacteria, it is both a nitrogen source and an electron acceptor. In agriculture and wastewater treatment, nitrate respiration by microorganisms is an important issue with respect to economics, greenhouse gas emission and public health. Several microbial processes compete for nitrate: denitrification, dissimilatory nitrate reduction to ammonium and anaerobic ammonium oxidation. In this review we provide an up to date overview of the organisms, genes and enzymes involved in nitrate respiration. We also address the molecular detection of these processes in nature. We show that despite rapid progress in the experimental and genomic analyses of pure cultures, knowledge on the mechanism of nitrate reduction in natural ecosystems is still largely lacking. 相似文献

Ulcerative colitis (colitis ulcerosa) is a non-specific inflammatory bowel disease of unknown etiology. The symptoms which are observed in the course of ulcerative colitis are: an increase in the number of leukocytes and blood platelets, an increase in the concentration of IL-6 and anemia. Blood platelets are the key element, linking the processes of hemostasis, inflammation and the repair of damaged tissues. Activation of blood platelets is connected with changes in their shape and the occurrence of the reaction of release. P-selectin appears on the surfaces of activated blood platelets and the concentration level of soluble P-selectin increases in the blood plasma. The aim of this study was to define whether the increased number of blood platelets in patients with ulcerative colitis accompanies changes in their activation and morphology. A total of 16 subjects with ulcerative colitis and 32 healthy subjects were studied. Mean platelet count, morphological parameters of platelets and MPC were measured using an ADVIA 120 hematology analyzer. Concentrations of sP-selectin and IL-6 in serum were marked by immunoassay (ELISA). MPC, concentration of sP-selectin and IL-6 were significantly higher in subjects with ulcerative colitis compared to those in the healthy group. There was a decrease of MPV in patients with ulcerative colitis, which is statistically significant. Chronic inflammation in patients with ulcerative colitis causes an increase in the number of blood platelets, a change in their morphology and activation. Decreased MPV value reflects activation and the role blood platelets play in the inflammatory process of the mucous membrane of the colon. A high concentration of sP-selectin, which is a marker of blood platelet activation, demonstrates their part in the inflammatory process. The increase in the concentration of sP-selectin correlated positively with the increase in concentration of IL-6. This is why it may be a useful marker of the activity of colitis ulcerosa. 相似文献

The current study aims to determine the involvement of cellular responses in combating Giardia intestinalis invasion. The study group consisted of 44 women and 18 men, aged 18-72 years, infected with G. intestinalis. The diagnosis was established based on laboratory investigations (examination of stool, choloscopy, GSA-65). Blood for analysis was collected before antiparasitic treatment and two weeks after treatment termination. The control group consisted of 22 women and 18 men aged 20-45 years. The serum concentrations of IL-5, IL-6, IL-13, TNF, IFN-g were assayed using a set of Quantikine human. The concentrations of NO in the serum were determined using a set of Total Nitric Oxide Assay. Patients infected with G. intestinalis showed a statistically significant increase in the levels of NO, IFN-g and IL-13. Even the antiparasitic treatment applied did not reduce the levels of these parameters and only caused a rise in IL-6. Our study showed a lack of acute inflammatory state in the course of G. intestinalis infection. 相似文献

The aim of this study was to evaluate regulatory T lymphocytes (Tregs) in the course of allergic contact dermatitis (ACD) and to elucidate the role of IL-10 and TGF-β in Tregs activity. Peripheral blood CD4(+)CD25(+) and CD4(+)CD25(high) cells were determined by flow cytometry in patients with acute disseminated ACD ('ad', n = 36), acute localized ACD ('al', n = 26), and disseminated ACD during remission ('rd', n = 27) as well as in controls (n = 22). Serum levels of cytokines were measured using ELISA. The mean percentage of CD4(+)CD25(+) and CD4(+)CD25(high) cells in patients with ad ACD was significantly higher than in controls (p < 0.01) and the remaining patients (p < 0.05). Both cell populations were significantly elevated in persons with widespread skin lesions (p < 0.05). In ad patients the CD4(+)CD25(+) increased during three weeks of disease, although the significant increase of CD4(+)CD25(high) was noted only in the third week. Patients with ad ACD showed a significantly decreased serum level of TGF-β1 as compared with controls and the remaining ACD patients. IL-10 level did not differ between all groups. The elevated population of CD4(+)CD25(high) cells in ad ACD patients, and its dependence on the extension of skin lesions, suggest a role of Tregs in regulating the course of ACD. The growing Tregs percentages may indicate their peripheral generation during ACD. The development of lesions despite an increased population of Tregs suggests their functional defect. The role of TGF-β1 in the suppressive activity of Tregs cannot be excluded. 相似文献

Rehmannia glutinosa Libosch., a valuable medicinal plant, was successfully propagated in vitro using shoot tip explants. Shoot multiplication was performed in glass tubes and in a nutrient sprinkle bioreactor. A mixture of 0.1 mg L?1 indole-3-acetic acid (IAA) and 1.0 mg L?1 of 6-benzylaminopurine in Murashige and Skoog (MS) agar-solidified medium proved the best combination for multiple shoot induction, yielding 8.2 shoots per explant after 4 weeks of culture in glass tubes. The number of shoots increased to 21 per explant when the same combination of growth regulators was used in a nutrient sprinkle bioreactor. The shoots rooted with a frequency of 93 % after 6 weeks of culture on MS agar medium supplemented with IAA (0.1 mg L?1) before being acclimatized in the greenhouse. The antioxidant activities of methanolic extracts from the leaves and roots of the in vitro-regenerated plants of R. glutinosa cultivated in the greenhouse were evaluated using four in vitro assays: scavenging of free radicals (DPPH and ABTS), transition metal reduction and total antioxidant activity phosphomolybdenum test. In all cases, the methanolic extract from leaves demonstrated better antioxidant activity than those taken from roots. A strong correlation was found between total phenolic and flavonoid content, and the antioxidant capacity of the studied extracts. 相似文献
