Phytochemical reinvestigation on the whole plants of Ypsilandra thibetica obtained five new spirostane glycosides, ypsilandrosides H-L (1-5), and a known saponin polyphylloside III (6). Among them, 1 and 2 are the first spirostane glycosides which possess novel 5(6 → 7) abeo-steroidal aglycones. Compounds 3 and 4 are rare saponins whose aglycones contain a hydroxyl group at C-7. Their structures were elucidated on the basis of MS, 1D and 2D NMR spectroscopic analysis and chemical evidences. The isolated compounds were evaluated for their cytotoxic activity on five tumor cell lines.  相似文献

Fluvastatin, a lipophilic statin, was known to inhibit proliferation and induce apoptosis in many cancer cells. Its potential anticancer was evaluated in three hepatocellular carcinoma (HCC) cell lines (HepG2, SMMC-7721 and MHCC-97H). Cells were treated with fluvastatin in vitro and its effect on cell proliferation, cell cycle, invasion and apoptosis was determined. Mechanism of apoptosis induced by fluvastatin on HCC cell lines was also investigated through western blotting and mitochondrial membrane potential (MMP) analysis. It was observed that fluvastatin inhibited proliferation of HCC cells by inducing apoptosis and G2/M phase arrest in a dose-dependent manner. The results of cell invasion assay revealed that fluvastatin significantly decreased the invasion potency of HCC cells. A mitochondria-operated mechanism for fluvastatin induced apoptosis might be involved and was supported by Western blotting and MMP analysis. After fluvastatin treatment, expression of Bcl-2 and procaspase-9 were downregulated, cytochrome c (cytosolic extract), Bax and cleaved-caspase-3 protein expression were increased. Furthermore, a breakdown of MMP in HCC cells was observed. To conclude, these results have provided a rationale for clinical investigations of fluvastatin in future as a potential anticancer reagent for growth control of HCC.  相似文献

Differentiation of human embryonic stem cells (hESCs) into hematopoietic lineages using various methods has been reported. However, the phenotype that precisely defines the hematopoietic progenitor compartment with clonogenic activities has yet to be determined. Here, we measured and characterized progenitor function of subfractions of cells prospectively isolated from human embryoid bodies (hEBs) during hematopoietic differentiation basing on surface markers CD45, CD34, CD43, and CD38. We report that hematopoietic progenitors predominantly resided in the CD45+ subset. CD43+ cells lacking CD45 expression were largely devoid of progenitor activity. However, progenitor activity and multipotentiality was more enriched in CD45+ cells co-expressing CD43. CD45+ subset co-expressing CD34 but lacking CD38 expression (CD45+CD34+CD38-) were further enriched for CFU capacity compared to the CD45+CD34+CD38+ subset. Our study demonstrates a role of CD43 in enriching hematopoietic progenitors derived from hEBs and reveals a hierarchical organization of hESC-derived hematopoietic progenitor compartments defined by phenotypic markers.  相似文献

Nocardia,Rhodococcus and Streptomyces,all members of the actinomycetes family,areGram-positive eubacteria with high G C content and able to form mycelium.We report here a newlyidentified plasmid pXT107 of Nocardia sp.107,one of the smallest circular plasmids found in Nocardia.The complete nucleotide sequence of pXT107 consisted of 4335 bp with 65% G C content,and encodedone replication extragenic palindromic (Rep) and six hypothetical proteins.The Rep,double-strand originand single-strand origin of pXT 107 resembled those of typical rolling-circle-replication plasmids,such aspNI100 of Nocardia,pRE8424 of Rhodococcus and pIJ101 of Streptomyces.The Escherichia coli-Nocardiashuttle plasmid pHAQ22,containing the rep gene of pXT107,is able to propagate in Nocardia but not inStreptomyces.  相似文献

Changes of a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor expression could impacts the viability of neurons and brain levels of brain-derived neurotrophic factor (BDNF) expression in the key brain structures in the pathophysiology of depressive disorder. In the present study, chronic unpredictable mild stress (CUMS) degraded performance decreased AMPA receptor expression and increased neuron apoptosis. Treatment with baicalin (20, 40 mg/kg) significantly reversed these changes. This study demonstrates that baicalin has potent antidepressant effect, likely through up-regulated the expression of AMPA receptor and suppression neuron apoptosis in CUMS-treated rats.  相似文献

To evaluate the effects of proteasome inhibitors lactacystin (LAC) and beta-lactacystin (beta-LAC) on the proliferation and activation of T lymphocytes, flow cytometry was used to analyze the proliferation and the expression of CD69, CD25 and CD3 of T lymphocytes activated by PHA. Furthermore, the expressions of PA28 and IL-2 mRNA were assayed by competitive RT-PCR. The results indicated that: (1) LAC and beta-LAC significantly decreased the incorporation of BrdU and inhibited T lymphocytes proliferation in T lymphocytes activated by PHA; (2) although LAC and beta-LAC did not affect the expression of CD69 at any time, they significantly inhibited the expression of CD25 (48 h, 72 h, P<0.05); (3) in comparison with control, LAC and beta--LAC significantly down-regulated the expression of PA28 and IL-2 mRNA (48 h, 72 h, P<0.05). LAC and beta-LAC significantly inhibited the proliferation and activation of T cells. Mechanisms involved are inhibition of CD25 and down-regulation of PA28 and IL-2 mRNA expressions.  相似文献

