启动子是决定基因表达水平的重要因素之一。组成型表达启动子被认为是工业上表达重要蛋白质的理想启动子。本研究利用蔗糖为唯一碳源的基本培养基对榨糖废水浸润的土壤微生物宏基因组文库进行筛选,获得两个阳性克隆。对其中一个克隆的柯斯质粒进行亚克隆,利用在线启动子预测和序列比对工具对其中一个亚克隆子进行分析,获得一个启动子序列。然后,利用PCR方法将该启动子和地衣芽孢杆菌的α-淀粉酶基因一起克隆到T载体上。结果表明该启动子在不加诱导剂的条件下能够在大肠杆菌中启动外源基因的高效表达。本研究结果为在生物领域中组成型启动子的应用研究提供了基础。  相似文献

甜菜碱醛脱氢酶(BADH)在植物抗逆反应中发挥着重要作用。文中从胡杨cDNA克隆到2个甜菜碱醛脱氢酶基因,分别命名为PeBADH1和PeBADH2。PeBADH1和PeBADH2均编码503个氨基酸的蛋白质,预测分子量分别是54.93 kDa和54.90 kDa。组织表达模式分析发现这2个基因在正常生长、盐和H2O2胁迫下,在不同组织中的表达模式有较大差异。在大肠杆菌中表达并纯化了2个基因的重组蛋白。酶活性分析显示PeBADH1和PeBADH2蛋白对底物的活性分别是0.073μmol/(min.mg)和0.107μmol/(min.mg)。热力学稳定性分析显示这2个蛋白的热力学稳定性具有明显差异。因此,基因表达模式差异与蛋白质酶学性质的不同预示着这2个基因可能存在功能上的分化。  相似文献

克雷伯氏菌(Klebsiella pneumonia)甘油歧化发酵生产1,3-丙二醇(1,3-PD)的过程中,乳酸是氧化途径最主要的副产物,乳酸的产生和积累,不仅限制了菌体本身的生长,而且严重影响了1,3-丙二醇的转化率。利用λRed重组技术对Klebsiella pneumonia中的酶乳酸脱氢酶基因(ldhA)进行改造。在λRed重组系统作用下,将带有300 bp的线性同源片段ldhA1-Cm-ldh A2与基因组DNA的同源重组,经过抗性筛选和PCR鉴定最终获得了ldhA基因缺失菌株K.pneumonia2-1ΔldhA。经过24 h发酵可知,乳酸最大产出浓度由原来的10.16 g/L降为0.49 g/L,1,3-PD由原来的78.83 g/L增长为85.76 g/L,甘油转化率由60.64%增长到65.97%,提高了5.33%。  相似文献

目的:观察八肽胆囊收缩素(CCK-8)对内毒素休克(ES)时海马损伤的影响,并探讨其可能的作用机制。方法:将日本大耳白兔经静脉注入内毒素的主要活性成分脂多糖(LPS,8mg/kg)复制ES模型。动物(32只)随机分为对照组、LPS组、CCK-8+LPS组和非特异性CCK受体拮抗剂丙谷胺(Pro)+LPS组(n=8)。监测平均动脉压(MAP)的变化,光、电镜观察海马的组织形态学改变,比色法检测海马NOS和SOD活性、N0和MDA含量的改变.用SD大鼠(12只,同上复制模型及分组)以免疫组织化学染色法观察海马iNOS和nNOS表达的变化。结果:与对照组相比,注入LPS后出现MAP显著而持续下降(P〈0.01);海马部位神经元损伤明显;iNOS和nNOS表达增强,NOS活性、NO和MDA含量显著升高(P〈0.05、P〈0.01和P〈0.01),SOD活性则降低(P〈0.01)。预先注入CCK-8可明显减轻上述变化,预先注入Pro则加剧以上变化。结论:CCK-8可减轻ES时脑内海马部位的损伤。其机制可能与其抗氧化作用和抑制NO的过量生成有关。  相似文献

