An integrated multi-trophic aquaculture (IMTA) system, with one fish cage model surrounded by an island and shellfish rafts, was used in the current study. Planktonic and sediment bacterial communities in the IMTA system were monitored over four seasons in 2019. In both plankton and sediment samples, the most dominant phyla were Proteobacteria and Bacteroidota. Sediment bacterial samples were more similar and had higher levels of biodiversity than planktonic bacterial samples. Obvious seasonal variations were found in plankton samples, but not in sediment samples. No obvious inter-site variations in planktonic and sediment bacteria (fish cages, shellfish rafts and control sites) were found and the results suggested that no obvious impact of feeding operations in fish culture cage model on bacterial communities in the IMTA system was observed in this study. Based on the sequence data, some faecal indicator bacteria and potentially pathogenic bacterial species were detected. According to the results, the bacterial water quality in the IMTA system was acceptable. PICRUSt (Phylogenetic Investigation of Communities by Reconstruction of Unobserved States) analysis revealed that the primary difference in potential functional roles of planktonic and sediment bacteria was amino acid transport and metabolism, which was active in different seasons.  相似文献

Non-specific phospholipase C (NPC) is involved in plant growth, development and stress responses. To elucidate the mechanism by which NPCs mediate cellular functions, here we show that NPC4 is S-acylated at the C terminus and that acylation determines its plasma membrane (PM) association and function. The acylation of NPC4 was detected using NPC4 isolated from Arabidopsis and reconstituted in vitro. The C-terminal Cys-533 was identified as the S-acylation residue, and the mutation of Cys-533 to Ala-533 in NPC4 (NPC4C533A) led to the loss of S-acylation and membrane association of NPC4. The knockout of NPC4 impeded the phosphate deficiency-induced decrease of the phosphosphingolipid glycosyl inositol phosphoryl ceramide (GIPC), but introducing NPC4C533A to npc4-1 failed to complement this defect, thereby supporting the hypothesis that the non-acylated NPC4C533A fails to hydrolyze GIPC during phosphate deprivation. Moreover, NPC4C533A failed to complement the primary root growth in npc4-1 under stress. In addition, NPC4 in Brassica napus was S-acylated and mutation of the S-acylating cysteine residue of BnaC01.NPC4 led to the loss of S-acylation and its membrane association. Together, our results reveal that S-acylation of NPC4 in the C terminus is conserved and required for its membrane association, phosphosphingolipid hydrolysis and function in plant stress responses.  相似文献

A hairpin RNA-encoding construct targeting gmFAD2-1 was transformed into soybean, and an optimised Agrobacterium-mediated embryonic tip system was employed. A novel intergeneric grafting method using transgenic plantlets as scions was used instead of the conventional rooting method. Compared with traditional acclimatisation, the survival ratio of cleft grafts increased by 70%, and the culture period was shortened by about 40 days. The regeneration frequency of the grafted shoots in this embryonic tip system was approximately 74.6%. Soybean transformants were confirmed by Southern and Northern blot hybridisation analyses. The fatty acid composition of the T1 and T2 seeds from the transformed plants was determined by gas chromatography. The resulting downregulation of the gmFAD2-1 gene substantially increased the level of oleic acid from 16% to 55% as indicated by the oleic desaturation proportion (ODP). The ratio of plants with high ODP, moderate ODP and low ODP was about 1:2:1, which was consistent with a single-gene segregation pattern.  相似文献

Statistical methodology was applied to the optimization of the ammonium oxidation by Nitrosomonas europaea for biomass concentration (CB), nitrite yield (YN) and ammonium removal (RA). Initial screening by Plackett-Burman design was performed to select major variables out of nineteen factors, among which NH4Cl concentration (CN), trace element solution (TES), agitation speed (AS), and fermentation time (T) were found to have significant effects. Path of steepest ascent and response surface methodology was applied to optimize the levels of the selected factors. Finally, multi-objective optimization was used to obtain optimal condition by compromise of the three desirable objectives through a combination of weighted coefficient method coupled with entropy measurement methodology. These models enabled us to identify the optimum operation conditions (CN = 84.1 mM; TES = 0.74 ml; AS = 100 rpm and T = 78 h), under which CB = 3.386×108 cells/ml; YN = 1.98 mg/mg and RA = 97.76% were simultaneously obtained. The optimized conditions were shown to be feasible through verification tests.  相似文献