用2m×2m的样方调查了湖南茶陵普通野生稻保护区群落,绘制了群落优势种水平分布图。选用斑块数量、总斑块面积、平均斑块面积、标准差、变异系数、中值、最大斑块面积、最小斑块面积和优势度等几个描述统计量,来刻画斑块大小的分布特征。选取9个不同生态条件的样地计测了其中10个主要物种的生态位宽度;以群落间接生境梯度代替生态因子梯度,计算了其生态位重叠值,进行了生态位重叠排序。结果表明:各物种斑块在群落中呈异质分布,物种在群落中发生现实生态位分离,这种分离主要是由生境条件特别是生境水位决定的。根据生态位重叠排序可把物种分为3组:莼菜(Brasenia schreberi)、水竹叶(Murdannia triqeutra)、莲(Nelumbo nucifera)、锐棱荸荠(Eleocharis acutangula)、水毛花(Scirpus triangulatus)、慈姑(Sagittaria trifolia)等分布于20cm以上的深水位生境;普通野生稻(Oryza rufipogon)、李氏禾(Leersia hexandra)等分布于10~20 cm中水位生境;柳叶箬(Isachne globosa)分布于10cm以下浅水位生境。排序的结果与物种在群落中的空间分布情况相符合。在普通野生稻的保护实践中,适宜生境尤其是适宜水位的维持十分重要。  相似文献

Regiocontrollable selectivity of enzymatic method for synthesis of polymerizable derivatives of methyl shikimate was described. Lipase acrylic resin from Candida antarctica (CAL-B) and immobilized lipase from Mucor miehei (MML) showed high regioselectivity toward the secondary hydroxyl of methyl shikimate, which presents three hydroxyl groups with similar reactivity. Catalysis by MML in acetone facilitated the single step synthesis of 5-O-acyl methyl shikimate derivatives in high yields, while the use of CAL-B in acetone afforded 4-O-acyl methyl shikimate derivatives. The obtained series of methyl shikimate derivatives would be important monomers for potential useful analogues of shikimic acid.  相似文献

We report on the results of laboratory evaluation of a dual-channel ultraviolet aerodynamic particle sizer spectrometer for real-time, continues monitoring of bio-aerosols. The detected fluorescence light was divided into two channels, simultaneously measuring the blue (400–500 nm) and green (500–600 nm) signal, which were corresponding to the fluorescence spectra of NAD(P)H and riboflavin, respectively. Primary results show the different fluorescence properties of the bacteria aerosols (Escherichia coli, Staphylococci and Bacillus subtilis var. niger spore) and the substrate (TSA eluate solutions, TSB and Nutrient broth). We demonstrated a promising method to improve the discrimination capability of bioaerosols from non-bioaerosol background components and to distinguish different types of bioaerosol particles.  相似文献

果蝇的视觉信号转导途径是已知的最快的G 蛋白偶联信号通路。这其中涉及到TRP/TRPL通道的开放以及钙离子的内流等一系列反应的形成。NINAC(neither inactivation nor afterpotential C)是一种特异性存在于果蝇感光细胞中的第3类肌球蛋白(Myosin III),其在终止果蝇的视觉信号转导通路中起着非常重要的作用。NINAC蛋白具有两种亚型:一种是132 kD的蛋白亚型 (p132),另一种则是174 kD的蛋白亚型(p174)。这两种不同的蛋白亚型都具有相同的激酶催化结构域(kinase domain),以及与肌球蛋白相似的马达结构域(motor domain)。但是,它们在C末端却存在着非常大的差异,这其中包括了钙调蛋白结合基序(IQ motif)。NINAC的这两种蛋白亚型在果蝇的感光细胞中的定位以及作用有很大不同,尤其是在与钙调蛋白的相互作用方面。钙调蛋白结合基序与钙调蛋白(CaM)之间的相互作用对于果蝇的视觉信号通路具有重要的意义:NINAC结合钙调蛋白能力的缺失将导致果蝇的视觉传导缺陷。本文通过蛋白共表达的方法,成功表达并纯化得到了不同版本的NINAC与钙调蛋白的蛋白复合物。静态光散射的结果表明,在Ca2+存在情况下,p174蛋白可以结合2个Ca2+-CaM,而p132只结合1个Ca2+-CaM。通过分析型凝胶过滤以及等温量热滴定技术,进一步鉴定了p174及p132的IQ2(第2个钙调蛋白结合基序)序列与Ca2+ CaM的相互作用。通过序列分析及进一步的突变实验发现,p174 IQ2中的3个疏水氨基酸(F1083,F1086 和 L1092)对于钙调蛋白的结合非常重要,并导致了p174与p132蛋白和Ca2+ CaM结合能力的差异。本文的研究提供了NINAC与Ca2+-CaM相互作用的生化机制,将为进一步在果蝇视觉信号通路中深入研究CaM是如何调节NINAC的体内功能实验打下基础。  相似文献