In this study, we evaluated the in vitro activity of echinocandins, azoles, and amphotericin B alone and in combination against echinocandin/azole-sensitive and echinocandin/azole-resistant Candida glabrata isolates. Susceptibility tests were performed using the broth microdilution method in accordance with the Clinical and Laboratory Standards Institute document M27-A3. The checkerboard method was used to evaluate the fractional inhibitory concentration index of the interactions. Cross-resistance was observed among echinocandins; 15% of the isolates resistant to caspofungin were also resistant to anidulafungin and micafungin. Synergistic activity was observed in 70% of resistant C. glabrata when anidulafungin was combined with voriconazole or posaconazole. Higher (85%) synergism was found in the combination of caspofungin and voriconazole. The combinations of caspofungin with fluconazole, posaconazole and amphotericin B, micafungin with fluconazole, posaconazole and voriconazole, and anidulafungin with amphotericin B showed indifferent activities for the majority of the isolates. Anidulafungin combined with fluconazole showed the same percentage of synergism and indifference (45%). Antagonism was detected in 50% of isolates when micafungin was combined with amphotericin B. Combinations of echinocandins and antifungal azoles have great potential for in vivo assays which are required to evaluate the efficacy of these combinations against multidrug-resistant C. glabrata strains.  相似文献

Grass carp, Ctenopharyngodon idellus (Valenciennes, 1844), is an economically important species widely cultured in the world, but its genome research resources are largely lacking. The objectives of this study were to construct normalized cDNA libraries for efficient EST analysis, to generate ESTs from these libraries, and to identify EST-related molecular markers such as microsatellites and single nucleotide polymorphisms (SNPs) for genetic analysis of this species. A total of 6,269 ESTs were generated representing 4,815 unique sequences, from which 105 putative microsatellites and 5,228 SNPs were identified. These genome resources provide the material basis for future genetic and functional analyses in this species.  相似文献

The objective of this study was to analyze the clinical manifestation, imaging characteristics, genotype, and the relationship between the three aforementioned parameters in two pedigrees suffering from spinocerebellar ataxia. To evaluate the clinical manifestation of the two pedigrees and to compare the characteristics, we performed the MRI analysis of some patients from both pedigrees, while 2 ml of the peripheral blood sample was collected for gene analysis. The gene analysis data showed that pedigree 1 was certified spinocerebellar ataxia type-2 (SCA2); the CAG repeats in the proband, proband’s mother, and proband’s brother were 44, 36, and 38, respectively. The MRI revealed brainstem cerebellar atrophy and “cross sign” and “ordinate sign” of pons. Pedigree 2 was certified SCA1; the CAG repeats of the proband, proband’s aunt, and proband’s asymptomatic cousin were 60, 51, and 52, respectively. The MRI revealed cerebellar atrophy in these individuals. We, therefore, concluded that it was difficult to diagnose the SCA subset solely through the clinical manifestation. The imaging characteristics analysis and final diagnosis depended basically on gene analysis data.  相似文献

A β-glucosidase effectively releasing diosgenin from spirostanosides of Dioscorea zingiberensis C. H. Wright (DZW), named AfG, was purified from a strain of Aspergillus fumigates. The molecular weight of AfG was 113 kDa. Analysis of protein fragments by ESI-Q-TOF indicated that AfG was a β-glucosidase. The circular dichroism spectrum suggested that the main secondary structure of AfG in Milli-Q water was α-helixes. Atomic force microscopy revealed that it was a globular protein. AfG maintained high activity from pH 3.6 to 5.0 and from 50 to 90°C. With the strong heat stability, AfG retained 55% of its original activity at 65°C for 120 h. AfG utilized muti-3-O-glycosides of various steroidal saponins from DZW as substrate, such as trillin, diosgenin diglucoside, dioscin, deltonin and gracillin, to yield diosgenin, suggesting the possibility of producing diosgenin from total saponins of DZW using a single enzyme.  相似文献
