In vitro synthesis of lectins in cell-free extracts from dry wheat and rye embryos

Cell-free extracts from dry wheat (Triticum vulgare L.) and rye (Secale cereale L.) embryos do not synthesize their corresponding lectins when incubated under conditions optimalized for translation of either exogenous or endogenous mRNA. Only when the extracts are prepared and incubated in the complete absence of sulphydryl containing compounds lectins are synthesized in vitro. Since immunoprecipitation techniques could not demonstrate unequivocally the presence of lectin among the cell-free translation products a specific affinity purification procedure has been developed in order to proof the presence of stored lectin mRNAs in dry wheat and rye embryos.Abbreviations DTT dithiothreitol - SDS-PAGE sodium dodecyl sulphate polyacrylamide gel electrophoresis - TCA trichloracetic acid     

Annuithrin,a new biologically active germacranolide from Helianthus annuus

Investigations on growth inhibition in Helianthus annuus led to the isolation of a new sesquiterpene lactone, a germacranolide with an α-methylene-γ-lactone moiety. The structure of this new germacranolide, annuithrin, was elucidated by spectroscopic methods. Its biological activity has been proven by growth inhibition in straight growth tests, antibacterial tests and inhibition of DNA-/RNA-synthesis in cells of the ascitic form of Ehrlich carcinoma.     

Reproductive strategies in the freshwater sphaeriid clam,Musculium partumeium (say), from a permanent and a temporary pond

Summary Reproduction was studied in two populations ofMusculium partumeium from temporary and permanent ponds. Adults of the single annual generation from the ephemeral pond have an annual selection ratio of 25:1 with 37.03 gC per newborn, and an intrinsic rate of increase (r) of 0.0084 day^-1. Fall-born adults from the permanent pond have an annual selection ratio of 38:1, 21.82 gC per newborn and anr of 0.0115 day^-1; springborn adults have an annual selection ratio of 136:1 (107:1 for their contribution to fall birth and 29:1 for the spring birth period) with newborns of 24.21 gC and anr of 0.0304. The trade off between quantity and quality of young is discussed in terms of adaptive strategies.     

H-Y antigen in 46,XY gonadal dysgenesis

Summary Presence of H-Y antigen has been correlated with testicular differentiation, and absence of H-Y with failure of testicular differentiation, in a variety of mammalian species. To determine more precisely the relationship between expression of H-Y antigen and development of the testis, we studied the cells of phenotypic females with the 46,XY male karyotype. Blood leukocytes were typed H-Y⁺ in five XY females with gonadal dysgenesis, although in other studies blood leukocytes from XY females with gonadal dysgenesis were typed H-Y^-. Thus mere presence of H-Y antigen is not sufficient to guarantee normal differentiation of the testis. In the present paper we review evidence for an additional factor in gonadal organogenesis, the H-Y antigen receptor. We infer that testicular development requires engagement of H-Y and its receptor. It follows that XY gonadal dysgenesis is the consequence of functional absence of the H-Y testis inducer as in the following conditions: failure of synthesis of H-Y or failure of specific binding of H-Y.     

Differential precipitation with concanavalin a as a method for the purification of glycoproteins: Human alkaline phosphatase

The purification of a glycoprotein from a mixture by first precipitating with a lectin and fractional solution of the precipitate could be applicable as an important preparative procedure. Another possibility could be fractional precipitation with the lectin by varying the concentration of Ca²⁺ and Mn²⁺ required for binding. These principles have been tested in relation to the preparation of human liver alkaline phosphatase. Both approaches have led to the purification of alkaline phosphatase from single human livers with a high recovery (28 and 34%) and purification factors of 16 × 10³ and 13 × 10³ over the initial aqueous butanl-ol extract. The specific activities for the preparations were 1610 and 1500 units/mg of protein, respectively; a unit is defined as the amount of enzyme catalyzing the hydrolysis of 1.0 μmol of p-nitrophenyl phosphate/min at 35°C in 0.1 m 2-amino-2-methylpropan-1-ol/HCl buffer, pH 10.5, containing 10 mmp-nitrophenyl phosphate, 0.1 mm MgCl₂, and 1 μm ZnSO₄.     

Influence of abiotic factors and physiological conditions on the formation of aggregations in cotton-bugs (Dysdercus spp., Heteroptera)

The subsocial habits of Dysdercus spp. are manifest first of all by the formation of aggregations, which are based on an inter-individual attraction. The tendency to establish aggregations is modified by several environmental factors such as temperature, intensity of light, relative humidity of the air and moisture of the substrate. In addition, the fluctuating physiological condition of the insects during their development as well as the daily periodicity of activity have a considerable influence on the aggregation-tendency.

Zusammenfassung Die subsoziale Lebensweise von Dysdercus-Arten kommt in der Bildung von Aggregationen zum Ausdruck, deren Entstehung ursächlich auf einer interindividuellen sozialen Attraktion beruht. Die Aggregationsbildungstendenz wird von einer Reihe abiotischer Faktoren stark beeinflußt: Ansteigende Temperatur, zunehmende Beleuchtungsstärke und abnehmende rel. Luftfeuchte vermindern sie. Hohe Substratfeuchtewerte wirken sich auf die Aggregationsbildungstendenz der Larven I fördernd, auf die der älteren Stadien hemmend aus. Weiterhin spielt der wechselnde physiologische Zustand der Tiere im Verlaufe der Ontogenese und die diurnale Aktivitätsrhythmik eine Rolle. Der Aggregationsgrad vermindert sich mit fortschreitender Larvenentwicklung bis hin zum Imaginalstadium, wobei zwischen den einzelnen Häutungen ausgeprägte Minima, während der Häutungen Maxima auftreten. Im Tagesverlauf liegt das Minimum der Aggregationsbildungstendenz in der Mitte der Lichtphase.

     

Staphylococcal nuclease reviewed: A prototypic study in contemporary enzymology

Summary This is the third in a series of four articles in which the chemical, enzymological and crystallographic work on Ribonucleate (deoxribonucleate)-3-nucleotidohydrolase, EC 3.1.4.4 (staphylococcal nuclease, micrococcal nuclease) will be reviewed and correlated. This article describes the structure of the nuclease and of a nuclease-inhibitor complex as determined by x-ray crystallography. The crystal structures are correlated with some of the known chemical and enzymological properties of the enzyme, and the three areas combined to propose a mechanism of action.This article is the third in a series of four devoted to the stapholoccal nuclease. Reviews concerning its isolation and enzymology (1) as well as the features of its ligand binding site (2) have appeared in previous issues. Collagen biochemistry of osteopetrotic bone: I. Quantitative changes in bone collagen cross-links in virus-induced avian osteopetrosis

The patterns of [³H]-NaBH₄-reduced bone collagen cross-links from osteopetrotic chickens were compared with those of age-matched controls. Ratios of the reduced cross-links, dihydroxylysinonorleucine (DHLNL)¹ to hydroxylysinonorleucine (HLNL), were dramatically increased in tibia bone samples from osteopetrotic birds compared to values from control birds. In addition, the initial HLNL peak from osteopetrotic bone collagen was heterogeneous, whereas DHLNL from osteopetrotic or normal bone collagen and HLNL from normal bone collagen were homogeneous.
