Rat liver tissue was used to examine the effect of an exogenous succinyl-CoA-generating system on the radiochemical assay for delta-aminolevulinic acid synthase (succinyl-CoA:glycine C-succinyltransferase (decarboxylating), EC 2.3.1.37) activity developed by Ebert et al. (Ebert, P.S., Tschudy, D.P., Choudry, J.N. and Chirigos, M.A. (1970) Biochim. Biophys. Acta 208, 236--250). In the absence of exogenous succinate thiokinase, 34--62% (average 55%) of the radioactivity in the final column eluate could be attributed to delta-amino-[4-14C]levulinic acid, as assessed by conversion of delta-aminolevulinic acid in the eluate to a pyrrole. The addition of succinate thiokinase markedly enhanced the formation of the contaminant(s), as succinyl-CoA was metabolized to a compound or compounds that eluted chromatographically with delta-amino-levulinic acid. This effect was abolished by 10 mM EDTA, probably because the generation of succinyl-CoA was suppressed due to the chelation of Mg2+. The steroid 21-hydroxylase activity present in the microsomes of bovine adrenals is stimulated by components of the cytosol. The nature of these activators has been examined by two procedures. The first consisted of treating cytosol with increasing amounts of acetone. When the concentration of the organic solvent reached 50%, a precipitate, presumably proteinaceous, formed. The portion of the precipitate that was redissolvable in 0.05 m potassium phosphate buffer, pH 7.2, contained 7–15% of the stimulatory activity originally present in the cytosol. When the acetone concentration was raised to 90%, another active material precipitated. It was identified as oxidized glutathione (GSSG) and it accounted for about 5% of the activity in the cytosol. In an attempt to avoid the harmful effects of acetone, the second procedure employed only gel filtration and ion exchange resin chromatography. By these means the cytosol was separated into 11 protein fractions and a small molecular weight material. Forty six percent of the proteins and the same fraction of the stimulatory activity present in the original cytosol were recovered. Because all 11 protein fractions contained some stimulatory activity, the results suggested that the protein constituents of these fractions were relatively nonspecific. Yet, of the several known proteins which were tested for activity (bovine serum albumin, ovalbumin, human γ-globulin, bovine pancreatic ribonuclease, and pig insulin) only bovine serum albumin proved to be active. An additional 8% of the stimulatory activity of the cytosol was present in the fraction containing the low molecular weight components and this was all attributable to its GSSG content. 相似文献
A procedure was developed to reduce premature bolting and flowering in Chinese cabbage grown in the field during the winter. This involved (a) the use of geothermic water as an inexpensive heat source for heating the transplants in the nursery; (b) application of the growth retardants daminozide (Alar) and paclobutrazol (PP333) to the heated plants before removing them to the field; and (c) application of daminozide to the plants in the field. This procedure markedly reduced stem elongation and the length of the internal stem in relation to the head without affecting the size of the head. Furthermore, application of daminozide in the field minimized the severity of tipburn, a common physiological disorder in Chinese cabbage fields. 1. 1.|Resistance to cold through non-shivering thermogenesis (the absolute increase in O2 consumption caused by noradrenaline injection) was increased 3-fold in individuals of the broad-toothed mouse A. mystacinus, kept for 3 weeks under a short photoperiod (long scotophase 8L:16D) at an ambient temperature of 28°C, compared to control conditions (12L:12D; 28°C), and did not differ significantly from the winter-acclimatized group.
2. 2.|Acclimation of the same individuals to long scotophase and cold (8L:16d; 7°C) caused a significant (P < 0.01) increase in absolute O2 consumption and maximal body temperature, as a response to noradrenaline injectin, when compared to long-scotophase individuals (8L:16D; 28°C).
3. 3.|The results of this study support the idea that winter acclimatization of heat-production mechanisims may be induced by the extension of scotophase, which cycles very regulary in nature and in the Mediterranean region occurs before the beginning of the cold season.

The urine composition of three rodent species, mole rat Spalax ehrenbergi, the golden hamster Mesocricetus auratus and the white rat Rattus norvegicus was studied. These three species represent different degrees of fossoriality. The results show that the urine of the species that show a higher degree of fossoriality, the mole rat and the hamster, contain high values of calcium and magnesium bicarbonates when compared with the white rat. From these results it may be assumed that the mole rat as well as the hamster can use the kidney as a pathway for releasing bicarbonate and carbonate. This mechanism may reduce the CO2 concentration in their hypercapnic environment. 相似文献
To determine the influence of body position on chest wall and pulmonary function, we studied the ventilatory, pulmonary mechanics, and thoracoabdominal motion profiles in 20 preterm infants recovering from respiratory disease who were positioned in both the supine and prone position. Thoracoabdominal motion was assessed from measurements of relative rib cage and abdominal movement and the calculated phase angle (an index of thoracoabdominal synchrony) of the rib and abdomen Lissajous figures. The ventilatory and pulmonary function profiles were assessed from simultaneous measurements of transpulmonary pressure, airflow, and tidal volume. The infants were studied in quiet sleep, and the order of positioning was randomized across patients. The results demonstrated no significant difference in ventilatory and pulmonary function measurements as a function of position. In contrast, there was a significant reduction (-49%) in the phase angle of the Lissajous figures and an increase (+66%) in rib cage motion in prone compared with the supine position. In addition, the degree of improvement in phase angle in the prone position was correlated to the severity of asynchrony in the supine position. We speculate that the improvement in thoracoabdominal synchrony in the prone position is related to alterations of chest wall mechanics and respiratory muscle tone mediated by a posturally related shift in the area of apposition of the diaphragm to the anterior inner rib cage wall and increase in passive tension of the muscles of the rib cage. This study suggests that the mechanical advantage associated with prone positioning may confer a useful alternative breathing pattern to the preterm infant in whom elevated respiratory work loads and respiratory musculoskeletal immaturity may predispose to respiratory failure. 相似文献
