New series of pyrazoles 4a – c and pyrazolopyrimidines 5a – f had been constructed. The newly synthesized compounds were assessed for their antimicrobial activity towards E. coli and P. aeruginosa (gram –ve bacteria), B. subtilis and S. aureus (gram +ve bacteria) and A. flavus and C. albicans (representative of fungi). The pyrazolylpyrimidine-2,4-dione derivative 5b is the most active candidate against B. subtilis (MIC=60 μg/mL) and P. aeruginosa (MIC=45 μg/mL). Regarding antifungal potential, compound 5f was the most effective against A. flavus (MIC=33 μg/mL). Similarly, compound 5c displayed strong antifungal activity towards C. Albicans (MIC=36 μg/mL) in reference to amphotericin B (MIC=60 μg/mL). Finally, the novel compounds had been docked inside dihydropteroate synthase (DHPS) to suggest the binding mode of these compounds.  相似文献

The potential toxicity of nanoparticles in plants is scarce and contradictory. Despite the diversity of research efforts, a detailed explanation of the TiO2NPS effects in plant photosynthesis is still missing. The present work gives a new approach to examine the impact of the TiO2NPs on crop production (development and photosynthesis) and plant protection (tolerance and defense systems) in fenugreek (Trigonella foenum graecum L.). Seedlings were assessed in greenhouse trials to estimate the influence of TiO2NPs on physiological characters for 16 days. They were treated with TiO2NPs at a size less than 20 nm. The results revealed that there were no significant effects on seedlings growth and biomass of stem, but a decrease in the fresh weight of leaves after TiO2NPs treatment. Plants treated with 100 mg·L?1 of TiO2NPs presented a reduction and chlorosis in leaf area due to a significant decrease in the chlorophyll a and b contents. The highest value of the photosynthetic pigments was recorded at 50 mg·L?1 of TiO2NPs. However, the treatment with 100 mg·L?1 of TiO2NPs caused a decrease in the levels of chlorophyll a, b and of carotenoids. Both doses of TiO2NPs induced an accumulation of anthocyanins compared to the control after 16 days of seedling development. A nano-stress significantly decreased the flavonoids level, but increased that of polyphenols compared to control after 16 days of exposure. The decrease in the translocation ratio of flavonoids suggests that many of them contain an enediol group, which suggests that they may act as bidentate ligands for anatase TiO2NPs. Accordingly, nano-stressed leaves exhibited significantly enhanced GPOX, CAT and APX activity levels. On the contrary, GPOX and CAT activities were reduced substantially in stems treated with 100 mg·L?1 TiO2NPs. The accumulation of MDA was found to be higher in stems than in leaves. This could be explained by the accumulation of nanoparticles in different organs; it could be that the stems are the favored targets of nanoparticles. These results underline the necessity for a deeper estimation of nanoparticle ecotoxicity and particularly concerning their interaction with plants.  相似文献

Macrophage migration inhibitory factor (MIF) is a multifunctional protein and a major mediator of innate immunity. Although X-ray crystallography revealed that MIF exists as a homotrimer, its oligomerization state in vivo and the factors governing its oligomerization and stability remain poorly understood. The C-terminal region of MIF is highly conserved and participates in several intramolecular interactions that suggest a role in modulating the stability and biochemical activity of MIF. To determine the importance of these interactions, point mutations (A48P, L46A), insertions (P107) at the monomer-monomer interfaces, and C-terminal deletion (Delta 110-114NSTFA and Delta 105-114NVGWNNSTFA) variants were designed and their structural properties, thermodynamic stability, oligomerization state, catalytic activity and receptor binding were characterized using a battery of biophysical methods. The C-terminal deletion mutants DeltaC5 huMIF 1-109 and DeltaC10 huMIF 1-104 were enzymatically inactive and thermodynamically less stable than wild type MIF. Analytical ultracentrifugation studies demonstrate that both C-terminal mutants sediment as trimers and exhibit similar binding to CD74 as the wild type protein. Disrupting the conformation of the C-terminal region 105-114 and increasing its conformational flexibility through the insertion of a proline residue at position 107 was sufficient to reproduce the structural, biochemical and thermodynamic properties of the deletion mutants. P107 MIF forms an enzymatically inactive trimer and exhibits reduced thermodynamic stability relative to the wild type protein. To provide a rationale for the changes induced by these mutations at the molecular level, we also performed molecular dynamics simulations on these mutants in comparison to the wild type MIF. Together, our studies demonstrate that intersubunit interactions involving the C-terminal region 105-114, including a salt-bridge interaction between Arg73 of one monomer and the carboxy terminus of a neighboring monomer, play critical roles in modulating tertiary structure stabilization, enzymatic activity, and thermodynamic stability of MIF, but not its oligomerization state and receptor binding properties. Our results suggest that targeting the C-terminal region could provide new strategies for allosteric modulation of MIF enzymatic activity and the development of novel inhibitors of MIF tautomerase activity.  相似文献

We have studied the configuration of genes encoding for the heavy and light chains in the tumoral cells of 6 patients affected by alpha heavy chain disease (alpha HCD). The results showed the presence of rearrangement of the alpha heavy chain as well as the kappa light chain genes whereas the lambda genes were in germinal configuration. Thus, these results suggest the presence of a monoclonal compound in the tumoral cells in the alpha HCD.  相似文献

