Electron microscopic and biochemical studies revealed a salient difference in the response to toxic doses of ouabain by cultured cardiac muscle and non-muscle cells from neonatal rats. Progressive cellular injury in myocytes incubated with 1 · 10?4–1 · 10?3 M ouabain ultimately leads to swelling and necrosis. The morphological damage in myocytes was accompanied by a drastic decrease in 14CO2 formation from 14C-labeled stearate or acetate but not glucose. Neither morphological nor biochemical impairments were observed in non-muscle cells. The interaction between ouabain and the cultured cells, using therapeutic doses of ouabain (i.e., <1 · 10?7 M), was characterized. Two binding sites were described in both classes of cells, one site is a saturable K+-sensitive site whereas the other is non-saturable and K+-insensitive. The complexes formed between the sarcolemma receptor(s) and ouabain, at low concentrations of the drug (e.g., 7.52 · 10?9 M), had Kd values of 8.9 · 10?8 and 2.3 · 10?8 M for muscle and non-muscle cells, respectively. The formation and dissociation of the complexes were affected by temperature and potassium ions.  相似文献

This study was conducted to evaluate the influence of brassinosteroid (24-epibrassinolide, EBL) seed priming and optimal nitrogen (N) supply in improving salt tolerance in soybean. The experimental treatments were (a) control (nutrient solution without N and without EBL priming), (b) nutrient solution without N and EBL seed priming, (c) N supplemented nutrient solution without EBL priming and (d) EBL seed priming + N supplemented nutrient solution under optimal (0 mM NaCl) and salt stress (0 mM NaCl) conditions. Salt stress caused significant reduction in growth and biomass accumulation of soybean. However, EBL seed priming and application of N improved the soybean performance under optimal and salt stress conditions. In this regard, treatments receiving both EBL and N were more effective. EBL priming and N, alone and in combination, triggered the accumulation of osmolytes including proline, glycine betaine and sugars resulting in better photo-protection through maintenance of tissue water content. Antioxidant activity and osmolyte accumulation significantly increased due to combined treatment of N and EBL under normal as well as salt stress conditions. In conclusion, salt stress caused reduction in growth and biomass soybean due to oxidative damage and osmotic stresses. However, soybean performance was improved by seed priming with EBL. Supplementation of N further improved the effectiveness of EBL treatment in improving salt tolerance in soybean.  相似文献

Understanding the mating system and reproductive success of a species provides evidence for sexual selection. We examined the mating system and the reproductive success of captive adult black sea bream (Acanthopagrus schlegelii), using parentage assignment based on two microsatellites multiplex PCR systems, with 91.5% accuracy in a mixed family (29 sires, 25 dams, and 200 offspring). Based on the parentage result, we found that 93.1% of males and 100% of females participated in reproduction. A total of 79% of males and 92% of females mated with multiple partners (only 1 sire and 1 dam were monogamous), indicating that polygynandry best described the genetic mating system of black sea bream. For males, maximizing the reproductive success by multiple mating was accorded with the sexual selection theory while the material benefits hypothesis may contribute to explain the multiple mating for females. For both sexes, there was a significant correlation between mating success and reproductive success and the variance in reproductive success of males was higher than females. Variation in mating success is the greatest determinant to variation in reproductive success when the relationship is strongly positive. The opportunity for sexual selection of males was twice that of females, as well as the higher slope of the Bateman curve in males suggested that the intensity of intrasexual selection of males was higher than females. Thus, male–male competition would lead to the greater variation of mating success for males, which caused greater variation in reproductive success in males. The effective population number of breeders (Nb) was 33, and the Nb/N ratio was 0.61, slightly higher than the general ratio in polygynandrous fish populations which possibly because most individuals mated and had offspring with a low variance. The relatively high Nb contributes to the maintenance of genetic diversity in farmed black sea bream populations.  相似文献

Background

Piper sarmentosum, locally known as kaduk is belonging to the family of Piperaceae. It is our interest to evaluate their effect on human hepatoma cell line (HepG2) for the potential of anticarcinogenic activity.

