The distribution of Vulpes cana in the extreme eastern Sahara is described on the basis of new photographic and sight records, as well as a recently acquired specimen from south-eastern Egypt. The proven range of this elusive fox now extends about 1000 km south into Africa along the Red Sea coastal mountains. The proven and predicted distribution ranges of this fox are discussed. Its phylogeography and evolutionary relationship to other foxes of the region are gleaned from analysis of mitochondrial cytochrome b DNA sequence and are discussed in light of climatic and paleogeographic history of the area. The results show that the species could have initially moved into Africa during the Pliocene-Pleistocene transition period when several land bridges occurred across the Gulf of Suez. More recent land bridges between the mountains of the Sinai Peninsula and the Eastern Desert of Egypt seem to have developed and disappeared several times during the Pleistocene, possibly allowing genetic exchange between V. cana populations in these two areas. The last of this population exchange and genetic flow between the two areas seems to have ended some 14,000 years ago when the present Gulf of Suez inundation took place.  相似文献

A challenging aspect of biomarker discovery in serum is the interference of abundant proteins with identification of disease-related proteins and peptides. This study describes enrichment of serum by denaturing ultrafiltration, which enables an efficient profiling and identification of peptides up to 5 kDa. We consistently detect several hundred peptide-peaks in MALDI-TOF and SELDI-TOF spectra of enriched serum. The sample preparation is fast and reproducible with an average CV for all 276 peaks in the MALDI-TOF spectrum of 11%. Compared to unenriched serum, the number of peaks in enriched spectra is 4 times higher at an S/N ratio of 5 and 20 times higher at an S/N ratio of 10. To demonstrate utility of the methods, we compared 20 enriched sera of patients with hepatocellular carcinoma (HCC) and 20 age-matched controls using MALDI-TOF. The comparison of 332 peaks at p < 0.001 identified 45 differentially abundant peaks that classified HCC with 90% accuracy in this small pilot study. Direct TOF/TOF sequencing of the most abundant peptide matches with high probability des-Ala-fibrinopeptide A. This study shows that enrichment of the low molecular weight fraction of serum facilitates an efficient discovery of peptides that could serve as biomarkers for detection of HCC as well as other diseases.  相似文献

A new technique for generating xylem cavitation and vulnerability curves was evaluated. The centrifugal force was used to lower the negative pressure in a xylem segment and to induce a positive pressure difference between sample's ends. This enabled the determination of sample hydraulic conductance during centrifugation and, hence, its variation with decreasing xylem pressures. The centrifuge technique was compared with standard methods on a large number of species including conifers, diffuse-porous and ring-porous woody angiosperms. A very good agreement was found for coniferous and diffuse-porous species. However, the technique was not appropriate for ring-porous species, probably because many vessels were cut open in the centrifuged xylem segments. The main advantage of this technique is its rapidity, the vulnerability curve of a xylem segment being constructed typically in less than half an hour. This will greatly facilitate the study of xylem cavitation in ecological or genetic researches.  相似文献

Backgrounds

With 10% of the general population aged 15–59 years chronically infected with hepatitis C virus (HCV), Egypt is the country with the highest HCV prevalence worldwide. Healthcare workers (HCWs) are therefore at particularly high risk of HCV infection. Our aim was to study HCV infection risk after occupational blood exposure among HCWs in Cairo.

Methodology/Principal Findings

The study was conducted in 2008–2010 at Ain Shams University Hospital, Cairo. HCWs reporting an occupational blood exposure at screening, having neither anti-HCV antibodies (anti-HCV) nor HCV RNA, and exposed to a HCV RNA positive patient, were enrolled in a 6-month prospective cohort with follow-up visits at weeks 2, 4, 8, 12 and 24. During follow-up, anti-HCV, HCV RNA and ALT were tested. Among 597 HCWs who reported a blood exposure, anti-HCV prevalence at screening was 7.2%, not different from that of the general population of Cairo after age-standardization (11.6% and 10.4% respectively, p = 0.62). The proportion of HCV viremia among index patients was 37%. Of 73 HCWs exposed to HCV RNA from index patients, nine (12.3%; 95%CI, 5.8–22.1%) presented transient viremia, the majority of which occurred within the first two weeks after exposure. None of the workers presented seroconversion or elevation of ALT.

