Microbes are responsible for cycling carbon (C) through soils, and predicted changes in soil C stocks under climate change are highly sensitive to shifts in the mechanisms assumed to control the microbial physiological response to warming. Two mechanisms have been suggested to explain the long-term warming impact on microbial physiology: microbial thermal acclimation and changes in the quantity and quality of substrates available for microbial metabolism. Yet studies disentangling these two mechanisms are lacking. To resolve the drivers of changes in microbial physiology in response to long-term warming, we sampled soils from 13- and 28-year-old soil warming experiments in different seasons. We performed short-term laboratory incubations across a range of temperatures to measure the relationships between temperature sensitivity of physiology (growth, respiration, carbon use efficiency, and extracellular enzyme activity) and the chemical composition of soil organic matter. We observed apparent thermal acclimation of microbial respiration, but only in summer, when warming had exacerbated the seasonally-induced, already small dissolved organic matter pools. Irrespective of warming, greater quantity and quality of soil carbon increased the extracellular enzymatic pool and its temperature sensitivity. We propose that fresh litter input into the system seasonally cancels apparent thermal acclimation of C-cycling processes to decadal warming. Our findings reveal that long-term warming has indirectly affected microbial physiology via reduced C availability in this system, implying that earth system models including these negative feedbacks may be best suited to describe long-term warming effects on these soils.  相似文献

Although the ray‐finned fishes are named for their bony, segmented lepidotrichia (fin rays), we are only beginning to understand the morphological and functional diversity of this key vertebrate structure. Fin rays support the fin web, and their material properties help define the function of the entire fin. Many earlier studies of fin ray morphology and function have focused on isolated rays, or on rays from only one or two fins. At the same time, relatively little is known about how different preservation techniques affect the material properties of many vertebrate structures, including fin rays. Here, we use three‐point bending tests to examine intra‐ and inter‐fin variation in the flexural stiffness of fin rays from yellow perch, Perca flavescens. We sampled fin rays from individuals that were assigned to one of three preservation treatments: fresh, frozen, and preserved with formalin. The flexural stiffness of the fin rays varied within and among fins. Pelvic‐fin rays were the stiffest, and pectoral fin rays the least stiff. The fin rays of the dorsal, anal, and caudal fins all had similar stiffness values, which were intermediate relative to those from the paired fins. The flexural stiffness of the fin rays was higher in rays that were at the leading edge of the fin. This variation in flexural stiffness was associated with variation in joint density and the relative length of the unsegmented proximal base of the fin rays. There was no significant difference in flexural stiffness between fresh and frozen specimens. In specimens preserved with formalin, there is a small but significant effect on stiffness in smaller fin rays.  相似文献

Oxygen consumption and lactate production above resting levels, and selected body temperatures, were measured in the lizard Agama stellio. Active and resting VO2 have low Q10 (1.7, 2.0) in the activity range 30-37 degrees C and higher Q10 (3.8, 4.0) below this. A correlation was found between published resting and active VO2 of lizards, and between VO2 and lifestyle. Four types were recognized, in order of increasing VO2: (a) fossorial; (b) sit-and-wait (including A. stellio); (c) cruising, and (d) widely foraging. A. stellio has a high capacity for lactate production, correlated with its short but rapid bursts of activity. This accounts for 80-90% of the energy used during 30 sec maximal activity.  相似文献

Antoantibodies to cholesterol were detected and purified from normal (nonimmunized) pig serum. The antibodies were assayed by ELISA with crystalline cholesterol as an Ag and by C-dependent damage to cholesterol-laden liposomes. Intravenous injection of liposomes containing cholesterol into anesthetized animals caused decreased hemolytic complement titers, and induced a reaction consisting of transient neutropenia, thrombocytopenia, respiratory distress, cyanosis, pulmonary and systemic hypertension, and decreased cardiac output. Plasma levels of thromboxane B2 and 6-keto-prostaglandin F1 alpha increased 1300 and 200%, respectively, and leukocyte and platelet counts decreased by 36 and 38%, respectively. Injection of cholesterol-free liposomes did not induce the reaction. These results show that naturally occurring autoantibodies to cholesterol can initiate C activation and can be associated with anaphylactoid reaction to exogenously administered cholesterol in pigs.  相似文献

An identification system for individual snakes, Natrix maura , was developed, based on photographs of the pattern on the ventral scales. The patterns appeared to be constant, and contained sufficient information (allowing for non-randomness within the pattern) to render duplication unlikely, even in extremely large populations. After digitization, the patterns were compared for overall similarity by computer, and those which matched were checked visually. Using this system on a third of the scales, only 1% of known recaptures (those in which one of the compared photographs was of poor quality) were missed, and coding was reduced to 2.2 min. This sifting makes recognition by natural marks practicable in very large samples, and is robust enough to absorb considerable error in coding.  相似文献
