The maximal amounts of growth of Selenomonas ruminantium were examined in the media containing various amounts of glucose. The yields of cells per unit weight of glucose are linear functions to glucose concentrations in the ranges between zero to 0.005% and 0.005 to 0.7%, Cell yields per glucose are greater in the former range, indicating greater a-mounts of energy are available per glucose at lower concentrations. Growth responses in lactate media containing various amounts of glucose showed that the preincubation with larger amounts of glucose is inhibitory for the following growth and metabolism of lactate. The organism produces predominantly lactate in the glucose medium. However, volatile fatty acid productions increase when the initial concentrations of glucose become low. Isotopic studies showed that the lactate utilization yielding volatile fatty acids is inhibited by the preceding metabolism of high concentrations of glucose. These results were discussed with regard to normal and abnormal fermentations in the rumen. 相似文献

Polynuclear aromatic hydrocarbons (PAH), including fluoranthene, pyrene, benzo(a)-anthracene, chrysene, benzo(a)pyrene (benzo(e)pyrene), dibenzo(a,h)anthracene and benzo(ghi)perylene, were identified and determined in sediments from Tokyo Bay. Their concentrations were proved to be in a range from several tens to several hundreds µg/kg of dry samples. This seems to suggest that the smaller are the average particle sizes of sediments and the higher are the total amounts of PAH concentrations.Statistically significant positive correlations were observed between the following pairs: total amount of PAH vs. clay content; total amount of PAH vs. the sum of (clay + silt) contents; total amount of PAH vs. ignition loss. Biosynthetic threonine deaminase was purified to an apparent homogeneous state from the cell extract of Proteus morganii, with an overall yield of 7.5%. The enzyme had a s020,w of 10.0 S, and the molecular weight was calculated to be approximately, 228,000. The molecular weight of a subunit of the enzyme was estimated to be 58,000 by sodium dodecyl sulfate gel electrophoresis. The enzyme seemed to have a tetrameric structure consisting of identical subunits. The enzyme had a marked yellow color with an absorption maximum at 415 nm and contained 2 mol of pyridoxal 5′-phosphate per mol. The threonine deaminase catalyzed the deamination of l-threonine, l-serine, l-cysteine and β-chloro-l-alanine. Km values for l-threonine and l-serine were 3.2 and 7.1 mm, respectively. The enzyme was not activated by AMP, ADP and ATP, but was inhibited by l-isoleucine. The Ki for l-isoleucine was 1.17 mm, and the inhibition was not recovered by l-valine. Treatment with mercuric chloride effectively protected the enzyme from inhibition by l-isoleucine. 相似文献

The changes during growth and sporulation in activities of cells of Bacillus subtilis to incorporate various amino acids were investigated with wild-type strain and its asporogenous mutant. In the case of wild type strain the uptake of valine, phenylalanine, and proline was largest during the logarithmic growth period. The uptake of these amino acids decreased rapidly during the early stationary phase. The uptake of valine and cysteine increased again to some extent just prior to the forespore stage. The uptake of glycine and serine, however, was largest at the forespore stage at which the formation of spore coat took place. From these observed phenomena it was assumed that the remarkable incorporation of glycine and serine into the wild type strain during sporulation was closely related to the formation of spore coat. 相似文献

Purification of the precipitates obtained from the juice oil of Citrus hassaku by chromatography afforded 7-geranyloxycoumarin (aurapten) and two compounds (mp 43~45°C and 122~124°C), whose structures were determined to be epoxyaurapten and marmin on the basis of spectral evidence. These compounds were isolated from Citrus hassaku for the first time. The spasmolytic activity was tested of aurapten, epoxyaurapten, marmin and their related compounds, which were synthesized from aurapten and marmin, on the small intestine removed from a male guinea pig. An N-acetylglutamokinase-deficient arginine-requiring mutant, KY9390 and an N-acetylornithine aminotransferase-deficient arginine-requiring mutant, AA-7 were derived from a wild-type strain and an l-arginine-producing mutant of Corynebacterium glutamicum, respectively. The tetradecapeptide of a renin substrate, DRVYIHPFHLLVYS, was used as a substrate for assaying several fungal aspartic and acidic proteinases in the acidic pH range. Pepsin primarily cleaved the valy3-tyrosyl4 (V3-Y4) and leucyl10-leucyl11 (L10-L11) bonds. The growth factor for malo-lactic fermentation bacteria was partially purified from tomato juice, and some properties of the factor were examined. The factor was comparatively a low molecule, thermo-stable and insoluble in nonpolar organic solvents. The purified factor fraction showed positive reaction with Molisch-, tetrazolium- and Nessler-reagents, suggesting the existence of sugar and nitrogen as the chemical components. The treatment with cellobiase or cellulase-II resulted in the inactivation of the factor and simultaneously released the sugar constituent composed of glucose from the factor. These results indicated that the glucose residue might exist as the β-1,4-glucosidic linkage in the growth factor. 相似文献
