The purpose of this study was to examine the effect of short-term high fat feeding on the inflammatory response in polymicrobial sepsis. Male C57BL/6 mice at 6 weeks of age were randomized to a high-fat diet (HFD) (60% kcal fat) or control diet (CD) (16% kcal fat) for 3 weeks. After 3 weeks of feeding, sepsis was induced by cecal ligation and puncture (CLP) and animals were monitored for survival. In a separate experiment, after 3 weeks of feeding mice underwent CLP and were sacrificed at various time points thereafter. Tissue was collected for biochemical studies. Mice fed a HFD gained more weight and had a greater fat mass compared to CD-fed mice. Mice on a HFD had a lower probability of survival and more severe lung injury compared with CD-fed mice following sepsis. Myeloperoxidase (MPO) activity, an indicator of neutrophil infiltration, was increased in the lung and liver after CLP in HFD-fed mice compared with CD (P < 0.05). The plasma cytokines tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 were increased in both groups after CLP, however, TNF-α and IL-6 levels were lower in HFD mice at 3 h after CLP compared with CD and consistent with lung, but not liver, messenger RNA (mRNA) expression. Leptin levels were higher in HFD-fed mice at 18 h after sepsis compared to baseline levels (P < 0.05). Polymicrobial sepsis increased hepatic nuclear factor-κB (NF-κB) activation in HFD-fed mice after CLP vs. CD-fed mice. Short duration high fat feeding increases mortality and organ injury following polymicrobial sepsis. These effects correspond to changes in NF-κB. 相似文献

Autumn migration routes and orientation of Swedish Ospreys Pandion haliaetus were studied by satellite tracking of 18 birds. Of these, 13 could be followed during the entire migration (6 females, 5 males and 2 juveniles). Most birds migrated across western and central Europe to winter in tropical West Africa. However, one juvenile flew to Cameroon and one female used a very easterly route and reached Mozambique. On average, the birds travelled a total distance of about 6700 km, with little variation except for the female wintering in Mozambique, who travelled more than 10 000 km. Of 21 stopovers (of >1 day), only five were made south of 45°N; three of these in Africa. Females departed before males and juveniles and flew to a stopover site they probably were familiar with. After 3–4 weeks there, they continued to their wintering grounds. Also males and juveniles usually made one or more stopovers. Adults seemed to travel to a known wintering site, where they remained stationary, whereas juveniles were more mobile after reaching tropical regions, probably looking for good wintering sites. Males generally left the breeding area in directions similar to the mean migratory direction, whereas a few females departed in diverging initial directions. Apart from these diversions, adult Ospreys followed very straight migratory routes, with overall mean directions of 185–209° and with mean angular deviations of 6–33°. Some juveniles also departed in diverging directions. Moreover, young birds tended to show a larger variability in orientation. Thus, the Ospreys kept a fairly straight direction and did not avoid geographical obstacles such as mountain ranges and desert areas. However, they seemed reluctant to cross large water bodies. There was no correlation between angular deviation and length of the migrational segment, indicating that the principles of orientation by vector summation may not be valid for Osprey migration. Moreover, the geographic direction of migration did not vary in accordance with variations in the magnetic declination, suggesting that the Ospreys did not orient along magnetic loxodromes. 相似文献

Thirteen male subjects lifted boxes of three different weights for 20 min periods each. Ventilation, oxygen uptake and heart rates were measured. Each subject made subjective estimates of the degree of difficulty of the work he was doing. Oxygen uptake and ventilation were found to reach steady state after about four minutes and remain there for the rest of the work session. Heart rate for the two lighter boxes reached steady state after about thirteen minutes while the heart rate continued to rise when lifting was done with the heaviest box. The relationships between oxygen uptake, heart rate and work load tended to be curvilinear, whereas the ventilation was linear when compared to work load. The heart rate and oxygen uptake relationship was also curvilinear, while ventilation and oxygen uptake had a linear relationship. The subjective ratings were expected to be about one-tenth of the heart rate. Only the ratings made while lifting the lightest box reflected this. Triploid intersexes homozygous for a mutant (msl-2) known to impede the hyperactivation of the X chromosome in diploid males differentiate into adults, sexually indistinguishable from their heterozygous sibs. A shift toward female sexual differentiation mediated by manipulating the rearing temperature is accompanied by an apparent increase in the level of an X-linked gene product. This unexpected result is rationalized in terms of differential lethality of individuals at the two extremities of the distribution of X-activity levels in intersexes raised at a particular temperature. Many phenotypic changes of eukaryotic cells due to changes in gene expression depend on alterations in chromatin structure. Processes involved in the alteration of chromatin are diverse and include post-translational modifications of histone proteins, incorporation of specific histone variants, methylation of DNA and ATP-dependent chromatin remodeling. Interconnected with these processes are the localization of chromatin domains within the nuclear architecture and the appearance of various classes of noncoding regulatory RNAs. Recent experiments underscore the role of these processes in influencing diverse biological functions.