Conclusions/Significance

HCWs of a general University hospital in Cairo were exposed to a highly viremic patient population. They experienced frequent occupational blood exposures, particularly in early stages of training. These exposures resulted in transient viremic episodes without established infection. These findings call for further investigation of potential immune protection against HCV persistence in this high risk group.  相似文献

Recently, we reported the role of Arabidopsis cyclic nucleotide-gated ion channel (AtCNGC) 11 and 12 in Ca2+-dependent physiological responses. AtCNGC11 and 12 have been reported to be involved in plant immunity, but whether these channels play additional physiological roles was not clear before. Using single and double knockout mutants, we have found that these channels play significant roles in Ca2+ signaling, which mediates several physiological processes, such as gravitropic bending and senescence. Here, we conducted a high throughput, non-biased chemical screen using the gain-of-function mutant of AtCNGC11 and 12, cpr22. Our data presented here indicates that Ca2+ but not K+ channel blockers suppress AtCNGC11/12-induced lethality. Our data further suggest that AtCNGC11 and 12 are involved in Ca2+-dependent, but not K+-dependent physiological responses in planta.  相似文献

Five flavonoids (rutin, asebotin, 3-hydroxyasebotin, quercetin-3-O-beta-D-xylopyranoside, and a racemic mixture of catechin) and caffeic acid were isolated and identified for the first time from seagrass, Thalassodendron ciliatum, collected from the Hurghada region in Egypt. The crude extract and the isolated pure compounds were evaluated for their cytotoxic activities against HCT-116, HEPG, MCF-7, and HeLa human cancer cell lines, for their antiviral activity against Herpes Simplex and hepatitis A viruses, and for their antioxidant activity.  相似文献

Introduction

Higher levels of high density lipoprotein (HDL) subfractions HDL3-chol and particularly HDL2-chol protect against cardiovascular disease (CVD), but inflammation reduces the HDL level and may impair its anti-atherogenic effect. Changed HDL composition through the impact of inflammation on HDL subfractions may contribute to the excess risk of CVD in rheumatoid arthritis (RA). In this study, we investigated whether HDL2-chol and HDL3-chol concentrations differ between RA patients and healthy controls, and whether these levels are related to the level of RA disease activity.

Methods

Non-fasting blood samples were collected from 45 RA patients and 45 healthy controls. None of the participants had a history of CVD, diabetes, or used lipid-lowering drugs. HDL2-chol and HDL3-chol concentrations were obtained by ultracentrifugation. Regression modeling was used to compare HDL subfraction levels between RA patients and healthy controls, and to analyze the effect of disease activity on HDL2-chol and HDL3-chol.

Results

HDL2-chol and HDL3-chol were significantly lower in RA patients compared to healthy controls (P = 0.01, P = 0.005, respectively). The HDL2:HDL3 ratio was significantly lower in patients compared to controls (P = 0.04). Reduced HDL2-chol and HDL3-chol levels were primarily present in female RA patients and not in male RA patients. A modest effect of the disease activity score in 28 joins ( DAS28) on HDL2-chol concentrations was found, after correction for disease duration, glucocorticosteroid use and body mass index (BMI), with a 0.06 mmol/L decrease with every point increase in DAS28 (P = 0.05). DAS28 did not significantly affect HDL3-chol concentrations (P = 0.186).

Conclusions

Both HDL subfractions but particularly HDL2-chol concentrations were decreased in RA, primarily in women. This seems to be associated with disease activity and is of clinical relevance. The reduction of the HDL subfraction concentrations, particularly the supposedly beneficial HDL2-chol, may negatively impact the cardiovascular risk profile of women with RA.  相似文献

In this paper, we introduce a plasmonic structure-based fiber optics gas sensor using gold nano-particles on the end face. The proposed optical sensors are designed and evaluated to investigate the influence of the shape of the nano-particles. Evaluation techniques such as wavelength and peak sensitivities, full width at half maximum, and figure of merit are studied. By extension, we numerically study the macroscopic electric and magnetic field distributions and the microscopic polarization and current density in order to interpret the physical mechanism. The optical sensor based on nano-particles of classical spherical shape shows the highest wavelength sensitivity. However, it was motivating that the sensor based on nano-particles of the conical shape offers a better performance. It exhibits resonance wavelength and peak intensity sensitivities of 157 nm/RIU and 44 %/RIU, respectively, which give a higher overall sensitivity. In addition, it offers a narrow stable line width and high figures of merit.  相似文献