Whether it is men or women who suppress female sexuality has important implications for understanding gendered relations, ultimately providing insight into one widespread cause of female disadvantage. The question of which sex suppresses female sexuality more avidly, however, neglects that our interests are never unambiguously masculine or feminine; each of us has a combination of male and female kin which alters how much of our future fitness derive from each sex. Here we exploit a nationally representative sample of 600 Tunisians to test whether support for Islamic veiling—a proxy for female sexual suppression—is more common amongst one sex than the other, and is affected by the relative sex of one's offspring (i.e., the number of sons relative to daughters). We find that men are more supportive of Islamic veiling than women, but women with more sons are more supportive of veiling and more likely to wear veils than women with fewer sons. All effects were robust to the inclusion of religiosity, which was weaker amongst men and unrelated to the number of sons a woman had. The number of daughters affected neither religiosity nor support for veiling, but did increase women's likelihood of wearing contemporary, fashionable Tunisian veils compared with no head covering. We further found that men were more religious if they had more sons. Overall, these findings highlight that far from being the fixed strategy of one sex or the other, female sexual suppression manifests facultatively to promote one's reproductive interests directly or indirectly by creating conditions beneficial to one's descendent kin. These results show that both men and women can suppress female sexuality, although the function in either case appears more closely aligned with male rather than female interests.  相似文献

Morphological markers/traits are often used in the detection of allelopathic stress, but optical signals including chlorophyll a fluorescence emission could be useful in developing new screening techniques. In this context, the allelopathic effect of barley (Hordeum vulgare subsp. vulgare) root exudates (three modern varieties and three landraces) were assessed on the morphological (root and shoot length, biomass accumulation), physiological (Fv/Fm and F0), and biochemical (chlorophyll and protein contents) variables of great brome (Bromus diandrus Roth., syn. Bromus rigidus Roth. subsp. gussonii Parl.). All the measured traits were affected when great brome was grown in a soil substrate in which barley plants had previously developed for 30 days before being removed. The response of receiver plants was affected by treatment with activated charcoal, dependent on barley genotype and on the nature of the growing substrate. The inhibitory effect was lower with the addition of the activated charcoal suggesting the release of putative allelochemicals from barley roots into the soil. The barley landraces were more toxic than modern varieties and their effect was more pronounced in sandy substrate than in silty clay sand substrate. In our investigation, the chlorophyll content and Fv/Fm were the most correlated variables with barley allelopathic potential. These two parameters might be considered as effective tools to quantify susceptibility to allelochemical inhibitors in higher plants.  相似文献

The study of the spatiotemporal distribution of talitrid amphipods was carried out along a transect in three stations belonging to the lagoon complex of Ghar El Melh, namely the supralittoral zones of the old harbour of Ghar El Melh, the opposite to Boughaz and Sidi Ali Mekki lagoon. Four species belonging to Talitridae family with two different genera were identified at all stations. These species are Orchestia gammarellus, Orchestia mediterranea, Orchestia stephenseni and Platorchestia platensis. The first two species were collected during the four seasons. The study of the density showed that the most important global mean density in talitrid was found in the supralittoral zone of opposite to Boughaz. In addition, statistical analysis showed a significant difference in the seasonal distribution of amphipods based on plant associations. Furthermore, the highest diversity of talitrid was observed in the winter in the supralittoral zone of opposite to Boughaz and in summer in the supralittoral zones of Sidi Ali Mekki lagoon and the old harbour of Ghar El Melh. Results showed that at all stations, a highly significant correlation of amphipod community with Cymodocea banquette was observed. Moreover, principal component analysis showed that the distribution of the different species depends on climatic and edaphic factors.  相似文献

Cytotoxic CD8+ T cells (CTLs) play a critical role in controlling viral infections. HIV-infected individuals develop CTL responses against epitopes derived from viral proteins, but also against cryptic epitopes encoded by viral alternative reading frames (ARF). We studied here the mechanisms of HIV-1 escape from CTLs targeting one such cryptic epitope, Q9VF, encoded by an HIVgag ARF and presented by HLA-B*07. Using PBMCs of HIV-infected patients, we first cloned and sequenced proviral DNA encoding for Q9VF. We identified several polymorphisms with a minority of proviruses encoding at position 5 an aspartic acid (Q9VF/5D) and a majority encoding an asparagine (Q9VF/5N). We compared the prevalence of each variant in PBMCs of HLA-B*07+ and HLA-B*07- patients. Proviruses encoding Q9VF/5D were significantly less represented in HLA-B*07+ than in HLA-B*07- patients, suggesting that Q9FV/5D encoding viruses might be under selective pressure in HLA-B*07+ individuals. We thus analyzed ex vivo CTL responses directed against Q9VF/5D and Q9VF/5N. Around 16% of HLA-B*07+ patients exhibited CTL responses targeting Q9VF epitopes. The frequency and the magnitude of CTL responses induced with Q9VF/5D or Q9VF/5N peptides were almost equal indicating a possible cross-reactivity of the same CTLs on the two peptides. We then dissected the cellular mechanisms involved in the presentation of Q9VF variants. As expected, cells infected with HIV strains encoding for Q9VF/5D were recognized by Q9VF/5D-specific CTLs. In contrast, Q9VF/5N-encoding strains were neither recognized by Q9VF/5N- nor by Q9VF/5D-specific CTLs. Using in vitro proteasomal digestions and MS/MS analysis, we demonstrate that the 5N variation introduces a strong proteasomal cleavage site within the epitope, leading to a dramatic reduction of Q9VF epitope production. Our results strongly suggest that HIV-1 escapes CTL surveillance by introducing mutations leading to HIV ARF-epitope destruction by proteasomes.  相似文献