Results

The anticarcinogenic activity of an ethanolic extract from Piper sarmentosum in HepG2 and non-malignant Chang's liver cell lines has been previously determined using (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide) (MTT) assays, where the IC50 value was used as a parameter for cytotoxicity. The ethanolic extract that showed anticarcinogenic properties in HepG2 cells had an IC50 of 12.5 μg mL-1, while IC50 values in the non-malignant Chang's liver cell line were greater than 30 μg mL-1. Apoptotic morphological changes in HepG2 cells were observed using an inverted microscope and showed chromatin condensation, cell shrinkage and apoptotic bodies following May-Grunwald-Giemsa's staining. The percentage of apoptotic cells in the overall population (apoptotic index) showed a continuously significant increase (p < 0.05) in 12.5 μg mL-1 ethanolic extract-treated cells at 24, 48 and 72 hours compared to controls (untreated cells). Following acridine orange and ethidium bromide staining, treatment with 10, 12 and 14 μg mL-1 of ethanolic extracts caused typical apoptotic morphological changes in HepG2 cells. Molecular analysis of DNA fragmentation was used to examine intrinsic apoptosis induced by the ethanolic extracts. These results showed a typical intrinsic apoptotic characterisation, which included fragmentation of nuclear DNA in ethanolic extract-treated HepG2 cells. However, the non-malignant Chang's liver cell line produced no DNA fragmentation. In addition, the DNA genome was similarly intact for both the untreated non-malignant Chang's liver and HepG2 cell lines.

Conclusion

Therefore, our results suggest that the ethanolic extract from P. sarmentosum induced anticarcinogenic activity through an intrinsic apoptosis pathway in HepG2 cells in vitro.  相似文献

In order to investigate the factors affecting the reproduction efficiency of Arab breeding mares in Tunisia, breeding data corresponding to 2340 mated cycles, over 5 years (from 2000 to 2004), from 555 mares offspring of 50 sires and bred with 17 stallions were analysed using a multivariate logistic regression. We chose logit link function and binomial distribution and we used log-likelihood-ratio (LL) and Wald tests (X2 Wald) to test the mean values. The factors of variation included in the model were the year, the stallion, the age of the mare at mating, the sire and the category of the mare. Logistic regression results showed that age and sire affected both the overall conception rate and the first-cycle conception rate of the mares whilst category and stallion influenced only the overall conception rate. To our knowledge, this is the first study showing that the sire of the mare can affect both the first-cycle conception rate and the overall conception rate of the mares.  相似文献

Retroviral insertional mutagenesis in mouse hematopoietic tumors provides a powerful cancer gene discovery tool. Here, we describe a high-throughput, single nucleotide polymorphism (SNP)-based method, for mapping retroviral integration sites cloned from mouse tumors, and a bacterial artificial chromosome (BAC) hybridization method, for localizing these retroviral integration sites to common sites of retroviral integration (CISs). Several new CISs were identified, including one CIS that mapped near Notch1, a gene that has been causally associated with human T-cell tumors. This mapping method is applicable to many different species, including ones where few genetic markers and little genomic sequence information are available. It can also be used to map endogenous proviruses.  相似文献

With the huge intensification of agriculture and the increasing awareness to human health and natural resources sustainability, there was a shift towards the development of environmental friendly N application approaches that support sustainable use of land and sustain food production. The effectiveness of such approaches depends on their ability to synchronize plant nitrogen demand with its supply and the ability to apply favored compositions and dosages of N-species. They are also influenced by farming scale and its sophistication, and include the following key concepts: (i) Improved application modes such as split or localized ("depot") application; (ii) use of bio-amendments like nitrification and urease inhibitors and combinations of (i) and (ii); (iii) use of controlled and slow release fertilizers; (iv) Fertigation-fertilization via irrigation systems including fully automated and controlled systems; and (v) precision fertilization in large scale farming systems. The paper describes the approaches and their action mechanisms and examines their agronomic and environmental significance. The relevance of the approaches for different farming scales, levels of agronomic intensification and agro-technical sophistication is examined as well.  相似文献

Plants possess highly sensitive perception systems by which microbial signal molecules are recognized. In the Bradyrhizobium-soybean (Glycine max (L.) Merr.) symbiosis, recognition is initiated through exchange of signal molecules, generally flavonoids from soybean and lipo-chitooligosaccharides (Nod factors) from the microsymbiont. Application of the Nod factor Nod Bj-V (C18:1, MeFuc) induced soybean resistance to powdery mildew caused by Microsphaera diffusa. Addition of Nod factor (concentrations ranging from 10(-6) to 10(-10) M) to soybean root systems led to reductions in disease incidence. The lowest disease incidence was caused by Nod factor treatment at 10(-6) M. The effect of Nod factor application on fungal growth and development was measured at 4, 12, 48, and 96 h after inoculation. Colony diameter and number of germ tubes per conidium were decreased by 10(-6) M Nod factor. Phenylalanine ammonia lyase (PAL, EC.4.3.1.1.) is the first enzyme of the phenyl propanoid pathway, and is commonly activated as part of plant responses to disease. Treatment of soybean seedlings with Nod factor, through stem wounds, induced PAL activity; the most rapid increase followed treatment with 10(-6) M Nod factor. These data show that soybean plants are able to detect root applied LCO and respond by increased disease resistance.  相似文献
